An Application of Molecular Genotyping in Mice

Abstract: Microsatellite markers are simple sequence repeats within the mammalian genome that can be used for identifying disease loci, mapping genes of interest as well as studying segregation patterns related to meiotic nondisjunction. Different strains of mice have variable CA repeat lengths and PCR based methods can be used to identify them, thus allowing for specific genotypes to be assigned. Molecular genotyping offers such identification at any developmental stage, which allows for a broad range of anomalies to be studied. We studied chromosomal segregation in relation to nondisjunction in early-gestation mouse embryos using molecular genotyping. Information on the parental origin as well as the number of chromosomes a given progeny carried was obtained in our analysis.
Microarray Data Reveal Relationship between Jag1 and Ddr1 in Mouse Liver

Abstract: Alagille syndrome is an autosomal dominant disorder involving bile duct paucity and cholestasis in addition to cardiac, skeletal, ophthalmologic, renal and vascular manifestations. Mutations in JAG1, encoding a ligand in the Notch signaling pathway, are found in 95% of patients meeting clinical criteria for Alagille syndrome. In order to define the role of Jag1 in the bile duct developmental abnormalities seen in ALGS, we previously created a Jag1 conditional knockout mouse model. Mice heterozygous for the Jag1 conditional and null alleles demonstrate abnormalities in postnatal bile duct growth and remodeling, with portal expansion and increased numbers of malformed bile ducts. In this study we report the results of microarray analysis and identify genes and pathways differentially expressed in the Jag1 conditional/null livers as compared with littermate controls. In the initial microarray analysis, we found that many of the genes up-regulated in the Jag1 conditional/null mutant livers were related to extracellular matrix (ECM) interactions, cell adhesion and cell migration. One of the most highly up-regulated genes was Ddr1, encoding a receptor tyrosine kinase (RTK) belonging to a large RTK family. We have found extensive co-localization of Jag1 and Ddr1 in bile ducts and blood vessels in postnatal liver. In addition, co-immunoprecipitation data provide evidence for a novel protein interaction between Jag1 and Ddr1. Further studies will be required to define the nature of this interaction and its functional consequences, which may have significant implications for bile duct remodeling and repair of liver injury.

Abstract: Es conocido que el movimiento tridimensional en las proximidades del asteroide (433) Eros es caótico para un amplio abanico de inclinaciones. La principal característica de este asteroide es su forma alargada, lo que hace que los procedimientos habituales de desarrollar el potencial en armónicos esféricos no tenga mucha utilidad. Por tal razón, consideramos un modelo simple que aproxime su forma alargada; en concreto, tomamos una varilla alargada y en rotación. De este modo se puede obtener el potencial de forma cerrada, sin necesidad de desarrollos. Con este modelo tan simple y mediante el cálculo de familias de orbitas periódicas, podemos obtener una explicación para la mayor a de las características dinámicas previamente halladas para el movimiento orbital alrededor de Eros.

Regularity for fully non linear equations with non local drift

Abstract: We prove Holder regularity for solutions of non divergence integro-differential equations with non necessarily even kernels. The even/odd decomposition of the kernel can be understood as a sum of a diffusion and a drift term. In our case we assume that such drift have the order smaller than or equal to the diffusion and at least one. For example we can say something about the following equation $\Delta^{1/2}u + |Du| = f$. The main step relies in a localized version of the Aleksandrov-Bakelman-Pucci estimate. Our estimates are also uniform as the order of the equation goes to two.

Abstract:

Purpose: The purpose of the present study was to compare the effect of intramuscular masseter administration of methylprednisolone using a split-mouth design, as a single 20-mg dose, after removal of impacted lower third molars. Patients and Methods: A non-blind, cross-over, comparative, non-randomized, clinical trial was planned. The sample was composed of 32 patients requiring extraction under local anesthesia of two lower bony impacted mandibular third molars. The difficulty of extraction was similar in all cases. The patients received 20 mg of methylprednisolone injected into the masseter muscle via the intrabuccal approach, immediately after suturing of the surgical wound on the right side. The left side received no intramuscular corticoid. Evaluations were made of postoperative pain and swelling. Results: A total of 32 subjects requiring surgical removal of two impacted mandibular third molars under local anesthesia were included in the present study. The patients administered methylprednisolone showed superior results after surgery in terms of pain and facial swelling parameters using self-evaluation, with statistically significant differences versus the control-side (p < 0.05). The results obtained show that 20 mg of methylprednisolone injected into the masseter muscle in the immediate postoperative period reduces swelling and pain. Conclusions: Injection of methylprednisolone is an effective therapeutic strategy to reduce swelling and pain after surgical removal of impacted lower third molars. It offers a simple, safe, painless, noninvasive, and cost effective therapeutic option for moderate and severe cases.
