Abstract: With the development of permafrost activities, it is clear that the discovery of viable microbes in permafrost raised their research status due to their peculiar physiological mechanism. Methane generation from bicarbonate and acetate showed an affected interest with microbes activities. The results of this work has great theory and economic meanings. In addition, it is also promising for the application prospects to the continual development of organisms research.

Abstract: To investigate the fluidized characteristic of powder particle fluidized bed(PPFB),the experiment of cold model fluidized bed was carried out.The results show that fluidization of PPFB could be maintained when gas velocity is in an appropriate range.Pyrolysis and catalytic pyrolysis of pine biomass was also investigated using the PPFB.It was found that there is a rapid release zone for the volatile during biomass pyrolysis,about 92% of the volatile matter of pine is released at 723 K.The total yield of inorganic gas(IOG),hydrocarbon gas(HCG) and hydrocarbon liquid(HCL) is only 3.1% at 773 K.The yields of IOG,HCG and HCL increase with the increase of pyrolysis temperature.The total yields of IOG,HCG and HCL reach 58.7% at 1 173 K,and the pyrolysis product is almost CO.CoMo-B catalyst greatly promotes the secondary reactions of the primary products of catalytic hydropyrolysis.At 863 K the yield of light aromatic hydrocarbons(benzene,toluene,xylene and naphthalene) is 6.3%,which is 2 times higher than that by using SiO2 at 1 173 K.

Abstract: 研究了金刚石光学特性与氮杂质及其含量的关系,从传统的金刚石氮含量标定方法出发,修正了金刚石氮含量的计算方法,并且用添加叠氮化钠的原料在六面顶压机上进行了高氮含量金刚石的制备研究.随着体系中叠氮化钠的添加,金刚石红外吸收谱在800—1400 cm-1范围的吸收强度相对于基线不断升高,这表明金刚石中存在的氮含量在随着叠氮化钠添加而升高,金刚石在单声子区域吸收强度大大增强.用含叠氮化钠的原料制备的金刚石呈现绿色、墨绿色甚至黑色,颜色的深浅依赖于叠氮化钠添加的多少.傅里叶红外光谱测试结果表明

Abstract: According to seismic attributes of time windows and instantaneous moments, an improved algorithm based on the window of the instantaneous intensity ratio is proposed. The basic theory is that instantaneous attributes are computed with complex trace analysis and first arrival with the intensity ratio method. The results show that the effect of rolling time windows is poor and larger errors exist in the process of picking some special points. When using the instantaneous intensity ratio method, the event of first arrival curve becomes more smooth, and abnormal points have greatly reduced, thus the accuracy of picking is greatly enhanced.

Abstract: The treatment of cutting the spring wheat roots in pot experiment shows that injuring the spring wheat roots moderately in early time can boost the photosynthetic rate and promote the accumulation of the photosynthetic production, improve the water use efficiency (WUE) of spring wheat leaves, and finally increase the grain yield. However, injuring the spring wheat roots in the time of stem elonging mostly restrains the growth of the spring wheat, and the yield is decreased.

Abstract: 采用生物素标记的(CA)15探针和磁珠富集法构建了大珠母贝(Pinctada maxima)微卫星DNA文库,随机挑选1200个菌落,经PCR筛选得到298个候选克隆,成功测序246个, 分析获得251个微卫星序列,其中完美型、非完美型和混合型分别占63%、32%和5%。除探针中使用的CA重复外, 还得到AT、GT、TC、AG、GC、TAA、CAA、AGG、GATA、GACA、GTGC、CGTC、GACG、CTGT等重复序列。设计引物90 对, 挑选其中30对合成并筛选出21 对能在大珠母贝基因组有效扩增的引物。种群PCR扩增后利用变性聚丙烯酰胺凝胶电泳法进行检测, 获得了10个多态位点, 共检测出59个等位基因, 片段长度范围为133~444 bp。各位点等位基因数2~8 个, 平均等位基因数5.9个; 有效等位基因数为1.342 3~6.000 0, 平均为4.124 0; 多态性信息含量(CPI)为0.222 5~0.811 8, 平均0.7179; 期望杂合度(He)为0.259 3~0.847 5, 平均0.717 9; 观测杂合度(Ho)为0.300 0~0.800 0, 平均0.533 3。这些微卫星多态性标记的获得, 为进一步开展大珠母贝遗传育种、保护生物学和种群遗传多样性等研究奠定了一定基础。

