Absorption and transport of 6 coumarins isolated from the roots of Angelica pubescens f. biserrata in human Caco-2 cell monolayer model

Abstract: Objective: To study the absorption and transepithelial transport of six coumarins (umbelliferone, osthole, columbianadin, columbianetin acetate, angelol-A and angelol-B, isolated from the roots of Angelica pubescens f. biserrata) in the human Caco-2 cell monolayer model.Methods: The in vitro cultured human colon carcinoma cell line, Caco-2 cell monolayer model, was applied to study the absorption and transport of the six coumarins from apical (AP) to basolateral (BL) side and from BL to AP side. The six coumarins were measured by reversed-phase high-performance liquid chromatography (HPLC) coupled with ultraviolet absorption detector. Transport parameters and apparent permeability coefficients (Papp) were calculated and compared with those of propranolol as a control substance of high permeability and atenolol as a control substance of poor permeability. The transport mechanism of angelol-B was assayed by using iodoacetamide as a reference standard to inhibit ATP-dependent transport and MK571 as a well-known inhibitor of MRP2.Results: The absorption and transport of six coumarins were passive diffusion as the dominating process. The Papp values of umbelliferone, osthole, columbianadin, columbianetin acetate, angelol-A and angelol-B from AP to BL side were (2.679±0.263)×10-5, (1.306±0.324)×10-5, (0.595±0.086)×10-6, (2.930±0.410)×10-6, (1.532±0.444)×10-5 and (1.413±0.243)×10-5 cm/s, and from BL to AP side were (3.381±0.410)×10-5, (0.898±0.134)×10-5, (0.510±0.183)×10-6, (0.222±0.025)×10-6, (1.203±0.280)×10-5 and (0.754±0.092)×10-5 cm/s, respectively. In this assay, the Papp value of propranolol was 2.18×10-5 cm/s and the Papp value of atenolol was 2.77×10-7 cm/s. Among the 6 coumarins, the Papp values of umbelliferone, osthole, angelol-A and angelol-B from AP to BL side were identical with that of propranolol, and columbianadin and columbianetin acetate lied between propranolol and atenolol. When replaced the HBSS with EBSS, and iodoacetamide or MK-591 were used in the experiment, the Papp of angelol-B had no statistical difference as compared with the control group. In the mean total recoveries, umbelliferone was (83.31±3.52) %, angelol-A was (77.39±7.38) %, osthole, columbianadin and angelol-B were between 50% to 65%, and columbianetin acetate was lower than 10%. The accumulation rates of osthole and columbianadin in the Caco-2 cells were (36.15±5.87) % and (53.90±4.39) %, respectively.Conclusion: The absorption and transport of umbelliferone, osthole, columbianadin, columbianetin acetate, angelol-A and angelol-B are passive diffusion as the dominating p

Abstract: The problem on study of the active and toxic constituents of Chinese materia medica is the core of the problems in the field of modern research of Chinese materia medica. But approved methods for ascertaining the active and toxic constituents in Chinese materia medica have not been developed yet and are in developing phases. In combination with the characteristics of Chinese materia medica in clinical use and the achievements of modern pharmaceutical research, series model may be a finer approach to make certain of the active and toxic constituents from Chinese materia medica, which includes the parent compound biotransformation by human intestinal bacteria to make biotransformation products, and the absorption and further biotransformation in intestinal wall, the metabolization in the liver, and the secretion by the kidney of the parent compounds and biotransformation products. This method is also an important research assignment in the field of experimental medicine study of Chinese materia medica.
Establishment of Caco-2 cell monolayer model and standard operation procedure for assessing intestinal absorption of chemical components of traditional Chinese medicine

Abstract: Objective: To establish Caco-2 (a human colon adenocarcinoma cell line) cell monolayer model and the standard operation procedure for studying and assessing intestinal absorption of chemical components of traditional Chinese medicine.Methods: Caco-2 cell monolayer model was established and evaluated by morphology feature using scanning electron microscope, inverted microscope and transepithelial electrical resistance (TEER) assay. Additionally, the model was further tested for the activity of alkaline phosphatase and the apparent permeability (Papp) of standard compounds, i.e. propranolol and atenolol, which were the control substances for high and poor transcellular transport marker, respectively.Results: The integrality of cell monolayer, cell differentiation (reflected by expression of alkaline phosphatase and cell monolayer morphology), and the Papp value of standard compounds in the established Caco-2 cell model were satisfactory. All parameters tested were in good agreement with those reported in the literature.Conclusion: The established Caco-2 cell model can be used to study the intestinal absorption of orally administrated chemical components of traditional Chinese medicine and their absorption mechanism.
Inhibitory effects of 11 coumarin compounds against growth of human bladder carcinoma cell line E-J in vitro

Abstract: Objective: To screen antitumor active compounds, drug-like or leading compounds from Chinese traditional and herbal drugs. Methods: Eleven coumarin compounds isolated from the Chinese traditional and herbal drugs were studied for their antitumor activities in vitro by determining the inhibition rates against growth of human bladder carcinoma cell line E-J. Results: It showed that umbelliferone, scoparone, demethylfuropinarine, isopimpinellin, forbesoside, columbianadin, decursin and glycycoumarin inhibited the growth of human bladder carcinoma cell line E-J in vitro and their activities showed a concentration-effect relationship. The inhibitory effects of forbesoside, columbianadin, decursin and umbelliferone, with IC50 values of 7.50×10-7, 2.30×10-6, 6.00×10-6 and 1.30×10-6 mol/L, respectively, were stronger than those of the other tested compounds. However, xanthotoxin, esculin and sphondin did not inhibit the growth of human bladder carcinoma cell line E-J in this assay condition. Conclusion: These findings indicate that forbesoside, columbianadin, esculin, decursin and umbelliferone would be effective or regarded as potent drug-like or leading compounds against human bladder carcinoma.

Abstract: This paper presents a predictive variable structure control method for the control and synchronization of chaotic nonlinear dynamic systems based on a unified frame for both the control and the synchronization. Two controllers are designed. One is the predictive controller for nominal model, and the other is a variable structure controller for system perturbation. The tracking control of reference signal is realized, and the method has the merit of robustness of variable structure control. The effectiveness of the proposed method are proved by numerical simulations.
Absorption and transport of pachymic acid in the human intestinal cell line Caco-2 monolayers

Abstract: Objective: To study the absorption and transport of pachymic acid (PA) isolated from the sclerotium of Poria cocos (Schw.) Wolf. in human intestinal epithelium.Methods: By using Caco-2 (the human colonic adenocarcinoma cell lines) cell monolayers as an intestinal epithelial cell model, the permeability of PA was studied from apical side (AP side) to basolateral side (BL side) or from BL side to AP side. The PA was measured by reversed-phase high performance liquid chromatography coupled with UV detector at maximum absorption wavelength of 210 nm. Transport parameters and apparent permeability coefficients (Papp) were then calculated and compared with those of propranolol and atenolol, which were the transcellular transport markers for high and poor permeability respectively.Results: The Papp values of PA were (9.50±2.20) 10-7 cm/s from AP side to BL side, and (11.30±5.90) 10-7 cm/s from BL side to AP side, respectively. Under the condition of this experiment, the Papp values were 1.45×10-5 cm/s for propranolol and 4.22×10-7 cm/s for atenolol.Conclusion: PA is transported through the Caco-2 cell monolayer in a concentration-dependent manner and the transport was linear with time. The absorption in apical to basolateral direction and secretion in basolateral to apical direction were poor and their Papp values were comparable to atenolol.
