New Standpoint of ALT Test for Blood Safety in Dongguan Blood Center  [PDF]

Abstract: Objective: To investigate the relationship between the single ALT positive (ELSIA-HBV/HCV negative) and NAT-HBV/HCV on blood donor samples. Methods: 28710 samples were surveyed retrospectively from Dec. 2010 to Dec. 2011. ALT was detected by rate method, and the single ALT positive samples were detected by ELSIA-HBV/HCV and NAT-HBV/HCV. The relationship between the single ALT positive and NAT-HBV/HCV were analyzed. 21 samples, values in 40 U/L ≤ ALT ≤ 70 U/L, were selected at random from 2516 samples with single ALT positive, which were second detected by ELISA and NAT in the second donation. Results: 2516 (8.74%) single ALT positive samples (ELSIA-HBV/HCV negative) were found in 28710 donors. Among these samples, 8 (2.8/10000) positive were detected by NAT, including 5 HBV-DNA-positive and 3 HCV-RNA-positive. Obviously, positive rate of NAT from the donors whose ALT value ≤ 70 U/L were lower than those of ≥ 71 U/L (P < 0.01). 21 donors were investigated in the second donation in following 153 to 401 days, All samples were negative by ELISA-HBV/HCV and NAT-HBV/HCV. Conclusions: Donors with single ALT positive (value in 40 U/L-70 U/L) are not likely to become HBV/HCV virus carriers or HBV/HCV patients after half or one year. So it is to set ALT abandone threshlod to ≤ 70 U/L can ensure blood safety, and reduce blood abandone in our center.
Separation of Sensitized and Non-Sensitized RBCs: Sephadex-Based Cell-Affinity Adsorbents

Abstract: Introduction In transfusion medicine, antibodies that cause RBCs positive DATs, may interfere with patients' phenotyping. Traditionally, these antibodies were removed using various antibody elution methodologies. However, the elution agents and conditions used have been only partially successful; and no one method is superior. The purpose of this study was to develop a general and efficient method to separate non-sensitized from sensitized RBCs using Sephadex-based cell-affinity adsorbents. Methods First, we coupled Sephadex support with Staphylococcal Protein G (SpG) with or without NHS. Then we simulated clinical conditions by mixing differe∏nt ratios of sensitized and non-sensitized RBCs in vitro. Sensitized cells were prepared by mixing antibody with corresponding antigen-positive RBCs. Finally, we checked the sensitization status of absorbed RBCs after absorption with modified Sephadex support. Results The number of sensitized RBCs bound to Sephadex-based cell-affinity adsorbents is approximately 5×108 RBCs/mL support. Activated Sephadex could separate sensitized from non-sensitized RBCs. Conclusion Sephadex-based cell-affinity adsorbents with an NHS spacer arm have bigger capacity for binding RBCs than unmodified Sephadex. The Sephadex-based cell-affinity adsorbents readily separate non-sensitized RBCs from sensitized RBCs, thus providing a new strategy to type the blood for transfused patients.
THE NUMERICAL CALCULATION OF SENSITIVITY COEFFICIENTS OF PERMEABILITY FIELD

Abstract: A formula of sensitivity coefficients in porous media is derived from fluid equations by Green's reciprocity principle. The discrete form is given by using numerical integration and finite difference method. The sensitivity coefficients of wellbore pressure to permeability parameters are given by Peaceman's equation and calculated by numerical method of 3D transient anisotropic fluid equation in porous media. The results are verfied by direct method. The computed results are in excellent agreement with those obtained by the direct method.
Zircon U-Pb geochronology of basement metamorphic rocks in the Songliao Basin

Abstract: Zircon LA-ICP MS U-Pb dating of six metamorphic rocks and a metagranite (breccia) from southern basement of the Songliao Basin are reported in order to constrain the formation ages of basement. The basement metamorphic rocks in the Songliao Basin mainly consist of metagabbro (L45-1), amphibolite (SN117), metarhyolitical tuff (G190), sericite (Ser) schist (N103), chlorite (Chl) schist (T5-1), biotite (Bi)-actinolite (Act)-quartz (Q) schist (Y205), and metagranite (L44-1). The cathodoluminesence (CL) images of the zircons from metagabbro (L45-1) and metagranite (L44-1) indicate that they have cores of magmatic origin and rims of metamorphic overgrowths. Their U-Pb isotopic ages are 1808±21 Ma and 1873±13 Ma, respectively. The zircons with oscillatory zoning from amphibolite (SN117) and Chl schist (T5-1), being similar to those of mafic igneous rocks, yield ages of 274 ± 3.4 Ma and 264 ± 3.2 Ma, respectively. The zircons from metarhyolitical tuff (G190) and Ser schist (N103) display typical magmatic growth zoning and yield ages of 424 ± 4.5 Ma and 287 ± 5.1Ma, respectively. Most of zircons from Bi-Act-Q schist (Y205) are round in shape and different in absorption degree in the CL images, implying their sedimentary detritals. U-Pb dating yield concordant ages of 427 ± 3.1Ma, 455 ± 12 Ma, 696 ± 13 Ma, 1384±62 Ma, 1649±36 Ma, 1778±18 Ma, 2450±9 Ma, 2579±10 Ma, 2793±4 Ma and 2953±14 Ma. The above-mentioned results indicate that the Precambrian crystalline basement (1808–1873 Ma) exists in the southern Songliao Basin and could be related to tectonic thrust, and that the Early Paleozoic (424–490 Ma) and Late Paleozoic magmatisms (264–292 Ma) also occur in the basin basement, which are consistent with the ages of the detrital zircons from Bi-Act-Q schist in the basement. Abstract:

