Growth regulation of simian and human AIDS-related non-Hodgkin's lymphoma cell lines by TGF-β1 and IL-6

Abstract: Growth stimulation or inhibition in response to cytokine treatment was quantified using trypan blue exclusion or colorimetric MTT assay. Intracellular flow cytometry was used to analyze the activation of signaling pathways and to examine the expression of anti-apoptotic proteins and distinguishing hallmarks of AIDS-NHL subclass. Apoptosis was quantified by flow cytometric analysis of cell populations with sub-G1 DNA content and by measuring activated caspase-3.Results confirmed the sensitivity of LCL8664, an immunoblastic SAIDS-NHL cell line, to TGF-beta1-mediated growth inhibition, and further demonstrated the partial rescue by simultaneous treatment with IL-6. IL-6 was shown to activate STAT3, even in the presence of TGF-beta1, and thereby to activate proliferative and anti-apoptotic pathways. By comparison, human AIDS-NHL cell lines differed in their responsiveness to TGF-beta1 and IL-6. Analysis of a recently derived AIDS-NHL cell line, UMCL01-101, indicated that it represents immunoblastic AIDS-DLCBL. Like LCL-8664, UMCL01-101 was sensitive to TGF-beta1-mediated inhibition, rescued partially by IL-6, and demonstrated rapid STAT3 activation following IL-6 treatment even in the presence of TGF-beta1.These studies indicate that the sensitivity of immunoblastic AIDS- or SAIDS-DLBCL to TGF-beta1-mediated growth inhibition may be overcome through the stimulation of proliferative and anti-apoptotic signals by IL-6, particularly through the rapid activation of STAT3.AIDS-related non-Hodgkin's lymphoma (AIDS-NHL) is the second most frequent cancer (following Kaposi's sarcoma) associated with AIDS, and is the cause of death in approximately 16% of HIV-infected individuals [1,2]. Abstract: Background Elevated stress can exacerbate cancer symptom severity, and after completion of primary cancer treatments, many individuals continue to have significant distress. Mindfulness-Based Cancer Recovery (MBCR) is an 8-week group psychosocial intervention consisting of training in mindfulness meditation and yoga designed to mitigate stress, pain, and chronic illness. Efficacy research shows face-to-face (F2F) MBCR programs have positive benefits for cancer patients; however barriers exist that impede participation in F2F groups. While online MBCR groups are available to the public, none have been evaluated. Primary objective: determine whether underserved patients are willing to participate in and complete an online MBCR program. Secondary objectives: determine whether online MBCR will mirror previous efficacy findings from F2F MBCR groups on patient-reported outcomes. Method/design The study includes cancer patients in Alberta, exhibiting moderate distress, who do not have access to F2F MBCR. Participants will be randomized to either online MBCR, or waiting for the next available group. An anticipated sample size of 64 participants will complete measures online pre and post treatment or waiting period. Feasibility will be tracked through monitoring numbers eligible and participating through each stage of the protocol. Discussion 47 have completed/completing the intervention. Data suggest it is possible to conduct a randomized waitlist controlled trial of online MBCR to reach underserved cancer survivors. Trial registration Clinical Trials.gov Identifier: NCT01476891
The broad emission-line region: the confluence of the outer accretion disc with the inner edge of the dusty torus

Abstract: (Abridged) We investigate the observational characteristics of BLR geometries in which the BLR clouds bridge the gap, both in distance and scale height, between the outer accretion disc and the hot dust, forming an effective surface of a "bowl". The gas dynamics are dominated by gravity, and we include the effects of transverse Doppler shift, gravitational redshift and scale-height dependent macro-turbulence. Our simple model reproduces many of the phenomena observed in broad emission-line variability studies, including (i) the absence of response in the core of the optical recombination lines on short timescales, (ii) the enhanced red-wing response on short timescales, (iii) differences between the measured delays for the HILs and LILs, and (iv) identifies turbulence as a means of producing Lorentzian profiles (esp. for LILs) in low inclination systems, and for suppressing significant continuum--emission-line delays between the line wings and line core (esp. in LILs). A key motivation of this work was to reveal the physical underpinnings of the reported measurements of SMBH masses and their uncertainties. We find that SMBH masses derived from measurements of the fwhm of the mean and rms profiles show the closest correspondence between the emission lines in a single object, even though the emission line fwhm is a more biased mass indicator with respect to inclination. The predicted large discrepancies in the SMBH mass estimates between emission lines at low inclination, as derived using the line dispersion, we suggest may be used as a means of identifying near face-on systems. Our general results do not depend on specific choices in the simplifying assumptions, but are in fact generic properties of BLR geometries with axial symmetry that span a substantial range in radially-increasing scale height supported by turbulence, which then merge into the inner dusty TOR.

