A Study on Spatio-temporal Data Model based on Feature and Event

Abstract: The feature is an essential element that comprises spatio-temporal data information and the event is a fundamental factor that result in changing spatio-temporal data.Basing on analyzing several existing Spatio-temporal Data Model,this paper introduces a new Spatio-temporal Data Model based on feature and event double mechanisms.The character of spatio-temporal data changing is analyzed.And how to organize data,build relations between features and storage data in spatio-temporal database are also emphatically discussed.At last,a software package of dynamic land use information system based on above model and methods are introduced.Its running network environment is Windows NT Server 2000 and client is Windows workstation 4.0/2k/xp professional operation systems.Another important technology used in dynamic land use information system is a self-developed Attribute Database Engine(Dblink) combined with Spatial Database Engine(SDE).It is proven efficient in realizing the query and retrospect of the history information during system's operating.
High-Level Overproduction of Thermobifida Enzyme in Streptomyces lividans Using a Novel Expression Vector

Abstract: In this study, we constructed a novel Streptomyces- E.coli shuttle vector pZRJ362 combining the xylose isomerase promoter and amylase terminator. A gene encoding the endoglucanase Cel6A in Thermobifida fusca was amplified by PCR, cloned into Streptomyces lividans host strain using the novel expression vector and Pichia pastoris GS115 host strain using the vector pPICZα-C, respectively. Afterwards, the expression pattern and the maximum expression level were comparatively studied in both expression systems. The maximum enzyme activity of Cel6A-(His) 6 secreted in S. lividans supernatant after 84-h of cultivation amounted to 5.56 U/mL, which was dramatically higher than that secreted in P. pastoris about 1.4 U/mL after 96-h of cultivation. The maximum expression level of Cel6A-(His) 6 in S. lividans supernatant reached up to 173 mg/L after 84-h of cultivation. The endoglucanase activity staining SDS-PAGE showed that there were some minor proteins in S. lividans supernatant which may be the Cel6A derivant by proteolytic degradation, while there was no proteolytic product detected in supernatant of P. pastoris.
Transient Ureteral Obstruction Prevents against Kidney Ischemia/Reperfusion Injury via Hypoxia-Inducible Factor (HIF)-2α Activation

Abstract: Although the protective effect of transient ureteral obstruction (UO) prior to ischemia on subsequent renal ischemia/reperfusion (I/R) injury has been documented, the underlying molecular mechanism remains to be understood. We showed in the current study that 24 h of UO led to renal tubular hypoxia in the ipsilateral kidney in mice, with the accumulation of hypoxia-inducible factor (HIF)-2α, which lasted for a week after the release of UO. To address the functions of HIF-2α in UO-mediated protection of renal IRI, we utilized the Mx-Cre/loxP recombination system to knock out target genes. Inactivation of HIF-2α, but not HIF-1α blunted the renal protective effects of UO, as demonstrated by much higher serum creatinine level and severer histological damage. UO failed to prevent postischemic neutrophil infiltration and apoptosis induction in HIF-2α knockout mice, which also diminished the postobstructive up-regulation of the protective molecule, heat shock protein (HSP)-27. The renal protective effects of UO were associated with the improvement of the postischemic recovery of intra-renal microvascular blood flow, which was also dependent on the activation of HIF-2α. Our results demonstrated that UO protected the kidney via activation of HIF-2α, which reduced tubular damages via preservation of adequate renal microvascular perfusion after ischemia. Thus, preconditional HIF-2α activation might serve as a novel therapeutic strategy for the treatment of ischemic acute renal failure.
Strategies for Optimization Expression of Heterologous Protein in Pichia pastoris

Abstract: Pichia pastoris is easy to genetically manipulate and can be grown to high cell densities, at the same time, P. pastoris is a eukaryote, and thereby has the potential for producing soluble, correctly folded recombinant protein with high yield, so the P. pastoris expression system is an ideal choice for the production of various heterologous proteins. However, at present not all the heterologous proteins can be successfully and efficiently expressed in P. pastoris, as a result, different proteins present different yield levels, bioactivity and stability. Here strategies for reducing proteolytic degradation and improving production of the expressed heterologous proteins are briefly summarized in terms of genetically factors and cultivation level.
Simulation of pedestrian evacuation flow with affected visual field using cellular automata

Abstract: Simulation of pedestrian evacuation flow with affected visual field is carried out on the basis of cellular automata. The room to be evacuated is divided into exit visible area, wall visible area and blind area by pedestrian sight radius, and two basic dynamic parameters in the dynamic parameters model are used to simulate the varied movement characteristics of pedestrian in different evacuation areas, including normal evacuation movement, movement along the wall and movement in fixed direction. The effect of pedestrian sight radius on evacuation time is studied with evacuation signs on the wall and pedestrian stochastically moving to find the wall in fixed directions and then moving along the wall. It is observed that the evacuation time depends not only on the pedestrian sight radius but also on the length and utilization rate of exit.

Abstract: The knowledge of subnuclear localization in eukaryotic cells is indispensable for under-standing the biological function of nucleus, genome regulation and drug discovery. In this study, a new feature representation was pro-posed by combining position specific scoring matrix (PSSM) and auto covariance (AC). The AC variables describe the neighboring effect between two amino acids, so that they incorpo-rate the sequence-order information; PSSM de-scribes the information of biological evolution of proteins. Based on this new descriptor, a support vector machine (SVM) classifier was built to predict subnuclear localization. To evaluate the power of our predictor, the benchmark dataset that contains 714 proteins localized in nine subnuclear compartments was utilized. The total jackknife cross validation ac-curacy of our method is 76.5%, that is higher than those of the Nuc-PLoc (67.4%), the OET- KNN (55.6%), AAC based SVM (48.9%) and ProtLoc (36.6%). Abstract: Using invariant sets of descending flow and variational methods, we establish some sufficient conditions on the existence of sign-changing solutions, positive solutions and negative solutions for second-order nonlinear difference equations with Dirichlet boundary value problem. Some results in the literature are improved.
