Protocol for Callus Induction and Somatic Embryogenesis in Moso Bamboo

Abstract: Moso bamboo [Phyllostachys heterocycla var. pubescens (Mazel ex J. Houz.) Ohwi] is one of the most important forest crops in China and the rest of Asia. Although many sympodial bamboo tissue culture protocols have been established, there is no protocol available for plantlet regeneration as indicated by callus induction for monopodial bamboos, such as Moso bamboo. In the present report, embryogenic callus induction, embryoid development, and germination were established for Moso bamboo from zygotic seed embryos. Callus was initiated from zygotic embryos after 10–20 d culture on MS media supplemented with 4.0 mg/L 2, 4-D and 0.1 mg/L zeatin (ZT). About 50% of the explants produced calli, and nearly 15% of the calli were found to be embryogenic in nature. These embryogenic calli can be subcultured for proliferation in the Murashige and Skoog media (MS) supplemented with 0.5–2.0 mg/L 2, 4-D. These calli were found to have maintained their capacity for regeneration even after one year of subculture. The viable somatic embryoids regenerated in medium containing 5.0–7.0 mg/L ZT. Nearly 5% of the calli were found capable of regenerating into plantlets directly in MS medium containing 5.0–7.0 mg/L ZT. Root growth was more pronounced when the plantlets were transferred to medium containing 2.0 mg/L NAA. After 30 days of subculture, the plantlets were transferred to a greenhouse.
A comparative study on genetic polymorphism and genetic relationship of 13 SNPs in three Chinese populations

Abstract: 采用荧光标记复合扩增毛细管电泳技术,基于等位基因特异性PCR原理,通过正交实验法建立了荧光标记复合扩增片段长度差异等位基因特异性SNPs分型体系,该体系可以根据产物长度和产物峰的数量一次完成13个SNPs分型,纯合子为单一产物峰,杂合子为长度相差4bp的两个产物峰.采用该体系对我国辽宁地区汉族、内蒙古地区蒙古族和广西地区壮族3个民族13个SNPs位点多态性进行群体调查,获得了3个民族13个SNPs等位基因分布频率,比较了3个民族等位基因的差异,并对其遗传学关系进行了研究.结果显示:3个民族13个SNPs的等位基因分布具有多态性,多个SNPs等位基因分布具有显著性差异(P<0.01),抽样调查结果符合Hardy-Weinberg平衡;辽南地区汉族人群与内蒙古蒙古族人群的亲缘关系更为接近,与广西壮族之间的亲缘关系相对较远.
Effects of microwave radiation on peripheral lymphocyte subpopulations in rats

Abstract: Objective To investigate the effects and mechanisms of microwave radiation on peripheral lymphocyte subpopulations in Wistar rats.Methods A total of 100 Wistar rats(180-220g) were exposed to microwave with different average power densities of 5,10,30 and 60 mW/cm2,and sham exposure of 0mW/cm2 was performed in a control group at the same time.At day 1,7,14 and 28 after microwave irradiation,the changes in peripheral CD3+,CD4+,CD8+ T cells,ratio of CD4+/CD8+ and CD45RA+ B lymphocyte in rats were analyzed by flow cytometry(FCM).Results The CD3+ T cells decreased significantly in 10-30mW/cm2 groups at day 7 and in 5-30 mW/cm2 groups at day 14 after radiation as compared with control group(P < 0.05),and CD4+ T cells decreased significantly in 10mW/cm2 group at day 14 after radiation as compared with control group(P < 0.01).From day 1 to day 14 after radiation,CD8+ T cells showed a reduction in number in all irradiated groups when compared with the control,but statistical significance was only found in the 30mW/cm2 group(P < 0.05).The CD4+/CD8+ ratio increased in 5mW/cm2 group on day 1,while decreased significantly in 5-30mW/cm2 groups on day 14 after radiation as compared with control group(P < 0.05).After microwave exposure,however,CD45RA+ B cells in 30mW/cm2 group at day 1 and in 30-60mW/cm2 groups at day 14 after radiation increased significantly in a dose-dependent manner.Conclusion A definite dosage of microwave radiation,ranging from 5-60mW/cm2,may induce changes in subpopulations of peripheral lymphocytes and cause acute immune function impairment in rats.
Research progress in neuro-immune interactions in Caenorhabditis elegans

Abstract: The innate immune response may be activated quickly once the organism is invaded by exotic pathogens. An excessive immune response may result in inflammation and tissue damage, whereas an insufficient immune response may result in infection. Nervous system may regulate the intensity of innate immune responses by releasing neurotransmitters, neuropeptides and hormones. Compared with the complicated neuro-immune system in mammals, it is much simpler in Caenorhabditis elegans. Besides, C. elegans is accessible to genetic, molecular biology and behavioral analyses, so it has been used in studies on neuro-immune interactions. It has been revealed recently in the studies with C. elegans that the neuronal pathways regulating innate immune responses primarily include a transforming growth factor-β (TGF-β) pathway, an insulin/insulin-like growth factor receptor (IGF) pathway and dopaminergic neurotransmission. Since these pathways are evolutionally conservative, so it might be able to provide some new ideas for the research on neuro-immune interactions at molecular levels. The recent progress in this field has been reviewed in present paper.
Characteristics and significance of change in Treg/Th17 balance in rats with combined trauma and γ-radiation