Abstract: Twenty-five years ago the desktop computer started becoming ubiquitous in the scientific lab. Researchers were delighted with its ability to both control instrumentation and acquire data on a single system, but they were not completely satisfied. There were often gaps in knowledge that they thought might be gained if they just had more data and they could get the data faster. Computer technology has evolved in keeping with Moore's Law meeting those desires; however those improvement have of late become both a boon and bane for researchers. Computers are now capable of producing high speed data streams containing terabytes of information; capabilities that evolved faster than envisioned last century. Software to handle large scientific data sets has not kept up. How much information might be lost through accidental mismanagement or how many discoveries are missed through data overload are now vital questions. An important new task in most scientific disciplines involves developing methods to address those issues and to create software that can handle large data sets with an eye towards scalability. This software must create archived, indexed, and searchable data from heterogeneous instrumentation for the implementation of a strong data-driven materials development strategy. At the National Center for Photovoltaics in the National Renewable Energy Lab, we began development a few years ago on a Laboratory Information Management System (LIMS) designed to handle lab-wide scientific data acquisition, management, processing, and mining needs for physics and materials science data. and with a specific focus on future scalability for new equipment or research focuses. We will present the decisions, process, and problems we went through while building our LIMS for materials research, its current operational state, and our steps for future development.

Abstract: This article argues that the relationship between Margaret Thatcher and her governments of the 1980s on the one hand, and the BBC on the other, differed from those of an ‘arm’s length’ regulator which had existed since the creation of the BBC through successive Conservative and Labour governments until 1979, in which ministers would not intervene in BBC policy or programming. Ministers and party members instituted a concerted attempt to intimidate the BBC, most especially its news and current affairs programmes, whenever the latter did not report national politics and international events to the government’s liking. Proposed neo-liberal reforms to the system of funding the Corporation and negotiations for regular increases in the value of the licence fee were similarly opportunities to exert pressure on the BBC and to call into question its public service remit. Finally, royal prerogative and royal patronage were used to manipulate the political colour of the Board of Governors and thus, indirectly, the management of the BBC.

Patient’s Experience of the Outcomes of Engaging in a Broadly Applicable Health Promotion Intervention for Individuals Facing Chronic Illness  [PDF]

Abstract: The aim of this study was to explore the health-related outcomes of a new health promotion intervention designed to be broadly applicable among people diagnosed with chronic illness. Qualitative process analysis was applied and a purposeful sample of 52 patients, representing a variety of long-term conditions, was invited to participate in the pilot-implementation and evaluation of the program. Participants attended individually or in groups in seven sessions held over a six-month period. A lay-based conceptual framework that revealed the bodily knowledge of patients with chronic illness as a resource for health was used as the frame for the program content and interdisciplinary health professionals who had undergone 80 hours of training led the program. The pedagogical approaches were varied in order to engage patients in health promotion work. In-depth evaluation interviews took place soon after program completion and data were analyzed in-depth by means of content analysis in order to identify patterns of health-related themes and the program’s possible active ingredients. The intervention was implemented successfully across clinical sites and diagnostic categories. Participants reported improved ability to manage illness-related problems and greater awareness and utilization of personal resources for health after they had participated in the program. They were able to think more positively about their own capabilities, gained greater understanding of factors that made their health condition better or worse, and became more active. Participants identified empowerment and social participation as important outcomes of engaging in the new program.

Abstract: This paper examines the effects of the divorce law liberalization of the early 1970s on the increase in divorce rates during the same time period. A review of the evidence suggests that the law changes were not a major driver of the divorce rates; but the policy changes appear to have affected behavior even for those who did not divorce. The results here suggest that as they saw the laws changing, young women in the divorce reform states redirected some of their investments from marriage to their own human capital. The perceived increase in the probability of divorce motivated women to improve their options outside of marriage.

Abstract: NEM® brand eggshell membrane is a novel dietary supplement that has been clinically shown to alleviate arthritis joint pain and stiffness; however the mechanism of action is not well understood. Preliminary evidence from an in vitro study of NEM® indicated that the mechanism of action may be based on the reduction of pro-inflammatory cytokines. In vivo studies were therefore initiated to evaluate the effects of NEM® on pro-inflammatory and anti-inflammatory cytokines following oral administration in rats. NEM® was administered daily at doses of 6.13 mg/kg bw/day (Study 1), 10.0 mg/kg bw/day (Study 2), or at doses of 0 (control), 26.0 or 52.0 mg/kg bw/day (Study 3) by oral gavage for 7 consecutive days. Inflammation was induced in the Study 3 rats by intraperitoneal injection of lipopolysaccharide. Changes in plasma cytokine levels from baseline following 7 days of oral supplementation with NEM® at 6.13 mg/kg bw/ day (Study 1) were statistically significant at Day 8 for IL-2, TIMP-1 and VEGF, at Day 21 for IL-10, and at Day 35 for MCP-1, MCP-3 and TIMP-1, and at 10.0 mg/kg
