Abstract: chromosomal sex is established at fertilization by the presence of an x or y chromosome. the first step of male and female development is gonadal specialization in testes or ovaries; all other processes that follow result from secondary effects produced by testis and ovary hormones. gonadal determination and differentiation and the development of external genitalia involve time- and tissue-specific expression of genes forming a gene cascade. those genes, their expression profile and their role in the pathological manifestations related to gonadal and external genitalia development will be discussed in this review.

Association of the germline TP53 R337H mutation with breast cancer in southern Brazil

Abstract: We conducted a case-control study to determine the prevalence of the R337H mutation by sequencing TP53 exon 10 in 123 women with breast cancer and 223 age- and sex-matched control subjects from southern Brazil. Fisher's test was used to compare the prevalence of the R337H.The R337H mutation was found in three patients but in none of the controls (p = 0.0442). Among the carriers, two had familial history of cancer meeting the Li-Fraumeni-like criteria. Remarkably, tumors in each of these three cases underwent loss of heterozygosity by eliminating the mutant TP53 allele rather than the wild-type allele. Polymorphisms were identified within the TP53 (R72P and Ins16) and MDM2 (SNP309) genes that may further diminish TP53 tumor suppressor activity.These results demonstrate that the R337H mutation can significantly increase the risk of breast cancer in carriers, which likely depends on additional cooperating genetic factors. These findings are also important for understanding how low-penetrant mutant TP53 alleles can differentially influence tumor susceptibility.Mutations in the TP53 tumor supressor gene usually occur within the highly conserved DNA-binding domain (aa 100–298) and, when inherited, are typically associated with Li-Fraumeni syndrome (LFS), in which carriers develop a broad spectrum of cancers (e.g., breast, brain, soft tissue, bone, blood, and adrenal cortex tumors) during childhood or young adulthood [1]. Families with incomplete features of LFS are referred to as having Li-Fraumeni-like (LFL) syndrome [2,3].In southern Brazil, a unique germline TP53 point mutation resulting in an Arg to His amino acid substitution (R337H) within the C-terminal oligomerization domain is strongly associated with childhood adrenocortical tumors (ACT) [4-6]. Abstract: fibromyalgia is a syndrome defined by chronic widespread pain and sensitivity to pressure at at least 11 of 18 tender points. the aim of this random blind clinical trial was to assess the effectiveness of acupuncture on pain relief, on quality of sleep and life in fibromyalgia patients. from 20 female fibromyalgic patients (diagnosed according to the american college of rheumatology), mean aged 44 years, 12 completed the study, randomly assigned into two groups: group a (ga, n=5) received acupuncture according to the traditional chinese medicine; and group b (gb, n=7) received acupuncture at eight tender points: at the occiput, trapezius muscle, supraspinatus muscle, and lateral epicondyle. pain was assessed by a visual analogue scale and by dolorimetry; quality of life, by the fibromyalgia impact questionnaire (fiq); and sleep, by the post sleep inventory. both groups received acupuncture once a week for eight weeks, with insertion of eight needles for 25 minutes. data were statistically analysed and significance level set at ±=0.05. results show significant sleep improvement in ga; and, in gb, lesser pain as detected by vas, higher pain threshold on tender points, better sleep and improvement in fiq items pain, morning tiredness, anxiety, and depression (p<0.05). these data suggest that acupuncture, especially that with tender point needle insertion, provides pain relief, better quality of life and of sleep in fibromyalgic patients.

Abstract: the aim of this study was to establish associations between quality of life and pain, anxiety, and depression, in subjects aged 35 to 60 years old. three hundred and four subjects were evaluated and divided into three groups according to pain features: chronic and widespread pain (cwp), local pain (lp), and sporadic pain (sp) groups. pain intensity was assessed by means of a visual analog scale (vas); anxiety by the state-trait anxiety inventory; depression, by the beck depression scale; and quality of life, by the medical outcomes study 36-item short-form health survey. subjects mean age was 49.1±6.8 years old; 80.3% were women; and 35% of the sample reported chronic widespread pain. results show that the cwp group presented more intense pain, anxiety and depression, as well as worst quality of life, followed by lp and sp groups (p<0.001). a strong correlation was found between quality of life and depression (r=-0.73), and moderate ones between quality of life and pain (r=-0.41), anxiety-trait (r=-0.65) and anxiety-state (r=-0.58) (p<0.0001). subjects with data show negative correlation for quality of life and the variables assessed and the cwp group reported the most intense symptoms.

