Histopathology and biochemistry analysis of the interaction between sunitinib and paracetamol in mice

Abstract: Background Sunitinib, a tyrosine kinase inhibitor to treat GIST and mRCC may interact with paracetamol as both undergo P450 mediated biotransformation and P-glycoprotein transport. This study evaluates the effects of sunitinib-paracetamol coadministration on liver and renal function biomarkers and liver, kidney, brain, heart and spleen histopathology. ICR male mice (n = 6 per group/dose) were administered saline (group-A) or paracetamol 500 mg/kg IP (group-B), or sunitinib at 25, 50, 80, 100, 140 mg/kg PO (group-C) or coadministered sunitinib at 25, 50, 80, 100, 140 mg/kg PO and paracetamol IP at fixed dose 500 mg/kg (group-D). Paracetamol was administered 15 min before sunitinib. Mice were sacrificed 4 h post sunitinib administration. Results Group-A serum ALT and AST levels were 14.29 ± 2.31 U/L and 160.37 ± 24.74 U/L respectively and increased to 249.6 ± 222.7 U/L and 377.1 ± 173.6 U/L respectively in group-B; group-C ALT and AST ranged 36.75-75.02 U/L and 204.4-290.3 U/L respectively. After paracetamol coadministration with low sunitinib doses (group-D), ALT and AST concentrations ranged 182.79-221.03 U/L and 259.7-264.4 U/L respectively, lower than group-B. Paracetamol coadministration with high sunitinib doses showed higher ALT and AST values (range 269.6-349.2 U/L and 430.2-540.3 U/L respectively), p < 0.05. Hepatic histopathology showed vascular congestion in group-B; mild congestion in group-C (but lesser than in group-B and D). In group-D, at low doses of sunitinib, lesser damage than in group-B occurred but larger changes including congestion were observed at high sunitinib doses. BUN levels were higher (p < 0.05) for group-B (33.81 ± 5.68 mg/dL) and group-D (range 35.01 ± 6.95 U/L to 52.85 ± 12.53 U/L) compared to group-A (15.60 ± 2.17 mg/dL) and group-C (range 17.50 ± 1.25 U/L to 26.68 ± 6.05 U/L). Creatinine remained unchanged. Renal congestion and necrosis was lower in group-C than group-B but was higher in group-D (p > 0.05). Mild cardiotoxicity occurred in groups B, C and D. Brain vascular congestion occurred at high doses of sunitinib administered alone or with paracetamol. Hepatic and renal biomarkers correlated with histopathology signs. Conclusions Paracetamol and sunitinib coadministration may lead to dose dependent outcomes exhibiting mild hepatoprotective effect or increased hepatotoxicity. Sunitinib at high doses show renal, cardiac and brain toxicity. Liver and renal function monitoring is recommended.

Abstract:

Purpose: To identify possible factors that influence sexual function in men undergoing maximal androgen deprivation therapy (ADT). Patients and Methods: A descriptive exploration was performed looking at characteristics of twenty-two men reporting sexual activity after nine months of maximal ADT. This previously published Phase II study, involved 250 prostate cancer patients undergoing intermittent ADT. An analysis between this cohort and the group that did not maintain sexual function was performed to ascertain if age, testosterone level, functional status or maintenance of quadriceps strength had an impact upon sexual function. Results: There was no difference in age, testosterone level or ECOG performance status between the sexually active and non-sexually active groups. Over the course of 9 months of ADT, the sexually active group appeared to maintain quadriceps muscle strength as measured with physical stands, and maintained overall health as measured by quality of life questionaries, compared to the non-sexually active group. Conclusions: This retrospective study suggests that exercise during ADT may reduce the impact of ADT on sexual function. This warrants further testing, and could be the focus of future randomised controlled trials.

Demographic Variables as Determinants of Women’s Participation in Child Nutrition Education in Osun State, Nigeria

Abstract: The study examined demographic variables as determinants of women participation in child nutrition education in Osun State, with a view to establishing the level of nutrients children have in breast-feeding and how child nutrition education impacted women participation and psychosocial development of mothers and children. The descriptive survey research design was employed. The random sampling technique was used to select 525 respondents which comprised mothers who brought their children to the hospitals and clinics. t-test was used to analyse the data There was significant difference in level of women participation with low and higher education in child nutrition awareness (t=1.96, df =525, p>0.5). There was no significant difference in the level of participation of unemployed and employed women in child nutrition awareness (t=.567, df =525,p>0.5). Similarly, there was significant difference between the level of participation of single and married women in child nutrition awareness’. (t=1.815, df =525, p>0.5. Therefore, the study recommended that every women, irrespective of age, level of education, level of employment, or marital status need to be educated and aware of the effects of nutrients children acquired in breast-feeding. Also, the government; non-governmental organizations, community workers and society at large should organize seminars and workshops on how to improve child nutrition practices.

