Cholesterol Depletion Disorganizes Oocyte Membrane Rafts Altering Mouse Fertilization

Abstract: Drastic membrane reorganization occurs when mammalian sperm binds to and fuses with the oocyte membrane. Two oocyte protein families are essential for fertilization, tetraspanins and glycosylphosphatidylinositol-anchored proteins. The firsts are associated to tetraspanin-enriched microdomains and the seconds to lipid rafts. Here we report membrane raft involvement in mouse fertilization assessed by cholesterol modulation using methyl-β-cyclodextrin. Cholesterol removal induced: (1) a decrease of the fertilization rate and index; and (2) a delay in the extrusion of the second polar body. Cholesterol repletion recovered the fertilization ability of cholesterol-depleted oocytes, indicating reversibility of these effects. In vivo time-lapse analyses using fluorescent cholesterol permitted to identify the time-point at which the probe is mainly located at the plasma membrane enabling the estimation of the extent of the cholesterol depletion. We confirmed that the mouse oocyte is rich in rafts according to the presence of the raft marker lipid, ganglioside GM1 on the membrane of living oocytes and we identified the coexistence of two types of microdomains, planar rafts and caveolae-like structures, by terms of two differential rafts markers, flotillin-2 and caveolin-1, respectively. Moreover, this is the first report that shows characteristic caveolae-like invaginations in the mouse oocyte identified by electron microscopy. Raft disruption by cholesterol depletion disturbed the subcellular localization of the signal molecule c-Src and the inhibition of Src kinase proteins prevented second polar body extrusion, consistent with a role of Src-related kinases in fertilization via signaling complexes. Our data highlight the functional importance of intact membrane rafts for mouse fertilization and its dependence on cholesterol.

Abstract: this article approaches the conflict argentina and uruguay mantain since 2002, due to the installation of cellulose plants in uruguayan territory, on the binational waters of the uruguay river. this conflict has gained international relevance and has transformed itself into a true public scientific controversy since 2005. this paper uses this case to analyze citizen participation in environmental conflicts, especially participation in cases of controversies related to the tension between environmental values and economic or political priorities. the scientific arguments were used by some of the actors in the "papermills" conflict as the basis for a political discussion, highlighting the complex relation between politics and scientific controversies that become public. we maintain that the case creates a historical precedent of extreme value for the future of citizen participation in these issues, in that it is an emblematic case of citizens organizing to inform themselves, act, and constitute themselves as a legitimate player in these debates.

Abstract: En este artículo se aborda el conflicto que mantienen desde el 2002 Argentina y Uruguay, debido a la instalación de plantas de celulosa en territorio uruguayo, sobre las aguas binacionales del Río Uruguay. El conflicto fue adquiriendo relevancia internacional y se transformó en una verdadera controversia científica pública a partir del 2005. Mediante el presente caso, analizaremos la participación ciudadana en conflictos medioambientales, en especial la participación en casos de controversias relacionadas con la tensión entre valores ambientales y prioridades económicas o políticas. Los argumentos científicos fueron utilizados por algunos de los actores como asiento de una discusión política de fondo, poniendo de relieve la compleja relación de la política con las controversias científicas que devienen públicas. Sostenemos que el caso sienta un precedente histórico y de sumo valor para el porvenir de la participación ciudadana en estas temáticas, en tanto constituye un caso emblemático de organización por parte de la ciudadanía, para informarse, actuar y constituirse en una voz privilegiada en estas temáticas. Abstract: the demand for information concerning the cataloguing process is a constant need among librarians working for the national health system. this need is required both for the development of the traditional functions of the institutions and to face the new challenges related to the web. with the aim of providing a version of the section devoted to cataloguing in the procedures manual designed for the national medical library, a detailed description is made of the documented procedure according to the iso-9000 standards. the knowledge of these techniques constitutes an urgent need for the development of the information systems in the current situation. its validity is renewed at present due to the spreading of the use of metadata in internet.

Abstract: the government information locator service, one of the latest technologies of information adopted by its strategic nature to solve the disorganization of internet and the deficiencies of the search tools is presented. based on the metadatum principle, this us standard offers a service to locate information that alows to discover, identify, describe and to accede directly to the information resources available in internet starting from international standards of search and information exchange. likewise, the health information locator is made known. this is an application of gils in the latin american health sector, a project developed by the latin american and caribbean health sciences information system in order to meet the present needs derived from the introduction of the new paradigm of information system represented by internet and to improve the efficiency of information retrieval.

Abstract: validity of the classification system of the national library of medicine of the united states. a quick examination of internet allows us to confirm the recognized capacity of the library methods such as cataloguing, classification and indexing for organizing information. the classification concepts, for example, are applied to useful tools like yahoo-type thematic directories or the guides prepared by experts, which take into account the experience of librarians to counterbalance the prevailing chaos in internet. the classification system of the national library of medicine of the united states that is officially applied in the national information system of medicine is thoroughly studied along with the presentation of its history and development. the structure of this system and the procedure to be followed in the classification process are described. the translation of the first updating of the classifiers published by the national library of medicine after its fourth edition is included.

Abstract: historically, wage-paid work has been part of the male body, in the masculine sexuality, in his relationship with productive work. this masculine image covers the organizations and their processes, leave aside women and contributes to maintain the genre segregation in women. in the case of the unions, measures pointing to develop a critical mass of women through dues or quotas constitute a bet to introduce necessary changes related to the organization culture and to introduce the genre perspective in the unionist action as well as the collective negotiation. in this paper, we will examine the application of the feminine unionist dues law (law no 25.674) in neuquén ceramists employees and workers union (soecn) to observe the degree of inclusion and participation of women in it. we will also study the strategies built in relationship to the implementation and appropriation of this space as a fighting place as well as the complex and heterogenous identities they build or try to build in opposition to the ones instituted by the androcentric system in which we are in. this approach gets more complex because it focuses primarily on the vital experiences of a group of women that, in the process of fighting in order to recover their work sources, they formed the women committee in the former zanon. nowadays, their work activities in the present fasinpat allow them to be in charge of executive positions in the union they are part of, the soecn, showing in this way, a different dynamics from the rest of work unions in our province and perhaps, in the country.

Abstract: the presentation and organization of collections built with greenstone system are endowed with a remarkable flexibility adds value to information access in the interests, characteristics and needs of end users. article creation and customization of a collection of digital documents based on greenstone, described the custom design consisting of a collection of files. doc and. pdf with regard to navigation and search mechanisms on the one hand, and changes in some elements of their about page, on the other, which were obtained from dissimilar formats for strings and macro classifiers extra.dm. to deepen the familiarity of the advantages of this system and expand the range of possibilities offered in the second part of this work, we continued to customizing a similar collection on the basis of the application of other format chains operating immediate changes in the provision of data as well as the appearance and organization of the information contained on the pages generated by this system. such a set of possible customizations illustrates, to some extent, the alternative configuration of the outputs of this application which also run in an expeditious manner.
