CXCR7 antagonism prevents axonal injury during experimental autoimmune encephalomyelitis as revealed by in vivo axial diffusivity

Abstract: C57BL6/J mice underwent adoptive transfer of MOG-reactive Th1 cells and were treated daily with either CXCR7 antagonist or vehicle for 28 days; and then evaluated by DTI to assess for axonal injury. After imaging, spinal cords underwent histological analysis of myelin and oligodendrocytes via staining with luxol fast blue (LFB), and immunofluorescence for myelin basic protein (MBP) and glutathione S-transferase-π (GST-π). Detection of non-phosphorylated neurofilament H (NH-F) was also performed to detect injured axons. Statistical analysis for EAE scores, DTI parameters and non-phosphorylated NH-F immunofluorescence were done by ANOVA followed by Bonferroni post-hoc test. For all statistical analysis a p < 0.05 was considered significant.In vivo DTI maps of spinal cord ventrolateral white matter (VLWM) axial diffusivities of na?ve and CXCR7 antagonist-treated mice were indistinguishable, while vehicle-treated animals exhibited decreased axial diffusivities. Quantitative differences in injured axons, as assessed via detection of non-phosphorylated NH-F, were consistent with axial diffusivity measurements. Overall, qualitative myelin content and presence of oligodendrocytes were similar in all treatment groups, as expected by their radial diffusivity values. Quantitative assessment of persistent inflammatory infiltrates revealed significant decreases within the parenchyma of CXCR7 antagonist-treated mice versus controls.These data suggest that CXCR7 antagonism not only prevents persistent inflammation but also preserves axonal integrity. Thus, targeting CXCR7 modifies both disease severity and recovery during EAE, suggesting a role for this molecule in both phases of disease.Axonal injury is a critical factor in the progression of neurologic deficits in patients with multiple sclerosis (MS) [1]. Cutaneous nociception evoked by 15-delta PGJ2 via activation of ion channel TRPA1

Abstract: Our search for endogenous chemical activators utilizing a bioactive lipid library screen identified a cyclopentane PGD2 metabolite, 15-deoxy-Δ12,14-prostaglandin J2 (15d-PGJ2), as a TRPA1 agonist. Similar to CA and AITC, this electrophilic molecule is known to modify cysteines of cellular target proteins. Electophysiological recordings verified that 15d-PGJ2 specifically activates TRPA1 and not TRPV1 or TRPM8 (thermoTRPs also enriched in DRG). Accordingly, we identified a population of mouse DRG neurons responsive to 15d-PGJ2 and AITC that is absent in cultures derived from TRPA1 knockout mice. The irritant molecules that activate TRPA1 evoke nociceptive responses. However, 15d-PGJ2 has not been correlated with painful sensations; rather, it is considered to mediate anti-inflammatory processes via binding to the nuclear peroxisome proliferator-activated receptor gamma (PPARγ). Our in vivo studies revealed that 15d-PGJ2 induced acute nociceptive responses when administered cutaneously. Moreover, mice deficient in the TRPA1 channel failed to exhibit such behaviors.In conclusion, we show that 15d-PGJ2 induces acute nociception when administered cutaneously and does so via a TRPA1-specific mechanism.The prostaglandins (PGs) are a class of biomolecules derived from arachidonic acid (AA) that are involved in a variety of signaling processes including inflammation. Abstract: Objective: To determine the effectiveness of antiplatelet and/or anticoagulant therapy (AP/AC) at reducing ischemic events in patients with Large Vessel Vasculitis (LVV). Methods: We performed a random effects meta-analysis of studies examining antiplatelet and/or anticoagulant therapy (AP/AC) and ischemic events in Takayasu’s Arteritis (TAK) or Giant Cell Arteritis (GCA). Severe ischemic events were defined as stroke, ischemic ocular manifestations and claudication symptoms. Any ischemic event included jaw claudication in addition to the above manifestations. Results: Seven studies met inclusion criteria: 1 TAK and 6 GCA. The majority of patients (>80%) were treated with ASA and treatment was initiated prior to diagnosis of LVV. Risk of severe and any ischemic event in patients with LVV treated with AP/AC versus no treatment was not significantly different (OR 0.570, 95% CI 0.243, 1.340 and OR 0.594, 95% CI 0.248, 1.421, respectively). For studies with follow-up data (26-76 months), AP/AC was protective for severe ischemic events (OR 0.18, 95% CI 0.04, 0.83). Findings were similar when excluding studies that did not account for potential confounders, such as cardiovascular risk factors. Conclusion: At follow-up, antiplatelet therapy significantly decreases ischemic events in patients with LVV. However, in most cases of GCA, the treatment was initiated prior to the diagnosis of vasculitis. The benefit of initiating anti-platelet therapy at the time of GCA diagnosis remains unclear.
Perceptions of Women on Marriage in Namibia  [PDF]