Abstract: 目的·探讨人毛囊隆突区是否存在上皮干细胞、黑素干细胞及自发荧光。方法·标本收集自整形外科健康志愿者后枕部和颞部全层头皮,分离完整毛囊。利用荧光显微镜观察毛囊自发荧光情况;采用间接免疫荧光法和免疫组织化学法检测毛囊角蛋白K15和K19的表达情况;采用RT-PCR检测毛球和隆突区上皮干细胞及黑素干细胞的标志物Dct、K15和K19,以及成熟细胞标志物Tyrase、Tyrp1、MC1R、MITF的mRNA水平。结果·人毛囊隆突区细胞可观察到自发荧光;免疫荧光和免疫组织化学检测结果显示隆突区外毛根鞘表达K15和K19;RT-PCR结果显示,隆突区Dct、K15的mRNA水平显著高于毛球,K19的mRNA水平与毛球相比无显著差异,而Tyrase、Tyrp1、MITF-M、MC1R的mRNA表达水平显著低于毛球。结论·人毛囊隆突区细胞可观察到自发荧光,且有上皮干细胞与黑素干细胞的存在。
: Objective·To investigate whether there are epithelial stem cells, melanocyte stem cells, and autofluorescence in the bulge region of hair follicles. Methods·Samples were collected from the posterior occipital and temporal whole layer scalp of healthy volunteers with complete separation of hair follicles. Autofluorescence in hair follicles was observed using fluorescence microscope. Expressions of hair follicle keratins K15 and K19 were measured using indirect immunofluorescence and immunohistochemistry. RT-PCR was used to measure the mRNA expression levels of epithelial stem cell and melanocyte stem cell markers Dct, K15, and K19 and mature cell markers Tyrase, Tyrp1,MITF-M, and MC1R in hair bulbs and bulge region. Results·Autofluorescence was observed in the bulge region of hair follicles. Immunofluorescence and immunohistochemistry showed K15 and K19 were expressed in bulge region. RT-PCR showed that the mRNA expressions of Dct and K15 in bulge region were significantly higher than those in hair bulbs. The mRNA expression of K19 in bulge region was not significantly different from that in hair bulbs, while the mRNA expressions of Tyrase, Tyrp1, MITF-M, and MC1R in bulge region were significantly lower than those in hair bulbs. Nesfatin-1 Stimulates Fatty-Acid Oxidation by Activating AMP-Activated Protein Kinase in STZ-Induced Type 2 Diabetic Mice

Abstract: Nesfatin-1 is an anorexigenic peptide involved in energy homeostasis. Recently, nesfatin-1 was reported to decrease blood glucose level and improve insulin sensitivity in high-fat diet-fed rats. However, little information is known about the influence of nesfatin-1 on lipid metabolism either in physiological or diabetic condition. This study undertook whether nesfatin-1 was involved in the pathophysiology in Streptozotocin-induced type 2 diabetic mice (T2DM), which was induced by a combination of high-calorie diet and two low-doses Streptozotocin. We observed that plasma nesfatin-1 was significantly increased while expression of nesfatin-1 neurons were decreased in hypothalamus in diabetes group compared to only high-calorie diet control group; intravenous injection of nesfatin-1 decreased 0–1h, 0–2h, 0–3h cumulative food intake in T2DM, but 0–24h total food intake had no difference between groups. Body weight and plasma FFA were normalized after nesfatin-1(10 μg/Kg) administration for 6 days. These results suggested that nesfatin-1 improved lipid disorder in T2DM. It was found that blood glucose and insulin resistance coefficient decreased with treatment of nesfatin-1 (both in 1 μg/Kg and 10 μg/Kg doses) in diabetes mice. For further understanding the role of nesfatin-1 on lipid metabolism, we detected p-AMPK and p-ACC of skeletal muscle in T2DM using western blotting. The expression of p-AMPK and p-ACC increased when nesfatin-1 was given with doses 1 μg/Kg but not in doses 10 μg/Kg. Taken together, nesfatin-1 participated in the development of T2DM and stimulated free fatty acid utilization via AMPK-ACC pathway in skeletal muscle in T2DM.

中国菰资源及其应用价值的研究

Abstract: 中国菰Z.Latifolia Griseb(Turcz)]是木本科菰属(Zinania Linnaeus)4种植物之一,是一种多年水生草本植物。我国从周代开始已有3000多年的应用记载。对中国南四湖、洪湖、洪泽湖、太湖、巢湖、白马湖、高邮湖、骆马湖、石臼湖、宝应湖的野生菰资源的调查显示,其菰草总面积大约488km2,估计每年菰草和菰米产量分别约是55×104t和1.8×104t。实验室和人群研究表明,中国菰米食用安全、风味独特,未见致突变作用和人体毒副作用;其蛋白质储量达13.2%,蛋白质功效比值达2.75;富含无机盐和B族维生素。菰草粉中蛋白质、脂肪、灰分、钙和磷含量分别是7.4%、2.3%、1.2%、0.49、和0.12%。菰植物的根、茎、叶、果都有多种用途和功能,具有很高的经济价值,亟待科学、合理、综合地开发利用。
