Fungicide and insecticide activity of Justicia procumbens extracts

Abstract: Justicia procumbens is traditional Chinese medicine herb. To confirm fungicidal and insecticidal activity of J. procumbens extracts, methanol, chloroform, acetone, ethyl acetate and hexane extracts were obtained from J. procumbens by the combined method of cold-soaking and ultrasonic extraction. Extract inhibitory effects were studied using the inhibition zone method involving five species of plant pathogenic fungus-Colletotrichum gloeosprioides, Phomopsis asparagi, Pyricularia grisea, Fusarium graminearum and Botrytis cinerea. Methanol extract toxicity against four species of plant pathogenic fungus and bacteria (C. gloeosprioides, P. asparagi, B. cinerea and Xanthomonas campestris) and three species of insects (Aedes albopictus, Musca domestica and Pieris rapae) were also assayed. Results showed that 8 mg?mL-1 extracts of J. procumbens with different polar solvents had some inhibitory effects on five species of plant pathogenic fungus. The inhibition rates of methanol extract against C. gloeosprioides, P. asparagi and B. cinerea were over 50%. Methanol extract from J. procumbens had better fungicidal and insecticidal activity. EC50 of methanol extract against mycelium growth of C. gloeosprioides, P. asparagi and B. cinerea were 5.94 mg?mL-1, 4.61 mg?mL-1 and 5.27 mg?mL-1, respectively. The corresponding EC90 were 63.69 mg?mL-1, 58.01 mg?mL-1 and 54.57 mg?mL -1, respectively. Methanol extract at 0.25~1.00 mg?mL-1 showed strong inhibitory activity against X. campestris. The resultant extract at 0.125 mg?mL-1 showed moderate inhibitory activity. Methanol extract MIC was 0.062 5 mg?mL-1. Methanol extract LC50 against A. albopictus, M. domestica and P. rapae were 0.195 8 mg?mL-1, 0.351 4 mg?mL-1 and 0.287 7 mg?mL-1, respectively. The corresponding EC95 were 0.988 4 mg?mL-1, 3.053 2 mg?mL-1 and 2.584 4 mg?mL-1, respectively. This suggested that J. procumbens extracts had better application prospects as a biological pesticide in agricultural production.
Investment and coordination problems of high-strength eco-environment reconstruction in Yangou Basin of loess hilly region

Abstract: Eco-environment construction in the loess-hilly regions of Yangou Basin was used as a case to analyze investment structure and management of hi-speed,histrength eco-environment engineering in order to harness and optimize investment.Prediction and expectations of future construction investment in eco-environment are discussed.The results offer theoretic foundation for investment mechanism of eco-environment reconstruction in the middle-western regions,especially the loess hilly regions.
AVO characters of tectonic coal and its modeling research

Abstract: 瓦斯突出是当前影响我国煤矿安全生产的主要因素之一.由于不同性质的煤层其瓦斯突出的可能性不同,原生煤具有低瓦斯炙出可能性,构造煤瓦斯突出可能性较大,而具有软分层特征的构造煤其瓦斯突出的可能性极高.由于不同性质煤层间的岩性差异较大,构造煤的泊松比要比原生煤的泊松比大0.1~0.2.因此,不同煤层与其顶、底板岩性差异不同,从而其顶、底板反射波的AVO曲线存在较大差异.从而可以通过AVO分析技术探测瓦斯突出可能性较高的构造煤的分布,特别是具有软分层特征构的造煤的分布.通过这种方式可以为煤矿安全生产提供第一手的地质资料.但目前AVO技术是以石油工业为基础发展而来的,并不完全适用于煤系地层.通过对煤层顶、底板AVO曲线的对比分析,发现目前常用的AVO近似方程对煤系地层来说精度较低,严重影响AVO分析精度.本文通过对Zoeppritz方程及其主要近似方程的分析对比,提出了一种改进的Hilterman近似方程,从而极大地提高了煤系地层AVO曲线的近似精度,使高精度的煤系地层AVO分析成为可能.另外,通过对构造煤顶、底板AV0曲线的分析,发现构造煤底板反射波较复杂并不适合AVO分析,而其顶板反射波规律性较强有利于AVO分析.
Autophagy Enhances Bacterial Clearance during P. aeruginosa Lung Infection

Abstract: Pseudomonas aeruginosa is an opportunistic bacterial pathogen which is the leading cause of morbidity and mortality among cystic fibrosis patients. Although P. aeruginosa is primarily considered an extacellular pathogen, recent reports have demonstrated that throughout the course of infection the bacterium acquires the ability to enter and reside within host cells. Normally intracellular pathogens are cleared through a process called autophagy which sequesters and degrades portions of the cytosol, including invading bacteria. However the role of autophagy in host defense against P. aeruginosa in vivo remains unknown. Understanding the role of autophagy during P. aeruginosa infection is of particular importance as mutations leading to cystic fibrosis have recently been shown to cause a blockade in the autophagy pathway, which could increase susceptibility to infection. Here we demonstrate that P. aeruginosa induces autophagy in mast cells, which have been recognized as sentinels in the host defense against bacterial infection. We further demonstrate that inhibition of autophagy through pharmacological means or protein knockdown inhibits clearance of intracellular P. aeruginosa in vitro, while pharmacologic induction of autophagy significantly increased bacterial clearance. Finally we find that pharmacological manipulation of autophagy in vivo effectively regulates bacterial clearance of P. aeruginosa from the lung. Together our results demonstrate that autophagy is required for an effective immune response against P. aeruginosa infection in vivo, and suggest that pharmacological interventions targeting the autophagy pathway could have considerable therapeutic potential in the treatment of P. aeruginosa lung infection.
Syntaxin Binding Protein 1 Is Not Required for Allergic Inflammation via IgE-Mediated Mast Cell Activation

