SHH1, a Homeodomain Protein Required for DNA Methylation, As Well As RDR2, RDM4, and Chromatin Remodeling Factors, Associate with RNA Polymerase IV

Abstract: DNA methylation is an evolutionarily conserved epigenetic modification that is critical for gene silencing and the maintenance of genome integrity. In Arabidopsis thaliana, the de novo DNA methyltransferase, DOMAINS REARRANGED METHYLTRANSFERASE 2 (DRM2), is targeted to specific genomic loci by 24 nt small interfering RNAs (siRNAs) through a pathway termed RNA–directed DNA methylation (RdDM). Biogenesis of the targeting siRNAs is thought to be initiated by the activity of the plant-specific RNA polymerase IV (Pol-IV). However, the mechanism through which Pol-IV is targeted to specific genomic loci and whether factors other than the core Pol-IV machinery are required for Pol-IV activity remain unknown. Through the affinity purification of NUCLEAR RNA POLYMERASE D1 (NRPD1), the largest subunit of the Pol-IV polymerase, we found that several previously identified RdDM components co-purify with Pol-IV, namely RNA–DEPENDENT RNA POLYMERASE 2 (RDR2), CLASSY1 (CLSY1), and RNA–DIRECTED DNA METHYLATION 4 (RDM4), suggesting that the upstream siRNA generating portion of the RdDM pathway may be more physically coupled than previously envisioned. A homeodomain protein, SAWADEE HOMEODOMAIN HOMOLOG 1 (SHH1), was also found to co-purify with NRPD1; and we demonstrate that SHH1 is required for de novo and maintenance DNA methylation, as well as for the accumulation of siRNAs at specific loci, confirming it is a bonafide component of the RdDM pathway.
SRA-Domain Proteins Required for DRM2-Mediated De Novo DNA Methylation

Abstract: De novo DNA methylation and the maintenance of DNA methylation in asymmetrical sequence contexts is catalyzed by homologous proteins in plants (DRM2) and animals (DNMT3a/b). In plants, targeting of DRM2 depends on small interfering RNAs (siRNAs), although the molecular details are still unclear. Here, we show that two SRA-domain proteins (SUVH9 and SUVH2) are also essential for DRM2-mediated de novo and maintenance DNA methylation in Arabidopsis thaliana. At some loci, SUVH9 and SUVH2 act redundantly, while at other loci only SUVH2 is required, and this locus specificity correlates with the differing DNA-binding affinity of the SRA domains within SUVH9 and SUVH2. Specifically, SUVH9 preferentially binds methylated asymmetric sites, while SUVH2 preferentially binds methylated CG sites. The suvh9 and suvh2 mutations do not eliminate siRNAs, suggesting a role for SUVH9 and SUVH2 late in the RNA-directed DNA methylation pathway. With these new results, it is clear that SRA-domain proteins are involved in each of the three pathways leading to DNA methylation in Arabidopsis.
Brain perfusion CT compared with15O-H2O-PET in healthy subjects

Abstract: RCBF was measured twice in 12 healthy subjects with15O-H2O PET and once with perfusion CT performed over the basal ganglia. Matching rCBF values in regions of interest were compared.Measured with perfusion CT, rCBF was significantly and systematically overestimated. White matter rCBF values were 17.4 ± 2.0 (mean ± SD) mL min-1 100 g-1 for PET and 21.8 ± 3.4 mL min-1 100 g-1 for perfusion CT. Grey matter rCBF values were 48.7 ± 5.0 mL min-1 100 g-1 for PET and 71.8 ± 8.0 mL min-1 100 g-1 for perfusion CT. The overestimation of grey matter rCBF could be reduced from 47% to 20% after normalization to white matter rCBF, but the difference was still significant.RCBF measured with perfusion CT does contain perfusion information, but neither quantitative nor relative values can substitute rCBF measured by15O-H2O PET yet. This, however, does not necessarily preclude a useful role in patient management.Quantitative measurement of the regional cerebral blood flow [rCBF] is a fundamental physiological parameter for characterizing the status of the brain tissue. RCBF measurements have important clinical implications in defining tissue ischemia [1,2], in diagnosing neurodegenerative diseases [3], and in locating and monitoring angiogenically active tumour tissues [4,5]. Positron-emission tomography [PET] measurements using a freely diffusible tissue tracer, oxygen-15-labelled water [15O-H2O], is regarded as one of the rCBF gold standards [6-8]. This cumbersome technique requires on-line tracer production from a cyclotron and continuous arterial blood sampling. It is expensive, technically demanding, and traumatic for the patient, and is rarely found outside specialized hospital units. Effects of Fructooligofructoses Chain Length on the Bifidobacteria of the Human Colon: A Pilot Study  [PDF]

Abstract: Human gastrointestinal health may be improved by the consumption of prebiotic food ingredients such as fructooligo-fructoses. A study was initiated to determine the effect of fructooligofructoses of different chain lengths on gastrointes-tinal parameters. Nineteen healthy subjects aged 20 - 57 y took part in a 10-week cross-over designed study. Subjects consumed either inulin or oligofructose for 3 weeks followed by a 2-week washout period between treatments. Stool samples were collected five times (baseline, 2 treatments, 2 washout) and analyzed for bifidobacteria. Daily records were kept for stool frequency, stool consistency and flatulence frequency. Bifidobacteria counts (cfu/ml) were higher (trending toward significance) during inulin and oligofructose intakes (1.2 × 107 ± 4.8 × 107 and 2.0 × 108 ± 4.7 × 108) and washout periods (2.9 × 106 ± 6.5 × 106 and 1.1 × 107 ± 1.6 × 107) than baseline counts (2.2 × 105 ± 5.1 × 105 and 2.9 × 106 ± 6.5 × 106), respectively. Inulin and oligofructose treatment periods had a significant effect on stool consistency (watery/very hard) and flatulence frequency, but not stool frequency, when compared to baseline (P < 0.05). Further research is needed to confirm these results due to small sample size and the need for a longer washout period between treatments.

