A Generic Mechanism of Emergence of Amyloid Protofilaments from Disordered Oligomeric Aggregates

Abstract: The presence of oligomeric aggregates, which is often observed during the process of amyloid formation, has recently attracted much attention because it has been associated with a range of neurodegenerative conditions including Alzheimer's and Parkinson's diseases. We provide a description of a sequence-indepedent mechanism by which polypeptide chains aggregate by forming metastable oligomeric intermediate states prior to converting into fibrillar structures. Our results illustrate that the formation of ordered arrays of hydrogen bonds drives the formation of β-sheets within the disordered oligomeric aggregates that form early under the effect of hydrophobic forces. Individual β-sheets initially form with random orientations and subsequently tend to align into protofilaments as their lengths increase. Our results suggest that amyloid aggregation represents an example of the Ostwald step rule of first-order phase transitions by showing that ordered cross-β structures emerge preferentially from disordered compact dynamical intermediate assemblies.
The historical pathway towards more accurate homogenisation

Abstract: In recent years increasing effort has been devoted to objectively evaluate the efficiency of homogenisation methods for climate data; an important effort was the blind benchmarking performed in the COST Action HOME (ES0601). The statistical characteristics of the examined series have significant impact on the measured efficiencies, thus it is difficult to obtain an unambiguous picture of the efficiencies, relying only on numerical tests. In this study the historical methodological development with focus on the homogenisation of surface temperature observations is presented in order to view the progress from the side of the development of statistical tools. The main stages of this methodological progress, such as for instance the fitting optimal step-functions when the number of change-points is known (1972), cutting algorithm (1995), Caussinus – Lyazrhi criterion (1997), are recalled and their effects on the quality-improvement of homogenisation is briefly discussed. This analysis of the theoretical properties together with the recently published numerical results jointly indicate that, MASH, PRODIGE, ACMANT and USHCN are the best statistical tools for homogenising climatic time series, since they provide the reconstruction and preservation of true climatic variability in observational time series with the highest reliability. On the other hand, skilled homogenizers may achieve outstanding reliability also with the combination of simple statistical methods such as the Craddock-test and visual expert decisions. A few efficiency results of the COST HOME experiments are presented to demonstrate the performance of the best homogenisation methods.
Telemedicine in veterinary practice

Abstract: Veterinary surgeons have a long tradition of consulting one another about problem cases and many have unwittingly practised telemedicine when discussing cases by telephone or by sending laboratory reports by telefax. Specific veterinary telemedicine applications have been in use since the early 1980s, but little research has been undertaken in this field. The Pubmed and CAB International databases were searched for the following Boolean logic-linked keywords; veterinary AND telemedicine, veterinary AND telecare, animal AND telemedicine, animal AND telecare and veterinary AND e-mail and an additional search was made of the worldwide web, using Google Scholar. This returned 25 papers which were reviewed. Of these only 2 report research. Sixteen papers had no references and 1 author was associated with 13 papers. Several themes emerge in the papers reviewed. These include remarks about the use of telemedicine, the benefits that can and are derived from the use of telemedicine, areas of practice in which telemedicine is being used, ethical and legal issues around the practice of telemedicine, image standards required for telemedicine, the equipment that is required for the practice of telemedicine, advice on ways in which digital images can be obtained and educational aspects of telemedicine. These are discussed. Veterinary practice has lagged behind its human counterpart in producing research on the validity and efficacy of telemedicine. This is an important field which requires further research.
Kardiovaskul re Effekte von Schilddrüsenhormonen und Schilddrüsenfunktionsst rungen

Abstract: Schilddrüsenhormone weisen zahlreiche Effekte auf das kardiovaskul re System auf. Eine Hypothyreose ist mit Hypercholesterin mie und Hypertonie assoziiert und kann zu manifesten kardiovaskul ren Erkrankungen führen. Auch eine subklinische Hypothyreose mit normalen peripheren Schilddrüsenhormonkonzentrationen wird mit einer Beschleunigung der Atherogenese bei lteren Frauen in Verbindung gebracht. Patienten mit Schilddrüsenüber- oder -unterfunktion zeigen charakteristische h modynamische Ver nderungen. Eine Schilddrüsenüberfunktion führt zu einem "high-output state" mit Zunahme des Herzminutenvolumens in Ruhe und einer Abnahme des peripheren Widerstandes. Genau gegens tzliche h modynamische Ver nderungen sind in Zusammenhang mit einer Hypothyreose zu erwarten. Sowohl die manifeste als auch eine latente Hyperthyreose ist mit einer Zunahme atrialer Rhythmusst rungen, vor allem von Vorhofflimmern, assoziiert. Dieser Artikel gibt eine übersicht über den Zusammenhang zwischen Schilddrüsenfunktionsst rungen und kardiovaskul ren Erkrankungen.
Self-Templated Nucleation in Peptide and Protein aggregation