Abstract: Objetivou-se nesta pesquisa quantificar as rea es vacinais em carca as de animais da espécie bovina e ra a Nelore de diferentes idades e sexos, provenientes de sistemas extensivos de cria o. Foram avaliados 850 animais abatidos em frigorífico do município de Sinop/MT. A detec o e retirada das les es foi realizada na linha I de inspe o de rotina de carca a. O ensaio estatístico foi conduzido no delineamento inteiramente casualizado com tratamentos dispostos em arranjo fatorial 2x4 (dois sexos e quatro grupos de idades). A avalia o do peso foi feita por análise de variancia, seguido de teste de médias com utiliza o do teste de Student-Newman-Keuls. O teste de Fisher mostrou uma intera o entre idade e sexo. Nos machos n o houve diferen a entre os grupos de diferentes idades, entretanto, nas fêmeas, o grupo acima de cinco anos foi estatisticamente inferior em rela o aos outros grupos. Na compara o entre machos e fêmeas apenas nos animais acima de cinco anos foi encontrada diferen a significativa, e as fêmeas apresentaram maior peso da amostra que os machos. O peso médio das por es retiradas foi de 1,28kg/animal, o que representa uma perda econ mica de US$1,68/animal, superior nas fêmeas (1,30kg) em rela o aos machos (1,24kg). Os resultados revelaram alta incidência de les es vacinais nas carca as dos animais em todas as categorias analisadas, o que leva à redu o da sua qualidade e valor, e mostra a importancia da melhoria dos manejos de vacina o para minimizar os prejuízos do produtor. The purpose of this research was to quantify the vaccine reaction in carcasses of Nellore cattle of different ages and sex, raised in extensive system. It was evaluated 850 animals that were at a slaughterhouse in Sinop/MT. The detection and removal of the vaccine lesions were performed in line-I of the routine carcass inspection. The statistical test was conducted in a completely randomized design with treatments in 2x4 factorial arrangement (two sexes and four age groups). The weight analysis was performed by ANOVA followed by the test of Student-Newman-Keuls to compare means. The Fisher test showed an interaction between age and sex. In males there was no difference between age groups and among females, the group of over 5 years old was statistically lower than the other two groups. In the comparison between males and females there was significant difference only in animals of over 5 years old and the females had lower sample weight than the males. The average weight of the sections removed from the carcass was 1.28kg/animal, which represents an economic loss

Abstract: it should take into account the pediatric context and the child's development moment to evaluate the quality of life in childhood from a standardized instrument. we present the questionnaire auqei validation by its application in 353 children, between 4 and 12 years-old, obtaining a cut off point of 48, internal consistency with a crombach's alpha of 0.71 and construct validity (r = 0.497). with these psychometrical qualities, the scale shows its importance as a diagnostic tool.

Apresentamos a valida o do questionário AUQEI, a partir de sua aplica o em 353 crian as, com idade entre 4 e 12 anos, obtendo um ponto de corte de 48, consistência interna representada por um alfa de Crombach da ordem de 0,71 e validade externa (r= 0,497). Com essas qualidades psicométricas, a escala mostra-se, assim, importante como instrumento diagnóstico.

Abstract: BACKGROUND: Fibromyalgia is a painful syndrome characterized by widespread chronic pain and associated symptoms with a negative impact on quality of life. OBJECTIVES: Considering the subjectivity of quality of life measurements, the aim of this study was to verify the discriminating power of two quality of life questionnaires in patients with fibromyalgia: the generic Medical Outcomes Study 36-Item Short-Form Health Survey (SF-36) and the specific Fibromyalgia Impact Questionnaire (FIQ). METHODS: A cross-sectional study was conducted on 150 participants divided into Fibromyalgia Group (FG) and Control Group (CG) (n=75 in each group). The participants were evaluated using the SF-36 and the FIQ. The data were analyzed by the Student t-test (α=0.05) and inferential analysis using the Receiver Operating Characteristics (ROC) Curve - sensitivity, specificity and area under the curve (AUC). The significance level was 0.05. RESULTS: The sample was similar for age (CG: 47.8±8.1; FG: 47.0±7.7 years). A significant difference was observed in quality of life assessment in all aspects of both questionnaires (p<0.05). Higher sensibility, specificity and AUC were obtained by the FIQ (96%, 96%, 0.985, respectively), followed by the SF-36 (88%, 89% and 0.948 AUC). CONCLUSION: The FIQ presented the highest sensibility, specificity and AUC showing the most discriminating power. However the SF-36 is also a good instrument to assess quality of life in fibromyalgia patients, and we suggest that both should be used in parallel because they evaluate relevant and complementary aspects of quality of life. OBJETIVOS: Considerando a subjetividade da mensura o de qualidade de vida, o objetivo deste estudo foi avaliar o poder de discrimina o de dois questionários que avaliam a qualidade de vida de pacientes com fibromialgia: o genérico Medical Short Form Healthy Survey (SF-36) e o específico Questionário do Impacto da Fibromialgia (QIF). MéTODOS: Foi conduzido um estudo transversal com 150 indivíduos, divididos em dois grupos: grupo fibromialgia (FM) e grupo controle (GC) (n=75 em ambos).
