The Trichoderma koningii aggregate species

Abstract: The morphological concept of Trichoderma koningii is found to include several species that differ from each other in details of phenotype (including conidium morphology, growth rate) and biogeography. Phylogenetic analysis utilizing partial sequences of the translation-elongation factor 1 alpha (tef1), as well as fragments of actin and calmodulin genes, indicate that phenotypic characters typical of T. koningii evolved independently in three well-separated main lineages. Combined molecular and phenotype data lead to the development of a taxonomy with the recognition of twelve taxonomic species and one variety within the three lineages. These lineages include: (1) T. koningii and T. ovalisporum and the new species T. caribbaeum var. caribbaeum, T. caribbaeum var. aequatoriale, T. dorotheae, T. dingleyae, T. intricatum, T. koningiopsis, T. petersenii and T. taiwanense; (2) the new species T. rogersonii and T. austrokoningii, and (3) the new anamorph T. stilbohypoxyli. Trichoderma koningii s. str. is an uncommon species restricted to Europe and eastern North America; T. caribbaeum var. aequatoriale, T. koningiopsis, and T. ovalisporum were isolated as endophytes of trunks of Theobroma species in tropical America, and T. ovalisporum from the woody liana Banisteropsis caapi in Ecuador; T. koningiopsis is common in tropical America but was isolated also from natural substrata in East Africa, Europe and Canada, and from ascospores in eastern North America, and as an endophyte in Theobroma species; T. stilbohypoxyli, originally described as a parasite of Stilbohypoxylon species in Puerto Rico, is found to be more common in the tropics, besides an endophytic isolate from Fagus in U.K. The additional new species are known almost exclusively from their teleomorphs. Isolates of T. ovalisporum and T. koningiopsis may have biological control potential. A morphophenetic key and a set of tools for molecular species identification were developed
Hypocrea rufa/Trichoderma viride: a reassessment, and description of five closely related species with and without warted conidia

Abstract: The type species of the genus Hypocrea (Hypocreaceae, Hypocreales, Ascomycota, Fungi), H. rufa, is re-defined and epitypified using a combination of phenotype (morphology of teleomorphs and anamorphs, and characteristics in culture) and phylogenetic analyses of the translation-elongation factor 1α gene. Its anamorph, T. viride, the type species of Trichoderma, is re-described and epitypified. Eidamia viridescens is combined as Trichoderma viridescens and is recognised as one of the most morphologically and phylogenetically similar relatives of T. viride. Its teleomorph is newly described as Hypocrea viridescens. Contrary to frequent citations of H. rufa and T. viride in the literature, this species is relatively rare. Although both T. viride and T. viridescens have a wide geographic distribution, their greatest genetic diversity appears to be in Europe and North America. Hypocrea vinosa is characterised and its anamorph, T. vinosum sp. nov., is described. Conidia of T. vinosum are subglobose and warted. The new species T. gamsii is proposed. It shares eidamia-like morphology of conidiophores with T. viridescens, but it has smooth, ellipsoidal conidia that have the longest L/W ratio that we have seen in Trichoderma. Trichoderma scalesiae, an endophyte of trunks of Scalesia pedunculata in the Galapagos Islands, is described as new. It only produces conidia on a low-nutrient agar to which filter paper has been added. Additional phylogenetically distinct clades are recognised and provisionally delimited from the species here described. Trichoderma neokoningii, a T. koningii-like species, is described from a collection made in Peru on a fruit of Theobroma cacao infected with Moniliophthora roreri.

Abstract: Dichotomophthora was erected by Mehrlich & Fitzpatrick in 1935.Its distinctive characteristics are regularly dichotomously branched or lobed at the apex of conidiophores and brown,elongate-ovoid to oblong conidia.Dichotomophthora is somewhat similar to Dichotomophthoropsis,but differs in having elongate-ovoid and straight conidia,presenting a striking contrast to helicoid or clavate conidia of the latter.The genus included two species,D.lutea(Routien)de Hoog & Oorschot and D.portulacae Mehrlich & Fitzpatrick ex M.B.Ellis.D.lutea was first described as Dactylaria lutea by Routien in 1957 and then moved to Dichotomophthora by de Hoog & Oorschot in 1983according to dichotomously branched conidiophores.There was additional species named D.indica Rao characterised by unbranched,rarely once dichotomously branched or forked,short,stout,lobed phialides borne on longer and stouter conidiophores,but it was not accepted by many mycologists and treated as synonym of D.lutea.D.portulacae was the pathogen which caused stem and leaf disease of Portulaca oleracea Linn.(Mehrlich & Fitzpatrick 1935; Klisiewicz et al.1983; Mitchell1986; Eken 2003)and was being tested in tomato and pepper crops in Florida for its ability to manage common purslane(Klisiewicz 1985).During a general survey of endophytic microfungi from Basella rubra Linn.at Chengyang district,Qingdao,Shandong province of China,D.portulacae was isolated from leaves of the plant for the first time in August 2006 and 2007.Isolates of the species studied were deposited in Mycology Herbarium at Qingdao Agricultural University(MHQAU).

