Screening and phylogenetic analysis of antibacterial strains isolated from antarctic

Abstract: Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Pseudomonas solanacearum, Pseudomonas aeruginosa and Bacillus thuringiensis were used as indicator strains respectively, four antibacterial strains, named as 97, Z11, Z18 and Z19 were screened from 580 strains of bacteria isolated from Antarctic by the method of agar diffusion. The growth curves, antibacterial curves and phylogenetic analysis of 4 antimicrobial strains were studied. The results showed that all of the four strains reached the log phase when they were cultured 24 h, strain 97 was in the log phase when it was cultured after 48 h, and the remaining three strains Z11, Z18 and Z19, were in the stationary when they were cultured after 60 h. They had the highest antibacterial activity when it was cultured after 84, 96, 72 and 72 h, respectively. Phylogenetic analysis indicated that these four strains belong to genera of Rheinheimera, Psychrobacter, Pseudomonas and Psychrobacter, respectively.
Genome-Wide Association Study Identified CNP12587 Region Underlying Height Variation in Chinese Females

Abstract: Introduction Human height is a highly heritable trait considered as an important factor for health. There has been limited success in identifying the genetic factors underlying height variation. We aim to identify sequence variants associated with adult height by a genome-wide association study of copy number variants (CNVs) in Chinese. Methods Genome-wide CNV association analyses were conducted in 1,625 unrelated Chinese adults and sex specific subgroup for height variation, respectively. Height was measured with a stadiometer. Affymetrix SNP6.0 genotyping platform was used to identify copy number polymorphisms (CNPs). We constructed a genomic map containing 1,009 CNPs in Chinese individuals and performed a genome-wide association study of CNPs with height. Results We detected 10 significant association signals for height (p<0.05) in the whole population, 9 and 11 association signals for Chinese female and male population, respectively. A copy number polymorphism (CNP12587, chr18:54081842-54086942, p = 2.41×10?4) was found to be significantly associated with height variation in Chinese females even after strict Bonferroni correction (p = 0.048). Confirmatory real time PCR experiments lent further support for CNV validation. Compared to female subjects with two copies of the CNP, carriers of three copies had an average of 8.1% decrease in height. An important candidate gene, ubiquitin-protein ligase NEDD4-like (NEDD4L), was detected at this region, which plays important roles in bone metabolism by binding to bone formation regulators. Conclusions Our findings suggest the important genetic variants underlying height variation in Chinese.
The effects of Suaeda salsa L. planting on the soil microflora in coastal saline soil

Abstract: Bioremediation,a rising environmental biotechnology,increasingly plays an important role in controlling the pollution in soil,surface water and ground water.It has many merits such as low cost,good cleanup effect and no secondary pollution compared with the traditional physical and chemical remediation techniques.Cultivating with saline-enduring and saline-avoiding plants can effectively improve saline soil.The improvement effects ought to be studied not only on the physical and chemical properties but also on the biological characters.Bioremediation of coastal saline soil was carried out by planting halophyte Suaeda salsa L in Tianjin estuarine area.The effects on the soil microflora were studied by comparing the content of soluble salt,the microbial amount and the transition of dominant microflora in soil.The results showed that the content of soluble salt in S.salsa planted soil decreased by 41%(method of weight) and 37%(method of electric conductivity) compared with that of control respectively.It revealed that the halophyte S.salsa was salt-absorbing plant for saline soil.The results also showed that the microbial amounts were increased with the decrease of the soluble salt in the soil around S.salsa L.root system.The bacteria,actinomycete and fugi were increased 2.3 times,4.3 times and 71 times respectively.The salinity for optimal growth of bacteria decreased synchronously with the decrease of salinity in the soil of root system.The bacteria with the lower ability of salt-tolerance have been becoming the dominant flora.The phylogenetic analysis based on the 16S rDNA indicated that the planting of S.salsa L also had effects on the sorts of bacteria in soil.Bacillus sp.became the dominant flora in saline soil after bioremediation by S.salsa.All these results demonstrated the remarkable improvement effect of ecosystem after S.salsa planting on coastal saline soil.

Abstract: 以南极菌 Pseudoalteromonas sp.S-15-13 为材料, 采用实时定量 PCR 的方法研究了温度、冻融循环及培养基中 NaCl、葡萄糖含量和 pH 对多糖合成关键酶基因 ugd 表达水平的影响。结果表明, 低温有利于ugd的表达, 在2℃和10℃培养温度下, 培养24 h后ugd的表达量约为20℃时的4倍; 冻融循环后, ugd 的表达量上调, 第 2 个冻融循环后 ugd 的表达量较对照提高了 6.85 倍; NaCl 对 ugd的影响表现为低促高抑, 即 NaCl 含量为 6.0%时 ugd 的表达量是对照(3.0%)的 3.97 倍, 但含量达 9.0%时ugd表达量仅为对照的2/5; 葡萄糖能够提高ugd的表达量, 当含量为2.0%时ugd表达量为对照的13.68 倍; 在一定范围内(pH 5.0—8.0), pH 的改变会促进ugd 的表达, 当 pH 为 6.0 时 ugd 表达量约为pH 7.0 时的 2.15 倍。
Spatio-temporal variation and associated affecting factors of soil pH in the past 20 years of Jiangsu Province, China

