Significance of nuclear morphometry in cytological aspirates of breast masses

Abstract: Background : Breast carcinoma is the most common malignancy globally. Cytological evaluation in breast lesions is largely subjective. Gradual progression of cells from normal to invasive involves nuclear changes that need to be viewed objectively. Aims : This study aims to apply nuclear morphometry on cytological breast aspirates. It evaluates its utility in differentiating benign vs. malignant lesions and correlates it with cytologic grading in malignant cases. Setting and Design : Nuclear morphometric parameters of malignant and benign cases were compared. Parameters of malignant cases were correlated with cytologic grading. Materials and Methods : Cytology was used to categorize aspirates from breast lumps into malignant (53 cases) and benign (29 cases). One hundred cells per case in both groups were mapped on DEBEL Cytoscan and six geometrical and three textural parameters obtained were compared. In malignant cases, morphometry was correlated with Robinson′s cytologic grading, which was further correlated in tissue sections (45 cases) with modified Scarff-Bloom-Richardson histologic grading. Statistical Analysis : Students "t"-test was applied for comparison between benign and malignant cases. One-way ANOVA followed by Bonferroni′s post hoc comparison was applied to compare the three cytologic grades. Results were considered significant when P<0.05. Results : Nuclear morphometry successfully differentiated between benign and malignant aspirates and correlated significantly with cytologic grades. Morphometry was especially useful in the diagnosis of atypical ductal hyperplasia and ductal carcinoma in situ. Useful parameters were mean nuclear area, long axis, short axis and total run length. Cytohistologic correlation was 83.3%, 88.9% and 88.9% for cytological grades 1, 2 and 3 respectively. Conclusions : Nuclear morphometry was thus a useful objective tool in the evaluation of breast masses.

Abstract: Pyogenic granuloma (PG) is a kind of inflammatory hyperplasia in response to chronic irritation. Here, we report a case of 64 year old male patient with PG on midline of the dorsum of the posterior third of the tongue. Its differential diagnosis, the importance of biopsy findings in establishing definitive diagnosis and about the new approaches for its treatment is discussed.? Elucidation of the Microbial Community in Activated Sludge Using PCR-DGGE Analysis in Arid and Semi Arid Regions of Rajasthan

Abstract: Activated sludge is the most commonly used process to treat sewage and industrial waste waterby micro organisms. The activated sludge system depends on the activities of microbial communitiespresent in the sludge. However, exact knowledge of the microbial community structure in waste watertreatment plants is limited. In this study, the bacterial diversity of activated sludge was investigated inthe two waste water treatment plants by using denaturing gradient gel electrophoresis (DGGE) of PCRamplified16S ribosomal DNA fragments. Dominant bands from DGGE profiles were excised andsubjected to sequencing to identify the dominant genotypes. Sequence analysis gave insights into theidentities of the predominant bacterial populations present. The DNA sequencing results indicated themicrobial diversity, revealing that the dominant bacteria present in Bramhapuri waste water treatmentplant is Acinetobacter sp. whereas the dominant bacteria in Pratapnagar (Delawas) waste watertreatment plant is Alpha-proteobacteria. Futhermore, cluster analysis of the DGGE profiles indicatedsignificant diversity in the bacterial community by depicting two distinct clusters for each waste watertreatment plant. These data endorse the ability of PCR-DGGE method to identify and characterizebacterial community from activated sludge.
Surrogate Regret Bounds for Bipartite Ranking via Strongly Proper Losses

Abstract: The problem of bipartite ranking, where instances are labeled positive or negative and the goal is to learn a scoring function that minimizes the probability of mis-ranking a pair of positive and negative instances (or equivalently, that maximizes the area under the ROC curve), has been widely studied in recent years. A dominant theoretical and algorithmic framework for the problem has been to reduce bipartite ranking to pairwise classification; in particular, it is well known that the bipartite ranking regret can be formulated as a pairwise classification regret, which in turn can be upper bounded using usual regret bounds for classification problems. Recently, Kotlowski et al. (2011) showed regret bounds for bipartite ranking in terms of the regret associated with balanced versions of the standard (non-pairwise) logistic and exponential losses. In this paper, we show that such (non-pairwise) surrogate regret bounds for bipartite ranking can be obtained in terms of a broad class of proper (composite) losses that we term as strongly proper. Our proof technique is much simpler than that of Kotlowski et al. (2011), and relies on properties of proper (composite) losses as elucidated recently by Reid and Williamson (2010, 2011) and others. Our result yields explicit surrogate bounds (with no hidden balancing terms) in terms of a variety of strongly proper losses, including for example logistic, exponential, squared and squared hinge losses as special cases. We also obtain tighter surrogate bounds under certain low-noise conditions via a recent result of Clemencon and Robbiano (2011).
Distribution Behaviour of Dimethoate in Tea Leaf  [PDF]

Abstract: A study was undertaken to assess the distribution behaviour of Dimethoate in tea leaf. Tea bushes were subjected to Dimethoate spray at recommended dose and double the recommended doses. The extraction of pesticide was done using chloroform and the analysis was done using a Hewlett-Packard 5890 series II gas chromatograph with Nitrogen Phosphorus Detector (NPD). The penetration behaviour of Dimethoate was studied in dry and wet seasons. Variations in penetration were observed in dry and wet seasons which was attributed to climatic factors like temperature, humidity, rainfall, sunlight and physicochemical properties of the residue like water solubility, partition coefficient and formulation type. Residues observed in the cell wall and tissues confirm its good penetrating ability inspite of its hydrophilic nature. Higher penetration in wet season as indicated can be attributed to the route through the stomatal pores.