Abstract: Mast cells play a central role in both innate and acquired immunity. When activated by IgE-dependent FcεRI cross-linking, mast cells rapidly initiate a signaling cascade and undergo an extensive release of their granule contents, including inflammatory mediators. Some SNARE (soluble N-ethylmaleimide-sensitive fusion factor attachment protein receptor) proteins and SM (Sec1/Munc18) family proteins are involved in mast cell degranulation. However, the function of syntaxin binding protein 1 (STXBP1), a member of SM family, in mast cell degranulation is currently unknown. In this study, we examined the role of STXBP1 in IgE-dependent mast cell activation. Liver-derived mast cells (LMCs) from wild-type and STXBP1-deficient mice were cultured in vitro for the study of mast cell maturation, degranulation, cytokine and chemokine production, as well as MAPK, IκB-NFκB, and NFAT signaling pathways. In addition, in vivo models of passive cutaneous anaphylaxis and late-phase IgE-dependent inflammation were conducted in mast cell deficient Wsh mice that had been reconstituted with wild-type or STXBP1-deficient mast cells. Our findings indicate that STXBP1 is not required for any of these important functional mechanisms in mast cells both in vitro and in vivo. Our results demonstrate that STXBP1 is dispensable during IgE-mediated mast cell activation and in IgE-dependent allergic inflammatory reactions.
Iterative non-overlapping domain decomposition method for optimal boundary control problems governed by elliptic equations

Abstract: 摘要: 研究了一类椭圆方程约束的最优边界控制问题的数值求解方法。为了避免运用传统数值方法所产生庞大的计算量,我们采用非重叠型区域分解迭代方法。 即:将求解区域Ω分解成若干个非重叠子区域,把上述的最优边界控制问题分解成这些子区域上的局部问题,这些局部问题间的内边界条件采用Robin条件。建立了求解这些局部问题的迭代格式,推导证明了迭代格式的收敛性。最后,给出一个数值算例,验证了迭代格式的有效性。
Abstract: A numerical method for solving optimal boundary control problems governed by elliptic equations is considered. In order to avoid large amounts of calculation produced by traditional numerical methods. An iterative non-overlapping domain decomposition method is established. The whole domain is divided into many non-overlapping subdomains, and the optimal boundary control problem is decomposed into local problems in these subdomains. Robin conditions are used to communicate the local problems on the interfaces between subdomains. The iterative scheme for solving these local problems is studied, and prove the convergence of the scheme is proved. Finally, a numerical example to prove the validity of the scheme is presented

Abstract: Myosin V利用ATP水解所释放的自由能,朝肌动蛋白微丝正端作连续的定向运动,平均步长约为36nm。最近几年,诸多实验数据表明,myosin V步长并不固定为36nm,马达各步长值和相应步长出现概率的柱状图符合高斯分布;且在负载力大于2pN的情况下会出现“中间步长”和“后退步子”的现象。可根据已有实验数据,同时考虑马达在跃迁过程中所受的溶液摩擦阻力、常负载力和高斯随机力对其跃迁距离的影响,提出一种跃迁模型,并以此为基础对上述现象进行理论解释。
Effects of V/III ratio on species diffusion anisotropy in the MOCVD growth of non-polar a-plane GaN films

Abstract: Non-polar a-plane (11\bar 20) GaN films have been grown on r-plane (1\bar 102) sapphire substrates by metal organic chemical vapour deposition. The influences of V/III ratio on the species diffusion anisotropy of a-plane GaN films were investigated by scanning electron microscopy, cathodoluminescence and high-resolution x-ray diffraction measurements. The anisotropy of a-plane GaN films may result from the different migration length of adatoms along two in-plane directions. V/III ratio has an effect on the growth rates of different facets and crystal quality. The stripe feature morphology was obviously observed in the film with a high V/III ratio because of the slow growth rate along the 1\bar 100] direction. When the V/III ratio increased from 1000 to 6000, the in-plane crystal quality anisotropy was decreased due to the weakened predominance in migration length of gallium adatoms.

Abstract: Asymmetric and periodic potential was used to describe the interaction between motor proteins and filaments that were periodic and polar. The current and effective potential in the two-state model were calculated. It was shown that the directed motion of motor protein was relevant to the effective potential. The slope of the effective potential corresponded to an average force. Therefore existence of the average force incarnated that detailed balance was broken in two-state transition. The curve of force-velocity theoretical relationship was also compared with the experimental data, it was qualitative tallied with the experiment.
Screening and phylogenetic analysis of antibacterial strains isolated from antarctic

Abstract: Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Pseudomonas solanacearum, Pseudomonas aeruginosa and Bacillus thuringiensis were used as indicator strains respectively, four antibacterial strains, named as 97, Z11, Z18 and Z19 were screened from 580 strains of bacteria isolated from Antarctic by the method of agar diffusion. The growth curves, antibacterial curves and phylogenetic analysis of 4 antimicrobial strains were studied. The results showed that all of the four strains reached the log phase when they were cultured 24 h, strain 97 was in the log phase when it was cultured after 48 h, and the remaining three strains Z11, Z18 and Z19, were in the stationary when they were cultured after 60 h. They had the highest antibacterial activity when it was cultured after 84, 96, 72 and 72 h, respectively. Phylogenetic analysis indicated that these four strains belong to genera of Rheinheimera, Psychrobacter, Pseudomonas and Psychrobacter, respectively.