Abstract: Marital pattern in a Namibian society is predominantly of never married women, and the rates are increasing, while the proportion of those getting married is falling, nevertheless, these proportions decline as age increases. Quantitative research revealed differences in marital status by place of residence, education level and age group suggesting that aspects of culture, marriage practices and customs change over time. Focus group discussions of 6 - 9 participants were conducted to construct supplementary in-depth understanding of women’s feelings and attitudes towards marriage in Namibia. Results indicated that women felt that marriage was still important for family formation. The practice of early marriage was reported to be still happening especially in remote rural areas but was on the decline. Marital union choice was dictated by parental guidance, childhood sexual abuse, poverty, culture, age, and religion. Most marriages were not stable and divorces were increasing due to issues of infidelity, poor communication, inpatience and intolerance, step children, financial matters, alcohol and drug abuse, young age at marriage, witchcraft allegations, in-law relationships and even poor cooking. Cohabitation was promoted by exorbitant lobola, marriage costs, modernization, media influences, poverty, and flexibility. The risks associated with cohabiting were gender based violence and passion killings in the event of dissolution and lack of legal representation framework especially with respect to property in the event of partner death. Among the never married women, reasons for not marrying included absence of acceptable mate on the marriage market; need for independence, infertility, fear of abusive relationships, bad past experiences, heart aches, and lack of good married role models to inspire them to also marry.

Abstract: Este artículo examina la dinámica que se desarrolla entre las principales características del constructo Orientación Empresarial (OE) –autonomía, innovación, propensión al riesgo y agresividad competitiva– y las relaciones de redes en el proceso de expansión internacional de un negocio. Este estudio emplea el enfoque cualitativo de estudio de casos como metodología. La principal fuente de recopilación de datos fue la entrevista personal con gerentes, directores, gerentes de mercadeo y de exportaciones. Nuestros hallazgos sugieren que algunos componentes del constructo OE reflejados por todos los casos constituyen herramientas para desarrollar, mantener y alcanzar mercados foráneos. Las relaciones de redes se convierten en un componente vital de las características emprendedoras reflejadas por cada caso. This paper examines the dynamic that is generated between the main traits of Entrepreneurial Orientation (EO) –autonomy, innovativeness, risk-taking, proactiveness, and competitive aggressiveness– and network relationships for the international expansion of a business. The current study uses a qualitative case studies approach. The main form of data collection was personal interviews with predominantly managing directors, chief executive officers, export managers, and marketing managers. Our findings suggest that some components of the EO construct reflected by all of the cases are tools to exploit, maintain, and reach foreign markets. The network relationships become a vital component of the entrepreneurial traits reflected by each case.

Abstract: While it is evident that the integration that European countries are involved in through the European Union (EU) is beneficial in terms of labour and trade opportunities, the effect of such integration on financial development is unclear. This paper implements an innovative approach that examines the impact of financial integration on financial development, and subsequently on economic growth within a sample of EU countries, pre-financial crisis of 2008. Traditionally monetary depth measures are used to assess the size and development of the financial sector. However due to the centralized control of monetary policy, these monetary depth measures lose their effectiveness in assessing financial development within the EU. This paper therefore looks at bank-based measures of financial development in an effort to establish whether a relationship exists between financial development and growth in the European Union countries, and if so, whether this relationship has been affected by financial integration. The paper focuses on the time period before the 2008 financial crisis to avoid the impact of capital flight, failed banking sectors and budgetary issues. The results support the hypothesis that the benefits of economic and financial integration are not uniform. Financial integration does not seem to have a very significant effect on growth where financial development has been controlled for.

Baggrunden var et nske om at etablere et virtuelt l ringsmilj , hvor der er mulighed for andet en blot formidling fra administration og undervisere til de studerende. I denne artikel vil jeg belyse den udviklingsproces og nogle af de didaktiske overvejelser, der har v ret i spil under implementeringen af Moodle. Endvidere inddrages evalueringer foretaget undervejs, samt resultater vedr. brugen af Moodle, indsamlet et stykke inde i processen med henblik p at belyse de p dagogiske forandringer.