Abstract: The ubiquitous Aeromonas hydrophila is responsible for several pathological conditions in fish and human. Like most gram negative bacteria, its virulence relies on outer membrane lipopolysachharide (LPS). The Leloir pathway enzyme UDP-galactose 4-epimerase (GalE), plays an important role in the LPS biosynthesis, and therefore is a potential drug target. We have earlier carried out extensive biochemical and biophysical studies with histidine-tagged recombinant GalE. However, for effective drug design it is desirable to understand the structure-function relation of a protein in its native form without any additional sequences or tags. In the present study, we report the high level expression, purification and characterization of recombinant GalE (rGalE) of Aeromonas hydrophila in its native form in E coli. The rGalE expressed as a soluble protein was purified to near homogeneity. From 1 L of shake flask culture ~15 mg of purified rGalE was obtained. The purified protein was biologically active with Km and Kcat values of 0.7 mM and 28.8 s–1, respectively. The enzyme exhibited a temperature and pH optima of 37C and 7 - 9, respectively. Thus, the present study employed for soluble expression and purification of functionally active rGalE without any tag bypasses the need for cumbersome strategies associated with removal of tag from purified protein.
Effect of Foundation and Non-Homogeneity on the Vibrations of Polar Orthotropic Parabolically Tapered Circular Plates  [PDF]

Abstract: The effect of Pasternak foundation and non-homogenity on the axisymmetric vibrations of polar orthotropic parabolically varying tapered circular plates has been analyzed on the basis of classical plate theory. Ritz method has been used to find the numerical solution of the specified problem. The efficiency of the Ritz method depends on the choice of basis function based upon deflection of polar orthotropic plates. The effects of different plate parameters viz. elastic foundation, non-homogeneity, taper parameter and that of orthotropy on fundamental, second and third mode of vibration have been studied for clamped and simply-supported boundary conditions. Mode shapes for specified plates have been drawn for both the boundary conditions. Convergence and comparison studies have been carried out for specified plates.

Abstract: In the present communication, the hydrodynamic model is used to investigate the amplitude modulation as well as demodulation of an electromagnetic wave of high power helicon pump wave into another helicon wave in strain dependent dielectric material incorporating carrier heating (CH) effects. The consideration of CH in modulation and demodulation is prime importance for the adding of new dimension in analysis of amplification of acoustic helicon wave. By using the dispersion relation, threshold pump electric filed and growth rate of unstable mode from the modulation and demodulation of the high power helicon wave well above from the threshold value will be discussed in the present analysis. The numerical analysis is applied to a strain dependent dielectric material, BaTiO3 at room temperature and irradiated with high power helicon wave of frequency 1.78 × 1014 Hz. This material is very sensitive to the pump intensities, therefore during studies, Gaussian shape of the helicon pump wave is considered during the propagation in stain dependent dielectric material and opto-acoustic wave in the form of Gaussian profile (ω00) is induced longitudinally along the crystallographic plane of BaTiO3. Its variation is caused by the available magnetic field (ωc), interaction length (z) and pulsed duration of interaction (τ). From the analysis of numerical results, the incorporation of CH effect can effectively modify the magnitude of modulation or demodulation of the amplitude of high power helicon laser wave through diffusion process. Not only the amplitude modulation and demodulation of the wave, the diffusion of the CH effectively modifies the growth rate of unstable mode of frequency in BaTiO3. The propagation of the threshold electric field shows the sinusoidal or complete Gaussian profile, whereas this profile is found to be completely lost in growth of unstable mode. It has also been seen that the growth rate is observed to be of the order of 108 - 1010 s-1 but from diffusion of carrier heating, and that its order is enhanced from 1010 - 1012 s-1 with the variation of the magnetized frequency from 1 to 2.5 × 1014 Hz.

Abstract: This paper deals with the interaction of femtosecond laser with strain dependent high dielectric material. For this investigation, ferroelectric material like BaTiO3 has been chosen because of centrosymmetric structure. Due to irradiation of laser light, the micro-structure of BaTiO3 is found to change along the direction of heat propagation. SEM and AFM tools have been used to detect the morphology and roughness of the femotosecond laser treated BaTiO3. The change of morphology and surface behavior depends upon the laser fluence and intensity of laser light. The maximum change in morphology has been observed at a higher laser fluence.

Abstract: High error rate and amplitude/phase imbalances can degrade the performance of any OFDM based wireless networks. Also the proper recovery of the input at the receiver end becomes questionable.The effect of such impairments can be compensated by using high order mapping techniques. This paper analyzes the performance of M-QAM and M-PSK (where M=256) using OFDM over a noisy channel, which is AWGN particularly. The parameters are modelled and simulated using MATLAB. The mapping techniques are compared for the parameter Symbol Error Rate. The simulation is done for transreceiving the OFDM signal at lesser Symbol Error Rate.
