Estimating firing rates from calcium signals in locust projection neurons in vivo

Abstract: Combining intracellular electrophysiology and multi-photon calcium imaging in vivo, we studied the relationship between calcium signals (sampled at 500–750?Hz) and spike output in principal neurons in the locust antennal lobe. Our goal was to determine whether the firing rate of individual neurons can be estimated in vivo with calcium imaging and, if so, to measure directly the accuracy and resolution of our estimates. Using the calcium indicator Oregon Green BAPTA-1, we describe a simple method to reconstruct firing rates from dendritic calcium signals with 80–90% accuracy and 50?ms temporal resolution.
Spectral Unmixing: Analysis of Performance in the Olfactory Bulb In Vivo

Abstract: Background The generation of transgenic mice expressing combinations of fluorescent proteins has greatly aided the reporting of activity and identification of specific neuronal populations. Methods capable of separating multiple overlapping fluorescence emission spectra, deep in the living brain, with high sensitivity and temporal resolution are therefore required. Here, we investigate to what extent spectral unmixing addresses these issues. Methodology/Principal Findings Using fluorescence resonance energy transfer (FRET)-based reporters, and two-photon laser scanning microscopy with synchronous multichannel detection, we report that spectral unmixing consistently improved FRET signal amplitude, both in vitro and in vivo. Our approach allows us to detect odor-evoked FRET transients 180–250 μm deep in the brain, the first demonstration of in vivo spectral imaging and unmixing of FRET signals at depths greater than a few tens of micrometer. Furthermore, we determine the reporter efficiency threshold for which FRET detection is improved by spectral unmixing. Conclusions/Significance Our method allows the detection of small spectral variations in depth in the living brain, which is essential for imaging efficiently transgenic animals expressing combination of multiple fluorescent proteins.

Abstract: the subject of this essay is the dilemmas of modernization such as viewed by sérgio buarque de holanda and edward c. banfield in raízes do brasil and the moral basis of a backward society. the comparative analysis searches to elucidate the concepts of cordiality and amoral familism as stated by the authors. when referred to their own theoretical models, such concepts are described as expressions of private ethic traditions which are not compatible with the social ethos demanded for the entrance in the modern age. within this scope, the conclusion is that banfield's vision states that a private ethic tradition shall be annulled by the modernizing action, whereas sérgio buarque de holanda seeks to reconcile them by setting a balance between tendencies which are apparently antagonistic.
