Abstract: this paper analyzes the key drivers of colombian household's indebtedness during the past 14 years, using financial accounts provided by the central bank (banco de la república) and a random sample of individuals tax income statements reported to the tax office (dian) between 1993 and 2004. based on a simple choice model, the results suggest that collateral and carried debt are the main drivers of indebtedness, in consequence policy should be household equity and interest rate oriented in order to avoid a massive default crisis such as the one ocurred in 1999.
Anti-glucuronoxylomannan IgG1 specific antibodies production in Cryptococcus neoformans resistant mice

Abstract: introduction. cryptococcus neoformans is a ubiquitous fungus which has been shown to be responsible for infections in individuals with impaired cell mediated immunity, such as patients infected with human immunodeficiency virus (hiv). c. neoformans has a polysaccharide capsule composed of glucuronoxylomannan (gxm), which acts as a major virulence factor and is considered to be a thymus independent type-2 antigen (ti-2). objective. to study the production kinetics of igg subclasses specific for gxm, the cross reactive antibodies to streptococcus pneumoniae polysaccharide and to determine spleen b cell subpopulations in murine models of cryptococcosis with different susceptibilities to the infection. materials and methods. antibodies were detected by elisa at different time intervals after c. neoformans infection in moderately resistant (balb/c), highly resistant (cba/j) and susceptible (c57bl/6) mouse strains. b cells subpopulations were determined by flow cytometry analysis. results. early production of igg1, described as protector antibodies, coincided with a decrease of the number of c. neoformans colony forming units (cfu) in the lungs. polysaccharide cross-reactive antibodies were detected in the three mouse strains, and were highest in the susceptible strain (c57bl/6), which showed the highest proportion of splenic cd5+ b lymphocytes. in contrast, cba/j mice showed the highest levels of cd43+ b. conclusions. these findings suggest that igg1 antibodies specific for gxm, are implicated in host protection against c. neoformans infection, and might be regulated by cd43+ cells. they also suggest that cross reactivity antibodies are not important in the protection of c. neoformans infection.
Un entorno para la representación de aspectos en esquemas preconceptuales

Abstract: since the mid-nineties, there has been a trend for identifying aspects from the earliest stages of software development lifecycle. the standard case (computer-aided software engineering) tools are used to represent aspects in uml (unified modeling language) diagrams. however, the analyst makes this task by interpreting the domain in a subjective way, with no validation from stakeholders. for this reason, in this paper we present an emf-based environment for representing aspects in pre-conceptual schemas, and then translating them into class and sequence diagrams. we exemplify the use of such environment with a case study.

Abstract: entity-relationship diagram (erd) is one of the used in modelling the domain information. several proposals have emerged for speeding up and improving the software development process by either automatically or semi-automatically obtain the erd. natural language, controlled languages, and intermediate representations have been used in such a task. the stakeholders (people with some concern in application development), when untrained, are incapable to understand several of such representations. as a consequence, stakeholder active participation in software development is highly restricted. trying to solve these problems, a set of heuristic rules for automatically obtaining erd and its sql-based equivalence is proposed in this paper. the starting point is un-lencep, a controlled language already used for generating other artefacts belonging to software application development.

Abstract: Introducción: las células asesinas naturales (NK y NKT) son una población de linfocitos que circulan en bajos porcentajes en sangre periférica. Ambos tipos de células realizan un papel importante en la respuesta inmune en condiciones normales o en procesos patológicos como ciertos tumores, abortos espontáneos y en el rechazo a trasplantes, lo cual les confiere un gran interés clínico. Objetivo: el presente trabajo pretende establecer valores para las células NK y NKT por medio de citometría de flujo en una población adulta de donantes de un banco de sangre de Bogotá. Metodología: se recolectaron 104 muestras de donantes y se realizó un estudio con los marcadores CD3, CD16 y CD56. Resultados: de las muestras 47.2% fueron mujeres y 52.8% hombres. Para las mujeres el porcentaje de células fue: NK 14.6% (± 12.1) y NKT 3.0% (± 2.5); para hombres NK 25.3% (± 21.3) y NKT 3.5% (± 2.9). Los valores absolutos de células para mujeres fueron NK 298.8 /μL (± 253.9) y NKT 70.1 /μL (± 47.7); para hombres NK 526.1 /μL (± 448) y NKT 95.5 /μL (± 77). Existe una diferencia estadística entre los valores absolutos par las células NK entre hombres y mujeres (p= 0.0004). Introduction: the natural killer cells (NK y NKT) are a population of lymphocytes that circulate in the peripheral blood but in low percentages. Both types of cells play an important role in immune response under normal conditions or in pathological processes as it is the case of certain tumors, spontaneous abortions and transplant rejections, what makes them very interesting from the clinical interest point of view. Objective: the aim of this work is to establish values for NK and NKT cells by means of flow cytometry in a group of adult population of blood donors in a blood bank in Bogotá. Methodology: 104 donor samples were collected and a study was carried out with CD3, CD56 and CD16 markers. Results: 47.2% of the samples were women and 52.8% men. In the case of women the cells percentage was: NK 14.6% (± 12.1) and NKT 3.0% (± 2.5); for men NK 25.3% (± 21.3) and NKT 3.5% (± 2.9). The absolute cells values for women were NK 298.8 /μL (± 253.9) and NKT 70.1 /μL (± 47.7); for men NK 526.1 /μL (± 448) and NKT 95.5 /μL (± 77). There is a statistical difference between the absolute values by the NK cells between men and women. (p= 0.0004).