Abstract: A sustained increase in intra-abdominal pressure (IAP) may result in abdominal compartment syndrome (ACS). This is a well documented complication in critically ill patients, but there appears to be a reluctance to routinely measure IAP in patients at high risk of developing intra-abdominal hypertension (IAH) and ACS. Relationship between stochastic inequalities and some classical mathematical inequalities

Abstract: The notions of association and dependence of random variables, rearrangements, and heterogeneity via majorization ordering have proven to be most useful for deriving stochastic inequalities. In this survey article we first show that these notions are closely related to three basic inequalities in classical mathematical analysis: Chebyshev's inequality, the Hardy-Littlewood-Pólya rearrangement inequality and Schur functions. We then provide a brief review of some of the recent results in this area. An overall objective is to illustrate that classical mathematical inequalities of this type play a central role in the developments of stochastic inequalities.

Savoirs mondains, savoirs savants : les femmes et leurs cabinets de curiosités au siècle des Lumières

Abstract: Dans la France des Lumières, la culture de la curiosité est un phénomène de mode mais surtout un jeu social et intellectuel. La présente étude entend retracer l’histoire d’une trentaine de cabinets féminins de curiosité tenus à cette époque. Des femmes fortunées de l’aristocratie et de la bourgeoisie parisienne et provinciale ont alors constitué sous l’emprise de leur libido sciendi des cabinets d’alchimie, de minéralogie, de physique chimie, d’histoire naturelle et d’anatomie naturelle et artificielle. Ces cabinets obéissent à une typologie particulière. Il en existe deux catégories : les cabinets d’amateurs, constitués pour la parade et le spectacle des visiteurs et fonctionnant comme de véritables écoles de plaisirs intellectuels et éducatifs ; les cabinets à finalité scientifique et didactique, formés par des savantes expérimentées qui se livrent dans leurs laboratoires à des recherches personnelles et expérimentales au nom des progrès de la science médicale et de l’instruction publique. La réflexion porte aussi sur le fonctionnement de ces cabinets privés de curiosité, particulièrement sur leur mode de constitution, leur décor intérieur ainsi que sur le contenu des collections qui nécessitaient certaines techniques d’organisation, d’acquisition et de conservation communes à celles de leurs homologues masculins. During the Enlightenment in France, curiosity culture constituted both a fashion and an intellectual and social game. This article explores thirty cabinets of curiosities run by women during this period. Wealthy women from the Parisiain and provincial aristocracy and middle classes organized cabinets in alchemy, mineralogy, physics and chemistry, natural history and biology studies. These cabinets can be divided into two distinct categories. The first represented amateur interests; they were developed for show and served as schools for intellectual and éducational pleasure. The second category were run by experienced women scientists who used their laboratories to conduct experimental research in the name of medical progress and public instruction. This article also describes how these private cabinets functioned, their composition, their interior décor as well as the contents of the collections, which required the organisation, acquisition and preservation techniques used in their male counterparts.

Abstract: Le métier de réalisateur de cinéma a été peu étudié en sciences humaines et sociales. Les études cinématographiques et la presse spécialisée s’intéressent beaucoup moins aux parcours des collaborateurs de l’équipe technique qu’aux comédiens et aux projets des auteurs que l’on pourrait croire retranchés dans leur intériorité créatrice. Pour échapper à ce travers biographique, le sociologue peut rendre compte de l’itinéraire d’un réalisateur en examinant ses liens avec ses collaborateurs. On prend ici l’exemple des liens de travail récurrents qui unissent Dominique Cabrera, réalisatrice de cinéma, à ses proches collaborateurs, en montrant de quelle manière des liens de travail peuvent s’approfondir en dépit d’un contexte sectoriel où l’organisation par projet de l’activité cinématographique favorise a priori la fugacité des relations de travail. On postule ainsi que le contexte pertinent pour éclairer les contours de cet itinéraire de carrière est le réseau des relations socioprofessionnelles au sein duquel naissent les projets de films et qui contribuent à ancrer ce parcours dans le paysage professionnel des pratiques cinématographiques. In France, social sciences and humanities have paid little attention to the profession of film director. Film studies and the press are much less interested in the professional careers of technical teams in comparison with the lives of actors and authors of projects who seem to be cut into their inner creativity. To escape the biographical pitfall, sociologists may account for a director’s itinerary by analyzing his/her ties with colleagues. Here, we examine the recurrent work relations between film director Dominique Cabrera and her close colleagues, to underline the strong relationships which paradoxically unite both the director and her staff in a project-based industry that is rather conducive to short-lived work relations. We postulate that the professional network produced consistently helps to describe and understand the course of a career, and that the relevant context to illuminate its contours is the network of professional relations that engenders film projects and is instrumental in fastening the career in the professional landscape of cinema.