UM2000 signal is a type of multi-audio frequency-modulated signal which is widely used for railway blocking. Principles of three typical demodulating algorithms are presented in details in this paper. Bit error rates of the three methods at different SNRs are achieved by Monte Carlo simulation experiments. Among the three algorithms, the quadrature demodulation has the best performance at the real working environment. However, the three methods have the same problem of phase hopping when noise is too strong.

Rice Protein Extracted by Different Methods Affects Cholesterol Metabolism in Rats Due to Its Lower Digestibility

Abstract: To elucidate whether the digestibility is responsible for the hypocholesterolemic action of rice protein, the effects of rice proteins extracted by alkali (RP-A) and α-amylase (RP-E) on cholesterol metabolism were investigated in 7-week-old male Wistar rats fed cholesterol-free diets for 3 weeks. The in vitro and in vivo digestibility was significantly reduced by RP-A and RP-E as compared to casein (CAS). The digestibility was lower in RP-E than that of RP-A. Compared with CAS, the significant cholesterol-lowering effects were observed in rats fed by RP-A and RP-E. Fecal excretion of bile acids was significantly stimulated by RP-E, but not by RP-A. The apparent cholesterol absorption was more effectively inhibited by RP-E than RP-A because more fecal neutral sterols were excreted in rats fed RP-E. There was a significant correlation between protein digestibility and cholesterol absorption ( r = 0.8662, P < 0.01), resulting in a significant correlation between protein digestibility and plasma cholesterol level ( r = 0.7357, P < 0.01) in this study. The present study demonstrates that the digestibility of rice protein affected by extraction method plays a major role in the modulation of cholesterol metabolism. Results suggest that the hypocholesterolemic action induced by rice protein with lower digestibility primarily contribute to the inhibition of cholesterol absorption.
Transgenic tobacco plants expressing synthetic Cry1Ac and Cry1Ie genes are more toxic to cotton bollworm than those containing one gene

Abstract: Transgenic tobacco plants carrying Cry1Ac, Cry1Ie or both genes were obtained. In the leaves of transgenic plants carrying both genes, the contents of Cry1Ac and Cry1Ie proteins were 0.173% and 0.131% of the total proteins, respectively. Cry1Ac protein content was 0.182% and Cry1Ie protein content was 0.124% of the total proteins in the leaves of transgenic plants containing only one Bt gene. Fresh leaves of transgenic tobacco and wild-type plants were used for the insect bioassay against wild-type and Cry1Ac-resistant cotton bollworm (Helicoverpa armigera). The bioassay results showed that transgenic plants carrying both genes were significantly more toxic to wild-type and Cry1Ac-resistant cotton bollworm than those carrying Cry1Ac or Cry1Ie alone. This study indicates that the higher toxicity of transgenic tobacco plants carrying both genes is caused by the cooperative function of both Bt proteins, thus providing a potential way to delay the development of insect resistance to transgenic crops.

Abstract: In this paper the research status of lateral shift of reflected electromagnetic wave at the interface of conductive media was reviewed, the lateral shift and the lateral delay were analyzed, and the characteristic of lateral shift curves varying with the angle of incidence was introduced in detail. For electromagnetic wave logging, the error correction method of lateral shift was stressed in this paper, the magnitude of measured errors caused by the lateral shift was established. It is noticed that the longer wavelength, the larger error of measured results. With respect to the effect of lateral shift, the problem which still needs to be settled in the measurement of electromagnetic wave, as well as prospects on the further research and applications of lateral shift are described.

Structure and Characteristics of Oxide Films Containing Ca and P on Ti Substrate

Abstract: The ceramic coating was formed by using microarc oxidation (MAO) in the electrolytic solution ofβ-Glycerophosphate sodium salt pentahydrate (β-GP) and calcium acetate (CA). The microstructure, phase structure and phase composition were investigated with scanning electron microscopy (SEM), atomic force microscope (AFM), energy dispersive X-ray microanalyser (EDX), X-ray photoelectron spectroscopy (XPS), electron probe micro analyzer (EPMA) and X-ray diffraction (XRD). Investigation of the chemical composition of the coating with EDX revealed that Ca and P of high concentration existed in oxide films. XRD patterns indicated that the oxide film was composed of only anatase at low voltage discharge, and that rutile peaks appear in addition to anatase peak with increasing MAO voltage. Despite the existence of Ca and P, compounds concerning Ca and P were not detected in the oxide film.