Abstract: Las células dendríticas son importantes reguladores de la respuesta inmune celular. Estas células son consideradas como presentadoras profesionales de antígenos, con potente capacidad de estimular linfocitos T vírgenes. El objetivo de este trabajo fue establecer un modelo de cultivo in vitro para la obtención de células dendríticas mieloides humanas a partir de monocitos CD14+ de sangre periférica. Se utilizaron citoquinas como IL-4 y factor estimulador de colonias granulo-monocíticas (GM-CSF) para la obtención de células inmaduras y la posterior incubación con PGE2 y TNFa para la obtención de células maduras.Utilizando este modelo se obtuvieron células dendríticas inmaduras al día 5 y células dendríticas maduras al día 7, lo cual fue confirmado por los cambios en la morfología y la expresión de marcadores de superficie como CD14, CD86, HLA-DR y CD83. El estudio de la cinética de maduración de células dendríticas, es de utilidad en protocolos de inmunoterapia para diferentes tipos de cáncer y en la definición de mecanismos inmunes involucrados en diferentes inmunopatologías causadas por antígenos provenientes de alergenos o de microorganismos.

Abstract: Desde mediados de los 90 se viene dando la tendencia a identificar aspectos desde las etapas más tempranas del desarrollo de software. Las herramientas CASE (Computer-Aided Software Engineering) estándar permiten representar los aspectos en diagramas UML (Unified Modeling Language) pero, para ello, se requiere que el analista interprete de manera subjetiva el dominio, sin que medie una validación de los interesados. Es por esto que en este artículo se proponeun entorno, basado en EMF (Eclipse Modelling Framework), que permite representar los aspectos en esquemas preconceptuales para luego traducirlos a los diagramas de clases y secuencias. Abstract: Entidad-relación es uno de los diagramas que se utilizan en el desarrollo de modelos para representar la información de un dominio. Con el fin de agilizar y mejorar el proceso de desarrollo de software, diferentes propuestas surgieron para contribuir en la obtención automática o semiautomática del diagrama entidad-relación. Varias de estas propuestas utilizan como punto de partida lenguaje natural o lenguaje controlado, mientras otras propuestas utilizan representaciones intermedias. Los interesados en el desarrollo de una aplicación de software no suelen comprender varias de las representaciones utilizadas sin tener previa capacitación, lo cual restringe la participación activa del interesado en todas las etapas del desarrollo. Con el fin de solucionar estos problemas, en este artículo se propone un conjunto de reglas heurísticas para la obtención automática del diagrama entidad-relación y su representación en SQL. Se toma como punto de partida el lenguaje controlado UN-Lencep, que ya se emplea para la generación de otros artefactos en el desarrollo de aplicaciones de software. Flow cytometry model for the detection of neutralizing antibodies against of IFN-β Modelo de detección de anticuerpos neutralizantes contra IFN-β mediante citometría de flujo

Abstract: Introduction. Interferon beta (IFN-β) is a treatment for relapsing remitting multiple sclerosis (RRMS). However, the therapeutic use of recombinant proteins induces a humoral immunologic response resulting in the induction of binding (BAbs) or neutralizing (NAbs) antibodies against the biological product. The presence of neutralizing antibodies has been associated with decreased IFN-β treatment efficacy. Materials and methods. Two different tumor cell lines (K562 and U937) were cultivated with human recombinant IFN-β1a at different concentrations and time in order to measure the expression of intracellular ISG15, an inducible molecule in the IFN-β1a signaling cascade. Blood was obtained from non-immunized and IFN-β1a immunized (100,000 IU) New Zealand rabbit. The presence of BAbs was evaluated by ELISA. For NAbs detection, sera 1:20 dilution were added to the IFN-β1a stimulated cell lines and ISG15 expression was evaluated by flow cytometry. Results. K562 cell was selected with a dose of 1000 IU of IFN-β1a, and a 1:100 dilution for the primary antibody and a 1:200 dilution for the secondary antibody as the best conditions for the assay. ISG15 expression was compared between cells alone or cultivated with IFN-β1a. Mean fluorescence intensity (MFI) for ISG-15 expression median was 198 arbitrary units (AU) with interquartile ranges of 173-231 AU for non-stimulated cells and 430 UA with interquartile ranges of 316-611.5 UA for IFN-β1a stimulated cells (p=0.008).Immunized rabbit sera decreased the expression of ISG-15 in K5652 cells stimulated with IFN-β1a, whereas non-immunized rabbit sera did not. Conclusions. This rabbit model demonstrates that ISG-15 expression evaluated with flow cytometry can be used as a detection assay for NAbs. Introducción. El interferón beta (IFN-β) se usa para tratamiento de la forma recaída-remisión de la esclerosis múltiple. Sin embargo, el uso de proteínas recombinantes como medicamentos, pueden generar la producción de anticuerpos, disminuyendo así la efectividad del tratamiento. Objetivo. Estandarizar una técnica de detección de anticuerpos neutralizantes contra IFN-β mediante citometría de flujo. Materiales y métodos. Se cultivaron dos líneas tumorales humanas (U937 y K562) con IFN-β1a humano recombinante y mediante citometría de flujo se determinó la expresión de la proteína ISG15 intracelular. Los sueros fueron obtenidos de un conejo Nueva Zelanda pre- y post-inmunización con 100.000 UI de IFN-β1a en adyuvante de Freund. Para la detección de anticuerpos neutralizantes, se estimularon células K562 con IFN-β1a pre-incubado