Abstract: The changes of soil pH significantly affect soil chemical properties and even whole environmental ecosystems. For this reason, the studies on them have attracted and con acidification is being accelerated with rapid industrial deve great deal of eco-environmental problems. Some authors tinue to attract much academic attention. Especially the soil lopment and unreasonable farming practices, which has led to a have conducted much research on the effects of some unique affecting factors on soil pH changes in some relatively uniform regions. However, in-situ studies of their comprehensive effects on soil pH changes in some complex regions have been seldom investigated. In this study, the spatio-temporal variation and associated factors which affect soil pH changes of Jiangsu province with high rapid industrial development and dense population were discussed by comparing the two spatial distribution maps of soil pH in 1980 and 2003. The results showed that the spatial distribution pattern of soil pH in 2003 was similar to that in 1980 with lower soil pHs in the south part of the province than those in the north because of the difference of soil types. However, significantly decreasing changes were also observed in some local regions from 1980 to 2003. Intensively acidified soils were identified in Yishu mound and depressed plain soil region, Lixiahe depressed plain soil region, Ningzhenyang low mountain and undulated soil region, and Taihu Lake plain soil region. In the former two regions the dominant affecting factors on decreased soil pH are farming practices such as fertilizing, incorporation of crop residues and soil utilization. As for the latter two, besides farming practices, industrialization and urbanization also are main affecting factors. Soil pH has increased in some areas of Ningzhenyang low mountain and undulated soil region and Taihu Lake soil region mainly due to soil utilization changes, structures, and road construction. Among soil groups, pH changes of Fluvo-aquic soils in the past 20 years were not significant, but those in paddy soils significantly decreased, particularly in the Lixiahe depressed plain soil region where decreasing trend of soil pH may be accelerated. This is because acidic buffering capacity of Fluvo-aquic soils is greater than that of paddy soils. In conclusion, farming practices, industrialization and urbanization are important affecting factors of soil pH changes and have led to significantly changes in some local regions. In order to the soil sustainable utilization in rapid economic developing regions, the government must mention the soil pH changes and take some effective measures to adjust and guide agriculture and industry so that the sparse soil resources in the densely populated area can be appropriately utilized.
Effects of wheat and soybean stubbles on soil microbial ecological characteristics in cucumber field.

Abstract: The study with traditional dilution plate method and BIOLOG ECO system indicated that under wheat and soybean stubbles, the amounts of soil fungi, bacteria and actinomyces in cucumber field increased, while that of Fusarium oxysporum f. sp. cucumerinum decreased (P <0.05). The Shannon-Wiener index, evenness index, Simpson index, and McIntosh index of soil microbial communities as well as soil microbial biomass C increased (P <0.05), basal respiration and metabolic quotient qCO2 decreased (P <0.05), and soil microbe's sole-carbon-source utilization enhanced. In addition, the contents of soil available P and K and the cucumber yields were also increased markedly. All of these illustrated that wheat and soybean stubbles improved the ecological environment of soil microbes.
Cardiac magnetic source imaging based on current multipole model

Abstract: It is widely accepted that the heart current source can be reduced into a current multipole. By adopting three linear inverse methods, the cardiac magnetic imaging is achieved in this article based on the current multipole model expanded to the first order terms. This magnetic imaging is realized in a reconstruction plane in the centre of human heart, where the current dipole array is employed to represent realistic cardiac current distribution. The current multipole as testing source generates magnetic fields in the measuring plane, serving as inputs of cardiac magnetic inverse problem. In the heart-torso model constructed by boundary element method, the current multipole magnetic field distribution is compared with that in the homogeneous infinite space, and also with the single current dipole magnetic field distribution. Then the minimum-norm least-squares (MNLS) method, the optimal weighted pseudoinverse method (OWPIM), and the optimal constrained linear inverse method (OCLIM) are selected as the algorithms for inverse computation based on current multipole model innovatively, and the imaging effects of these three inverse methods are compared. Besides, two reconstructing parameters, residual and mean residual, are also discussed, and their trends under MNLS, OWPIM and OCLIM each as a function of SNR are obtained and compared.
Inverse computation for cardiac sources using single current dipole and current multipole models

Abstract: Two cardiac functional models are constructed in this paper. One is a single current model and the other is a current multipole model. Parameters denoting the properties of these two models are calculated by a least-square fit to the measurements using a simulated annealing algorithm. The measured signals are detected at 36 observation nodes by a superconducting quantum interference device (SQUID). By studying the trends of position, orientation and magnitude of the single current dipole model and the current multipole model in the QRS complex during one time span and comparing the reconstructed magnetocardiography (MCG) of these two cardiac models, we find that the current multipole model is a more appropriate model to represent cardiac electrophysiological activity.
