Abstract: Dicranostigma Leptopodum (Maxim) Fedde (DL- F), which had been previously documented to suppress oxidative hemolysis of erythrocytes and enhance immune functions of murine peri- toneal macrophages, was investigated for its effect on anti-tumor activity. Of alkaloids extracted from DLF, five have been identified with employment of chromatographic analysis. An antiproliferative role of these alkaloids was determined on SMMC-7721 Human Hepatoma Ce- lls in an apoptosis-inducing manner, through MTT assaying, Trypan blue exclusion assaying and cytometric analysis of cell cycle distribution. To further examine their inhibitory effects on tumor progression, murine H22 cells were inoculated into Kunming mice to determine the role of these alkaloids of DLF in inhibiting tumor growth in the tumor-implanted mice. It was found that these alkaloids of DLF enhanced the tumor shrinkage effectively wherein its tumor inhibitory rate and immunohistochemistry stain- ing of the tumor were determined and profiled, respectively.
Caffeine-Induced Ca2+ Oscillations in Type I Horizontal Cells of the Carp Retina and the Contribution of the Store-Operated Ca2+ Entry Pathway

Abstract: The mechanisms of release, depletion, and refilling of endoplasmic reticulum (ER) Ca2+ were investigated in type I horizontal cells of the carp retina using a fluo-3-based Ca2+ imaging technique. Exogenous application of caffeine, a ryanodine receptor agonist, induced oscillatory intracellular free Ca2+ concentration ([Ca2+]i) responses in a duration- and concentration-dependent manner. In Ca2+-free Ringer’s solution, [Ca2+]i transients could also be induced by a brief caffeine application, whereas subsequent caffeine application induced no [Ca2+]i increase, which implied that extracellular Ca2+ was required for ER refilling, confirming the necessity of a Ca2+ influx pathway for ER refilling. Depletion of ER Ca2+ by thapsigargin triggered a Ca2+ influx which could be blocked by the store-operated channel inhibitor 2-APB, which proved the existence of the store-operated Ca2+ entry pathway. Taken together, these results suggested that after being depleted by caffeine, the ER was replenished by Ca2+ influx via store-operated channels. These results reveal the fine modulation of ER Ca2+ signaling, and the activation of the store-operated Ca2+ entry pathway guarantees the replenishment of the ER so that the cell can be ready for response to the subsequent stimulus.

Abstract:

Liver cirrhosis portal hypertension patients to reduce the number of blood cells are common in clinical, and often affect the prognosis. This paper discusses cirrhotic portal hypertension patients complicated by the reason of the decrease in the number of peripheral blood cells and what is the clinical significance of these reasons so as to provide theoretical support for the choice of treatment. Splenomegaly and hypersplenism caused should be the main reason for reducing the number of blood cells, but not all, other reasons are alcohol and virus inhibition of bone marrow, liver function impairment, autoimmune damage and loss of blood, etc. If it is a function of the spleen hyperfunction caused by blood cells decreases, blood should rise to normal after splenectomy, or consider other reason or there are other reasons at the same time.

Abstract: Multi-core processor provides multiple threads parallel execution capability, and makes applications to have huge potential for performance improvement, but makes it enormously challenge to efficiently develop high-performance program Meanwhile, through the old process of performance optimization, the scalability is difficult to be guarantee.From application point of view,attributing core calculation to patterns and motifs,and optimizing these motifs can produce reusable library that can support efficient develop new application,and also can guarantee the scalability of the application performance. A layered parallel computing model was used for guidance in this article. From the perspective of application-driven parallel program performance optimization, this article designed a new parallel multi-core processor computing modcl,which can be used in the architecture of multi core processor chip. Based on this model,g-scan algorithm was designed which has good extendibility together with high performance after analyzing and optimizing some fundamental parallel algorithms. At the last, newly designed parallel algorithm was applied to OpenSeesSP which is a finite element software widely used in structure engineering.
Optimization of technology for dietary fiber extraction from Maixiansan by response surface methodology

