Functional Characterization of the Small Heat Shock Protein Hsp12p from Candida albicans

Abstract: Hsp12p is considered to be a small heat shock protein and conserved among fungal species. To investigate the expression of this heat shock protein in the fungal pathogen Candida albicans we developed an anti-CaHsp12p antibody. We show that this protein is induced during stationary phase growth and under stress conditions including heat shock, osmotic, oxidative and heavy metal stress. Furthermore, we find that CaHsp12p expression is influenced by the quorum sensing molecule farnesol, the change of CO2 concentration and pH. Notably we show that the key transcription factor Efg1p acts as a positive regulator of CaHsp12p in response to heat shock and oxidative stress and demonstrate that CaHsp12p expression is additionally modulated by Hog1p and the cAMP-PKA signaling pathway. To study the function of Hsp12p in C. albicans we generated a null mutant, in which all four CaHSP12 genes have been deleted. Phenotypic analysis of the strain shows that CaHSP12 is not essential for stress resistance, morphogenesis or virulence when tested in a Drosophila model of infection. However, when overexpressed, CaHSP12 significantly enhanced cell-cell adhesion, germ tube formation and susceptibility to azole antifungal agents whilst desensitizing C. albicans to the quorum sensing molecule farnesol.

Abstract: this article refers to the quality of the textual information, more specifically in extensive documents, addressing the cohesion between text sections. in this sense, an approach was proposed and tested which integrates two technical resources: social network analysis and internal references. the methodology employed was participatory research, including cohesion analysis between 109 sections of an extensive document text developed by the actual researchers. the results have proved satisfactory in helping the researchers to identify points for improvement of cohesion between the various text units in the extensive document analyzed.
CO2 Acts as a Signalling Molecule in Populations of the Fungal Pathogen Candida albicans

Abstract: When colonising host-niches or non-animated medical devices, individual cells of the fungal pathogen Candida albicans expand into significant biomasses. Here we show that within such biomasses, fungal metabolically generated CO2 acts as a communication molecule promoting the switch from yeast to filamentous growth essential for C. albicans pathology. We find that CO2-mediated intra-colony signalling involves the adenylyl cyclase protein (Cyr1p), a multi-sensor recently found to coordinate fungal responses to serum and bacterial peptidoglycan. We further identify Lys 1373 as essential for CO2/bicarbonate regulation of Cyr1p. Disruption of the CO2/bicarbonate receptor-site interferes selectively with C. albicans filamentation within fungal biomasses. Comparisons between the Drosophila melanogaster infection model and the mouse model of disseminated candidiasis, suggest that metabolic CO2 sensing may be important for initial colonisation and epithelial invasion. Our results reveal the existence of a gaseous Candida signalling pathway and its molecular mechanism and provide insights into an evolutionary conserved CO2-signalling system.

Abstract: this paper discusses the importance of alternative models of regulation of the quality of public schools in response to the purposeful evaluation models focused on student achievement on standardized tests. given the paucity of studies on public policy implementation becomes the experience of a municipal school system as a reference for the analysis of the strengths and vulnerabilities of the evaluation process implemented taking as participatory institutional focus on the negotiation meetings between the managers of the central and the actors of the schools. the aim is to identify concepts of quality and assessment governing the negotiation process and the consequences for decision making. it is observed that the actors tend to enhance the understanding of his role in negotiating and there is evidence of expanding the role and involvement of players from schools with the logic evaluation of the model with the continuity of the process.

Abstract: the study aims to think about the meaning of the self evaluation commissions created as a strategy that permits institutional self- evaluation processes in higher education. it aims to establish comparisons from a rereading of the model used by sinaes in brazil higher education. the results confirm the importance of the self evaluation commissions as an instance that promotes the meeting of the social actors in collective spaces to search for the improvement of learning quality in an organized and plural form. the advantages of the use of such commissions in fundamental school education are appointed by the proximity of the self evaluation processes of the internal and external actors related to the school.

Abstract: the text addresses evaluation as an essential knowledge for teachers' work processes. facing the external evaluation processes increasingly more present in the context of educational policies, the authors propose that the processes of teacher education incorporate a discussion about institutional evaluation presenting it as a mediation between the evaluation of learning and the evaluation of systems. the articulation between the three levels of evaluation reverberates in the processes of qualification of the forms of participation of the teacher in the pedagogical project and indirectly improves the students' learning.

Abstract: accuracy is an essential characteristic of quality information. and the summary is the main resource for organizational information. the summarty provides the researchers with a quick understanding of information, improves the levels of access and utilization of collections of corporative information. the research evaluated the capability of software for automatic generation of summaries, for selection of units of text which express the central ideas in extensive textual information. therefrom, summaries have been generated for a widely known article. together with the author's original summary, they were refereed by twenty expert ressearchers. human authors present higher quality summaries, but the quality level of summaries created by the new generations of software summarizers enables them to be considered as important tools for the centers of organizational information that have to add aggregate values to their information collections.

Abstract: the article deals with strategic potential of information systems oriented to organizational networks. the objective is to show the importance of the domain and metadata discrimination and communication protocols by network actors. the reflection is done from the analysis of two examples of network information systems. it is concluded that the semantic discussion between the represented organizational networks - in the context of information management- is as important for the success of the net as is the planning and cooperative action by other dimensions of the management.