Abstract: Objective  To observe the process of wound healing and investigate the characteristics of Treg/Th17 balance in rats with combined wound and 5 Gy whole-body γ-radiation (CWR) for exploring the interrelationship between wound healing and the immune balance of Treg/Th17. Methods  Sixty-five female Wistar rats (clean grade) were randomly divided, depending on weight, into normal control group (normal), wound only group (MW) and wound plus 5 Gy whole body γ-radiation group (RW). The wound area was measured with image analyzer, the peripheral white blood cells (WBC) and lymphocytes (LYM) were counted with automatic cytometry, and the percentage of splenic regulatory T cells (Tregs) and Th17 cells was analyzed by flow cytometry at different time points post CWR. Results  (1) The residual wound area was much greater in RW group than in MW group (P<0.01) 7d to 21d after CTR, the wound surface in MW group almost healed on 21d, while that in RW group healed on 28d post CTR. (2) The peripheral WBC and LYM counts were significantly lower in MW group than in normal group (P<0.05) 1d to 7d after CWR, and in RW group than in normal and MW group 1d to 14d after CWR. Furthermore, the WBC count was still obviously lower in RW group than in MW and normal groups 21d to 28 d after CWR. (3) The percentage of splenic Tregs was significantly higher in RW group than that in normal and MW groups (P<0.01) at 3d and 7d after CWR, and returned to normal level at 28 d after CWR. (4) The percentage of Th17 cells was notably higher in RW group than in normal and MW groups (P<0.01) on the first day, reached the peak value on 3 d, and returned to normal level on 28 d after CWR. (5) The ratio of Treg/Th17 was distinctly lower in RW group than in normal and MW groups from 1d to 21d, and approximated normal level on 28d after CWR, indicating that 5Gy whole-body γ-radiation could induce Treg/Th17 imbalance in wounded rats. Conclusions  5 Gy whole-body γ-radiation may delay the process of wound healing in rats. Radiation-induced changes in Tregs and Th17 may lead to the disequilibrium of Treg/Th17, which may be an important factor for delayed wound healing in CWR.
A bound system in the expanding universe with modified holographic Ricci dark energy and dark matter

Abstract: The evolution of a bound system in the expanding background has been investigated in this paper. The background is described by a FRW universe with the modified holographic dark energy model, whose equation of state parameter changes with time and can cross the phantom boundary. To study the evolution of the bound system, an interpolating metric is considered, and on this basis the geodesics of a test particle are given. The equation of motion and the effective potential are also derived from the geodesics. By studying the the effective potential and the evolution of the radius of a test particle in the bound system of the Milky Way galaxy, we have found that the galaxy would go through three stages: expands from a singular point; stays in a discoid for a period of time; big rip in the future. With the help of analysing the critical angular momentum, we find that the test particle needs less angular momentum to escape from the center mass as time passes.
The Database System in the Grid Environment

Abstract: Until now, almost all of the Grid applications use files to store data, so little has been done on the integration of database systems into the Grid. If the Grid is to support a wide range of applications ,we must solve the problem of integrating database systems into the Grid. This paper firstly examines the requirement of the Grid environment to database systems, and then summarizes two methods to integrate database systems into the Grid. Finally,we give a short evaluation on these two methods.
Building Reliable Content-Based Routing Protocol over Structured P2P Networks

Abstract: Much work has been done on building content-based publish/subscribe systems over structured P2P networks, so that the two technologies can be combined together to better support large-scale and highly dynamic systems. However, existing content-based routing protocols can only provide weak reliability guarantees over P2P networks. Based on the routing protocols of structured P2P networks, a new type of content-based routing protocol for pub/sub systems is designed, which is called Identifier Range Based Routing (IRBR) protocol. The IRBR protocol guarantees that the subscribing nodes always receive the interested events exactly once as long as the message from publishing nodes to subscribing nodes is arrivable in the P2P network. At the same time, it can also disseminate an event to all interested subscribers with less network traffic. A prototype pub/sub system has been developed on Pastry, and the experimental results demonstrate the fault-tolerance and routing efficiency of the protocol.

Chromosome numbers are closely correlated with floral structure. 2n = 38 is always found in the species with dingy-colored flower, swollen stigma and stamens inserted at the base of perianth tube;the species with bright-colored flower,smaller stigma and stamens inserted at upper or middle part of perianth tube usually have the chromosome num- ber 2n=36. 3. The basic chromosome number x= 19 is considered as original one in the genus,while x= 18 is derived.
