Inhibition of Glioma Cell Lysosome Exocytosis Inhibits Glioma Invasion

Abstract: Cancer cells invade by secreting enzymes that degrade the extracellular matrix and these are sequestered in lysosomal vesicles. In this study, the effects of the selective lysosome lysing drug GPN and the lysosome exocytosis inhibitor vacuolin-1 on lysosome exocytosis were studied to determine their effect on glioma cell migration and invasion. Both GPN and vacuolin-1 evidently inhibited migration and invasion in transwell experiments and scratch experiments. There are more lysosomes located on the cell membrane of glioma cells than of astrocytes. GPN decreased the lysosome number on the cell membrane. We found that rab27A was expressed in glioma cells, and colocalized with cathepsin D in lysosome. RNAi-Rab27A inhibited lysosome cathepsin D exocytosis and glioma cell invasion in an in vitro assay. Inhibition of cathepsin D inhibited glioma cell migration. The data suggest that the inhibition of lysosome exocytosis from glioma cells plays an important modulatory role in their migration and invasion.
Network Motifs in Object-Oriented Software Systems

Abstract: Nowadays, software has become a complex piece of work that may be beyond our control. Understanding how software evolves over time plays an important role in controlling software development processes. Recently, a few researchers found the quantitative evidence of structural duplication in software systems or web applications, which is similar to the evolutionary trend found in biological systems. To investigate the principles or rules of software evolution, we introduce the relevant theories and methods of complex networks into structural evolution and change of software systems. According to the results of our experiment on network motifs, we find that the stability of a motif shows positive correlation with its abundance and a motif with high Z score tends to have stable structure. These findings imply that the evolution of software systems is based on functional cloning as well as structural duplication and tends to be structurally stable. So, the work presented in this paper will be useful for the analysis of structural changes of software systems in reverse engineering.
Trace elemental records of short-lived heating during exhumation of the CCSD eclogites

Abstract: Trace element compositions of garnet, omphacite and apatite in ultrahigh-pressure eclogites from the main hole of the Chinese Continental Scientific Drilling (CCSD) project were in situ analyzed by the LA-ICP-MS method. Although both garnet and omphacite have homogeneous major element compositions, their trace elements show zonations from core to rim in rare earth elements. In particular, middle rare earth elements in the garnet, heavy rare earth elements in the apatite and all rare earth elements in the omphacite increase from core to rim, respectively. Based on dependence of partition coefficients on temperature and pressure in these minerals, we suggest that the trace elemental zonations in these minerals may record a short-lived heating event during exhumation.

Abstract: The development of Web service has changed the process of software production, and requirements engineering becomes the key issue of service-oriented software engineering. Meantime, it reduces the degree of difficulty of software production, which facilitates end-users to customize software according to their personalized requirements. The paper proposes a method for service-oriented personalized requirements analysis, which is based on domain goal model and process model. The method can inform users of potential errors in requirements by detecting the correctness of requirements, which is driven by users’ personalized operations on goal models, and customize personalized processes to satisfy users’ requirements by reusing domain processes. The personalized processes are the basis for Web service discovery and composition.
Bragg gap solitons in $\mathcal{PT}$ symmetric lattices with competing nonlinearity

Abstract: The effect of competing nonlinearity on beam dynamics in parity-time $(\mathcal{PT})$ symmetric potentials is investigated. By using numerical methods, the existence of gap solitons is demonstrated in the first Bragg gap of optical $\mathcal{PT}$ symmetric lattices with competing nonlinearity. Meanwhile, the stability of such solitons is analyzed through introducing a small perturbation to the solitary solutions. The abrupt annihilation of the solitons during propagation demonstrates the Bragg gap solitons in $\mathcal{PT}$ symmetric potentials are not stable. In comparison with the on-site gap solitons, the off-site gap solitons exhibit more robust properties during propagation.
Reappraisement and refinement of zircon U-Pb isotope and trace element analyses by LA-ICP-MS

Abstract: A protocol was established for simultaneous measurements of zircon U-Pb ages and trace elements by LA-ICP-MS at spot sizes of 16–32 μm. This was accomplished by introducing N2 into ICP to increase the sensitivity. The obtained U-Pb ages for zircon standards GJ-1, TEMORA and SK10-2 are consistent with the preferred values within about 1% uncertainty (2 σ) by simple external calibration against zircon standard 91500. Different data reduction softwares could yield different uncertainties for calculation of U-Pb ages. The commercially available program GLITTER4.4 could apply an improper uncertainty calculation strategy, but it may yield artificial high precisions for single analyses. Our trace element analyses indicate that Si is not an ideal internal standard for zircon when calibrated against the NIST glasses. Calibration against the NIST glasses using Si as an internal standard, a systematic deviation of 10%–30% was found for most trace elements including Zr. However, the trace element compositions of zircon can be accurately measured by calibration against multiple reference materials with natural compositions (e.g., BCR-2G, BHVO-2G and BIR-1G), or calibration against NIST SRM 610 and using Zr as an internal standard. Analyses of two pieces of GJ-1 demonstrate that it is relatively homogenous for most trace elements (except for Ti).

Abstract: 辽宁四道的沟金矿定位于鸭绿江断裂西侧。其容矿围岩为辽河群盖县组的变质岩系,成矿流体中富含Na^+、Ca^2+、Cl^离子和还原性所体(CO、N2、CH4、H2)。成矿温度为160~280℃,成矿压力为45~55MPa。Eh,pH,fo2和氢氧同位素值表明,成矿为相对弱还原环境,成矿流体为一种近中性的混合流体。

Abstract: Naturally, like the web, integrated software systems in Internet will have to be distributed and heterogeneous. To im-prove the interoperability of services for SAAS, it is crucial to build requirements semantics that will cross the entire lifecycle of services especially on requirements stage. In this paper, a requirements semantics interoperability extend-ing approach called Connecting Ontologies (CO) that will act as semantics information carrier designing to facilitate the requirements identification and services composition is proposed. Semantic measurement of Chinese scenario is explored. By adopting the approach, a series of tools support for transport domain are developed and applied based on CO and DPO (Domain Problem Ontology) to enforce requirements engineering of networked software efficiently.

Abstract: In situ trace element compositions of single minerals in eclogites from the CCSD main hole(100 - 1100m) were analyzed using LA-ICP-MS. Trace element partitioning between garnet and omphacite suggests that Ti and Co are controlled by Mg( for example, Dco Grt/Omp =3.43dMg Grt/Omp -0. 34), and REE, Sr and Y are controlled by Ca. Concentrations of REE, Pb and Th in omphacite are significantly controlled by presence of apatite. Petrologic and geochemical features suggest that trace element compositions of the minerals formed by retrograde metamorphism were controlled by original minerals, the mineral assemblage and fluid activity. For example, presence of epidote decreased greatly the LREE and Sr concentrations of the coexisting amphibole compared with the omphacite, parent mineral of the amphibole based on the petrologic features; and decomposition of phengite increased Rb and Ba concentrations of amphibole. MREE enrichments in the rim of garnets in several samples imply that the eclogites could suffer a shortlived heating during exhumation based on variations of partition coefficients with temperature and pressure in eclogite, as suggested by overprinting of granulite facies on eclogites and widespread partial melting of UHP rocks in the Sulu UHP terrane. On the other hand, the highly variations of Nb and Ta in rutile could provide additional evidence for the hypothesis that rutile in the eclogite was transformed from Ti-bearing magnetite during UHP metamorphism.