Abstract: Five species of higher marine fungi were observed on the incubated drift and intertidal woods collected from the coasts of Yellow Sea and Bohai Sea. Among them, Halosphaeriopsis was a genus newly recorded for China. Taxonomy and morphology of these species were discussed in this paper. The specimens were deposited in Mycology Herbarium at Qingdao Agricultural University (MHQAU).
PPM1B and P-IKKβ expression levels correlated inversely with rat gastrocnemius atrophy after denervation

Abstract: activated inhibitor of nuclear factor-κb kinase β (ikkβ) is necessary and sufficient for denervated skeletal muscle atrophy. although several studies have shown that mg2+/mn2+-dependent protein phosphatase 1b (ppm1b) inactivated ikkβ, few studies have investigated the role of ppm1b in denervated skeletal muscle. in this study, we aim to explore the expression and significance of ppm1b and phosphorylated ikkβ (p-ikkβ) during atrophy of the denervated gastrocnemius. thirty young adult female wistar rats were subjected to right sciatic nerve transection and were sacrificed at 0 (control), 2, 7, 14, and 28 days after denervation surgery. the gastrocnemius was removed from both the denervated and the contralateral limb. the muscle wet weight ratio was calculated as the ratio of the wet weight of the denervated gastrocnemius to that of the contralateral gastrocnemius. rt-pcr and western blot analysis showed that mrna and protein levels of ppm1b were significantly lower than those of the control group at different times after the initiation of denervation, while p-ikkβ showed the opposite trends. ppm1b protein expression persistently decreased while p-ikkβ expression persistently increased for 28 days after denervation. ppm1b expression correlated negatively with p-ikkβ expression by the spearman test, whereas decreasing ppm1b expression correlated positively with the muscle wet weight ratio. the expression levels of ppm1b and p-ikkβ were closely associated with atrophy in skeletal denervated muscle. these results suggest that ppm1b and p-ikkβ could be markers in skeletal muscle atrophy.
Some insights into clinical detection and analysis of serum lipoprotein for studies on integrated traditional Chinese and Western medicine

Abstract: Dyslipoproteinemia is correlated with the occurrence and development of atherosclerosis and cardiocerebral vascular diseases. The metabolism of serum lipoprotein, however, is a complicated and ordered metabolic process. On one hand, every organ, tissue and even cell of the body participates in and affects the state of serum lipoprotein dynamic balance (SLDB) in different degrees. On the other hand, SLDB is a direct factor for maintaining the normality of whole vascular system, especially the vascular endothelium system. The serum lipoprotein dynamic imbalance, as an inducement or even etiological factor, can bring about various clinical diseases. Therefore, integrated analysis is needed to appropriately assess the state of SLDB. It is suggested that serum lipoprotein dynamic imbalance syndrome should be the key risk factor for those related diseases. Serum lipoprotein electrophoretogram can ideally express the state of SLDB and be taken as a new index for cognizing the serum lipid metabolism and the function of speen-qi to distribute Jing(Essonse) in the integrated traditional Chinese and western medicine. Practice has showed that concrete integration and bilateral development should be one of the main tasks in current researches of integrated traditional Chinese and western medicine.

Abstract: During investigation and identification of fungi from Wuyi Mountains, Fujian Province, Huangshan Mountains, Anhui Province and Kunyushan Mountains, Shandong Province, three new Chinese records of Phaeosphaeria were found. They are Phaeosphaeria humerata, P. marciensis and P. typharum. Phaeosphaeria are usually saprophytic on dead stems or other wood of monocotyledon of Poaceae, Cyperaceae and Juncus. In this paper, morphological characters, illustrations and discussions of the species are provided and the specimens were deposited at mycology laboratory of Qingdao Agricultural University (MHQAU).

Abstract: During investigation of saprophytic fungi on deadwoods collected from Shandong Peninsula, four new Chinese records of Leptosphaeria agnita, L. dumetorum, L. eustomoides and L. solani were found. In this paper, these species are described, illustrated and discussed, and a key to reported saprophytic species of Leptosphaeria from China is provided. The specimens are deposited in the Mycology Lab of Qingdao Agricultural University.