Abstract: Concurrent dural and leptomeningeal metastatic carcinomatosis are very rare and have a poor prognosis. Here we present a woman with advanced estrogen receptor (ER) positive and progesterone receptor (PR) positive breast cancer who presented with leptomeningeal disease. Patient underwent multi targeted epigenetic therapies applied in a protocol called MTET. She continued to respond to the interval treatment, which consisted only of the nutraceutical agents. Here we discuss her case in detail and we believe that such an example might be applied to other patients in this situation resulting clinical improvement and less toxicity.
Sex-Role Orientation in Men Is Related to Salivary Testosterone Levels  [PDF]

Abstract: Previous research has implicated the involvement of androgens in sex-role orientation in males, from studies of 2nd to 4th digit ratio (a purported marker of prenatal testosterone). The present pilot study investigates the relationship between salivary testosterone levels and sex-role orientation using Bem Sex Role Inventory (BSRI) scores in adult males. Twenty-one males (aged 18-24) completed the BSRI and provided saliva samples for assay. BSRI Femininity scores were significantly negatively correlated with testosterone levels; the higher the Femininity scores, the lower the testosterone levels. There was no relation of BSRI Masculinity scores with testosterone levels. Our preliminary results add to the research suggesting that sex-role orientation in males may be partially related to underlying hormone levels.
Response shift and glycemic control in children with diabetes

Abstract: At year 1, thirty-eight children with type 1 diabetes attending a diabetes summer camp participated. At baseline and post-camp they completed the Problem Areas in Diabetes (PAID) questionnaire. Post-camp, the PAID was also completed using the 'thentest' method, which requires a retrospective judgment about their baseline functioning. At year 2, fifteen of the original participants reported their HbA1c.PAID scores significantly decreased from baseline to post-camp. An even larger difference was found between thentest and post-camp scores, suggesting scale recalibration. There was a significant positive correlation between year 1 HbA1c and thentest scores. Partial correlation analysis between PAID thentest scores and year 2 HbA1c, controlling for year 1 HbA1c, showed that higher PAID thentest scores were associated with higher year 2 HbA1c.Results from this small sample suggest that children with diabetes do show scale recalibration, and that it may be related to glycemic control.Diabetes is one of the most common chronic diseases of childhood. Adjustment to the disease and the demands of its complex regimen are formidable tasks even for adults. Children face these demands in the context of already challenging normative developmental tasks. Understanding children's diabetes-related problems can inform intervention designed to improve medical outcomes and quality of life for this population.Response shift is a theoretical construct that provides a framework for this investigation. In essence, it posits that people can adjust how they think about their quality of life when they encounter relevant new information. In this model, antecedents (e.g., demographics, personality), interact with a catalyst (intervention or change in health status) to elicit psychological mechanisms (e.g., social comparison) in order to accommodate the catalyst. Abstract: this article aims to evaluate the importance of americanist studies and the place of french researchers in the constitution of an international research network in the early 20th century. we try to understand how this disciplinary field emerged centered on the native american people, especially their cultural and social specificities. an analysis of the methodological underpinnings of the first collections reveals that special attention was given to the collection and the study of material culture since it bore witness to the diversity of human societies. young collaborators were hired to be in charge of and to analyze the collection of objects for the musée de l'homme in order to fill the lacuna of knowledge of the "native americans cultures". lévi-strauss' two missions (1935 and 1938) to make the inventory of the material culture that were part of this project inaugurated a new phase of americanist studies, focussed on the study of social structure. we evaluate the importance of this change for americanist research in the amazon and the consolidation of an international research network that came into being prior to the second world war. such a documental review, which questions the 'museologized' ethnographic object, is important because it allows us to reread the history of the discipline, in the light of the importance of the historical contexts, political debates and the place of americanist studies in brazil and in france. it is also possible to define the limits of an anthropology which becomes autonomous and takes a critical position in relation to colonial states and welfare services provided to native americans.
Spoke Top Antenna for Transient Radiation

Abstract: When an antenna transmits a short pulse of energy such as used for ultra-wide band applications, the pulse gets distorted upon transmission. To examine the properties of pulse transmission it is helpful to analyze the system in the time domain versus the frequency domain. Presented is a spoke top antenna for transient radiation. The spokes reduce the reflection from the open end of the antenna significantly reducing the trailing pulses commonly seen in the time domain. Comparisons are made with a dipole antenna. Both analytical, modeled and experimental results are presented.

Abstract: The time-space configurations of place and space are used to ground an analysis and discussion of the constitutive role of place, or virtual team members' respective local contexts, in the conduct of virtual teamwork. In contrast to the majority of current virtual teams research, which emphasizes the "spatial," or virtual aspect of virtual teamwork, this study uses an extended example, the establishment of a computer-conferencing infrastructure, to show the constitutive role played by local institutionalized rhythms, relationships, rules, politics, and resources in the enactment of virtual team tasks. Implications for studying, designing, and managing virtual teams are discussed.
