Nitrilases in nitrile biocatalysis: recent progress and forthcoming research

Abstract: Over the past decades, nitrilases have drawn considerable attention because of their application in nitrile degradation as prominent biocatalysts. Nitrilases are derived from bacteria, filamentous fungi, yeasts, and plants. In-depth investigations on their natural sources function mechanisms, enzyme structure, screening pathways, and biocatalytic properties have been conducted. Moreover, the immobilization, purification, gene cloning and modifications of nitrilase have been dwelt upon. Some nitrilases are used commercially as biofactories for carboxylic acids production, waste treatment, and surface modification. This critical review summarizes the current status of nitrilase research, and discusses a number of challenges and significant attempts in its further development. Nitrilase is a significant and promising biocatalyst for catalytic applications.
Preparation of Spherical UO2 Ceramic Particles by Sol-Gel Method

Abstract: The spherical UO2 ceramic particles were prepared by a sol-gel method with U3O8 powders as raw material, which combines advantages of external gelation and internal gelation methods. The process includes steps of sol preparation, gel particles forming, aging, ashing, drying, calcining, reducing, sintering, sieving and separating. The crystal form of the ceramic particle is face-centered cubic, and its density reaches 98.18% of the theoretical density value. All of the O/U ratio, diameter, sphericity and odd-shaped particles fraction can reach the design specification. Influences of many factors are also analyzed.

Bioassay-Guided Isolation of DPP-4 Inhibitory Fractions from Extracts of Submerged Cultured of Inonotus obliquus

Abstract: Inonotus obliquus is a medicinal mushroom used in Russian and Eastern European folk medicine for the treatment of gastrointestinal cancer, cardiovascular disease and diabetes. Previous studies in our laboratory have demonstrated that the mycelium powders of I. obliquus possess significant antihyperglycemic effects in a mouse model of diabetic disease induced by alloxan. However, the active ingredients of mycelium powders responsible for the diabetes activity have not been identified. This study aims to identify the active ingredients of I. obliquus mycelium powders by a bioassay-guided fractionation approach and explore the mechanism of action of these active ingredients by using a well-established DPP-4 (an important enzyme as a new therapeutic target for diabetes) inhibitory assay model. The results showed the chloroform extract of mycelium was potential inhibitory against DPP-4. Bioactivity guided fractionation led to the identification of 19 compounds using UPLC-Q-TOF-MS. Molecular docking between the compounds and DPP-4 revealed that compounds 5, 8, 9, 14, 15 may be the active components responsible for the DPP-4 inhibitory activity.

Abstract: A numerically robust and low-complexity method of signal subspace estimation is proposed in the paper. The transform matrix to tridiagonalize the covariance matrix of observation data is constructed in the forward recursion of multistage Wiener filter (MSWF), and its columns span the signal subspace. Compared with the traditional method of correlation subtractive structure, the forward recursion in the method is implemented with the Householder unitary transform. Therefore, it strengthens significantly the orthogonality of basis vectors in the signal subspace and improves the numerical robustness, especially in the finite-precision implementation. Besides, a method of calculating the transform matrix is proposed to reduce the computational complexity based on the unitary property of the Householder matrix and backward accumulation of matrices. Finally, simulation results demonstrate the numerical robustness and computational efficiency of the proposed method.

Abstract: This paper first analyzed the heavy-tailed distribution of gradients in natural scenes and proposed an algorithm about motion deblurring based on hyper-Laplacian moldel.It adopted an alternating minimization scheme to optimize the energy equation.Used a look-up table(LUT) for rapidly solving the image deconvolution.Our method can yield good result with medium size pictures in seconds.Compared with the Wiener and Richardson-Lucy methods, our method yields the best overall performance both in the restoration effect and suppression of ringing.

Abstract: Outdoor exposure ageing test and artificial accelerated ageing test of high-strength polyethylene(HSPE) monofilament without stabilizers and antioxidants were studied in laboratory.As a result,there are some differences in ageing stability between HSPE and common polyethylene(PE) monofilament,according to preservation rate of breaking strength and preservation rate of percentage of breaking elongation under given conditions,the former has a superiority in ageing stability,the ageing stability of HSPE monofilament is better than that of common PE monofilament.The results got from outdoor exposure ageing test and artificial accelerated ageing test are consistent on the whole.