Abstract: Peptides and proteins exhibit a common tendency to assemble into highly ordered fibrillar aggregates, whose formation proceeds in a nucleation-dependent manner that is often preceded by the formation of disordered oligomeric assemblies. This process has received much attention because disordered oligomeric aggregates have been associated with neurodegenerative disorders such as Alzheimer's and Parkinson's diseases. Here we describe a self-templated nucleation mechanism that determines the transition between the initial condensation of polypeptide chains into disordered assemblies and their reordering into fibrillar structures. The results that we present show that at the molecular level this transition is due to the ability of polypeptide chains to reorder within oligomers into fibrillar assemblies whose surfaces act as templates that stabilise the disordered assemblies.

Abstract: Au cours des premières décennies du dix-neuvième siècle, la paysannerie des Hautes Terres d’Ecosse connut une profonde et douloureuse mutation sous l’impulsion de divers facteurs culturels et économiques. La presse des Basses Terres se fit l’écho de ces transformations et se montra le plus souvent critique envers les habitants d’une région per ue comme étrangère. L’analyse du discours du Blackwood’s Edinburgh Magazine, l’une des publications littéraires majeures en Grande-Bretagne, et de deux des journaux qui firent preuve de la plus grande virulence à l’encontre des Highlanders, le Perthshire Advertiser et le Fifeshire Journal, permet d’identifier la nature des stratégies de distanciation auxquelles la presse des Basses Terres eut recours. Dans cet article je m’efforcerai de démontrer que les analyses proposées par Edward Said dans Orientalism et Culture and Imperialism peuvent s’appliquer à la description des mécanismes mis en uvre par la presse des Lowlands.Said, en effet, assimile le discours colonial à un système statique d’essentialisme synchronique . Or le discours de la presse des Lowlands avait comme vocation première de présenter la communauté des Highlands comme essentiellement statique, de la rejeter dans l’altérité et d’en discréditer les schémas culturels. La perception fondamentalement négative de la celtitude s’articulait autour d’un ensemble de couples antithétiques (travail-indolence, supériorité-infériorité, modernité-archa sme ou encore civilisationbarbarie). Une démarche de type ethnocidaire s’imposait donc afin que le Highlander puisse quitter son statut d’étranger pour bénéficier, au même titre que les autres membres de la communauté britannique, des bienfaits de la civilisation et du progrès.

Abstract: Die Herzinsuffizienz stellt eine der h ufigsten internistischen Erkrankungen dar. Grundpfeiler der pharmakologischen Therapie der chronischen Herzinsuffizienz mit dem Ziel einer Prognoseverbesserung ist die Blockade der im Rahmen des Syndroms Herzinsuffizienz stattfindenden neurohumoralen Aktivierung. Die Ger tetherapie (CRT, ICD) gewinnt im Therapiekonzept der Behandlung von Patienten mit chronischer Herzinsuffizienz zunehmende Bedeutung. Effektive Therapien etwa für spezifische Behandlung der diastolischen Herzinsuffizienz sind limitiert. Interessante neue Therapieans tze zur Behandlung der chronischen Herzinsuffizienz werden gegenw rtig in klinischen Studien untersucht. Gro e klinische Untersuchungen mit neuen Substanzen brachten zum Teil gemischte Ergebnisse. Das unterstreicht die Tatsache, dass es schwierig ist, zus tzlich zur momentan verfügbaren Therapie einen additiven Benefit zu erreichen. Die folgende Arbeit gibt eine übersicht, was sich auf dem Gebiet der Herzinsuffizienztherapie in den vergangenen Jahren entwickelt hat.