Abstract: A Box–Behnken design was employed to optimize the extraction parameters, including α-amylase concentration (X1: 0.3 – 0.5%), enzymolysis time (X2: 30 – 60 min) and NaOH content (X3: 1.0 – 5.0%), of dietary fiber from Maixiansan using an enzyme–alkali extraction technique.The optimal technological conditions were as follows: α-amylase concentration: 0.4%; enzymolysis time: 45 min; NaOH content: 4.0%. Under these conditions, the extraction yield reached 57.14%, which was well consistent with the predicted models with a coefficient of determination (R2) of 0.9818. An evaluation of the anti-inflammatory activity indicated that Maixiansan was able to significantly inhibit dextran sodium sulfate-induced ulcerative colitis in rats by increasing the concentration of short-chain fatty acids (acetate, propionate and butyrate), among which the butyrate content was significantly higher in the Maixiansan group than in the other groups.Our experiments showed that response surface methodology can optimize the extraction of dietary fiber from Maixiansan. Maixiansan could be explored as an anti-ulcerative colitis agent.Inflammatory bowel disease includes Crohn’s disease and ulcerative colitis (UC). UC is a chronic inflammatory disorder of the colon characterized by alternating periods of flare-ups and quiescent disease [1]. Dietary fiber is an integral constituent of normal human nutrition and may contribute to a healthy colonic environment [2]. The protective effects of a fiber-rich diet have been attributed to short-chain fatty acids (SCFAs), including acetate, propionate and butyrate [3]. SCFAs are the major end products of fiber fermentation in the gastrointestinal tract [4-6]. Abstract: Through the performance analysis of an open source finite clement software based on MPI program model on multi core cluster platform, some performance bottlenecks were founded. Based on the performance bottleneck analysis,two optimization plans based on MPI/OpenMP hybrid parallel program model were proposed, one of them resolves the inefficiency in solving linear or nonlinear system equations, and the other one elevates processes communication performance. Experiment results show that hybrid parallel solver can efficiently promote the pure MPI based parallel program performance, as up to 3 times. I}he multi-thread multi-process communication plan can do some optimization, but is not the best solution in this case. The overall optimized performance analysis indicates that on multi-core cluster computing platform, MPI/OpenMP parallel program model can more efficiently utilize hardware system computation resource.

Abstract: AIM: To investigate the role of hepatic peroxisome proliferator-activated receptor-γ (PPAR-γ) in increased susceptibility to endotoxin-induced toxicity in rats with bile duct ligation during endotoxemia. METHODS: Male Sprague-Dawley rats were subjected to bile duct ligation (BDL). Sham-operated animals served as controls. DNA binding were determined by polymerase chain reaction, Western blotting analysis, and electrophoretic mobility shift assay, respectively. BDL and sham-operated rats received a non-lethal dose of intraperitoneal lipopolysaccharide (LPS) injection (3 mg/kg, i.p.). Additionally, the potential beneficial effects of the PPAR-γ agonist rosiglitazone were determined in BDL and sham-operated rats treated with a non-lethal dose of LPS. Survival was assessed in BDL rats treated with a non-lethal dose of LPS and in sham-operated rats treated at a lethal dose of LPS (6 mg/kg, i.p.). RESULTS: PPAR-γ activity in rats undergoing BDL was significantly lower than in the sham-controls. Hepatic PPAR-γ gene expression was downregulated at both the mRNA and protein levels. In a parallel group, serum levels of pro-inflammatory cytokines were nearly undetectable in the sham-operated rats. When challenged with a non-lethal dose of LPS (3 mg/kg), the BDL rats had approximately a 2.4-fold increase in serum IL-6, a 2.7 fold increase in serum TNF-α, 2.2-fold increase in serum IL-1 and 4.2-fold increase in serum ALT. The survival rate was significantly lower as compared with that in sham-operated group. Additionally, rosiglitazone significantly reduced the concentration of TNF-α, IL-1β, IL-6 and ALT in sham-operated rats, but not in BDL rats, in response to LPS (3 mg/kg). Also, the survival was improved by rosiglitazone in sham-operated rats challenged with a lethal dose of LPS, but not in BDL rats, even with a non-lethal dose of LPS (3 mg/kg). CONCLUSION: Obstructive jaundice downregulates hepatic PPAR-γ expression, which in turn may contribute to hypersensitivity towards endotoxin.

Abstract: The shallow decay phase of the early X-ray afterglow in gamma-ray bursts discovered by {\it Swift} is a widely discussed topic. As the spectral index does not change at the transferring of the shallow decay phase to a normal phase, it implies this transferring should be a dynamical change rather than a spectral evolution. We suggest both the shallow decay phase and the normal phase are from the external shock in a wind environment, while the transferring time is the deceleration time. We apply this model to GRBs 050319 and 081008, and find them can be well explained by choosing a proper set of parameters.
