Abstract: 在YEPD培养基中添加NaCl,可以诱导酿酒酵母(Saccharomyces cerevisiae)细胞内3-磷酸甘油脱氢酶的形成,当NaCl浓度达5%时,酶比活从0.05U/mg提高到0.5U/mg;若再限制培养基中葡萄糖浓度在100mg/L以下,酶比活可达到0.89U/mg。酶比活与培养基中的NaCl浓度的函数关系式为:Sa-0.129C3-0.038C2+0.034C+0.063(O≤C≤5%)。粗酶液经Sephadex G一25凝胶过滤,Blue Sepharose亲和层析以及Mono Q离子交换等步骤,提纯123.6倍,得纯酶液。经SDS一凝胶电泳测得分子量为45 000±2 000。酶的最适温度为51℃,最适pH值为6.8。保温30分钟的半失活温度(t1/2)为41℃。NADH和DHAP的Km(mmol/L)值分别为:0.0177和0.134。
Purification and the Secondary Structure of Fucoidanase from Fusarium sp. LD8

Abstract: The fucoidanase from Fusarium sp. (LD8) was obtained by solid-state fermentation. The fermented solid medium was extracted by citric acid buffer, and the extracts were precipitated by acetone and purified by Sephadex G-100 successively. The results showed that the specific fucoidanase activity of purified enzyme was 22.7-fold than that of the crude enzyme. The recovery of the enzyme was 23.9%. The purified enzyme gave a single band on SDS-PAGE gel, and the molecular weight of fucoidanase was about 64?kDa. The isoelectric point of the enzyme was 4.5. The enzyme properties were also studied. The results showed that the optimum temperature and pH were 60°C and 6.0, respectively; the temperature of half inactivation was 50°C, and the most stable pH for the enzyme was 6.0. , and the of the enzyme was 8.9?mg·L?1 and 2.02?mg·min?1·mL?1 by using fucoidan from Fucus vesiculosus as substrate. The compositions of the secondary structure of fucoidanase were estimated by FTIR, the second derivative spectra, and the curve-fitting analysis of the amide I bands in their spectra. The results showed that -sheet was the dominant component (58.6%) and -helix was the least (12%); the content of -turn and random coil were 15.39% and 14.5%, respectively. 1. Introduction Sulfated polysaccharides, named fucoidan, could be extracted from many marine brown algae. The main differences of structural characterization in algal fucoidan originate from their species; the backbone of fucoidan isolated from Laminaria cichorioides [1] was consisting of (1–3) linked fucopyranose residues, and that from L. japonica was primarily (1–3) linked fucopyranose residues (75%) and a few (1–4) fucopyranose linkages (25%) [2]; The fucoidan from Fucus serratus [3], F. evanescens [4], and F. distichus [5] was shown to contain a backbone consisting of alternating (1–3) and (1–4) linked a-L-fucopyranose residues. Investigators were interested in screening fucoidan which was isolated from different brown algal species; fucoidan was attributed several different bioactivities including anticoagulant [6–8], antithrombotic/antithrombin activity [9], antiviral [10], and other activities [11]. The biological effects of fucoidan depended on the molecular mass, sulfate content, and sugar constituents [12, 13]. The low molecular weight fucoidan (LMWF) could be obtained by fucoidanase (E.C. enzymolysis, and it could hydrolyze fucoidan to sulfated LMWF without removal of its side substitute groups. Abstract: The production process of xylanase of solid-state fermentation from Aspergillus nigerwas studied. The maximum enzyme activity can be obtained when the fungus wat cultured for 3days. The ratio between the solid culture and the extracting agent was about 1:7(w / v). Thesaturation condition of (NH4)2SO4 is the range from 6O%~65%. Two kinds of xylanase enzymepowder were obtained by both freezer drying and at 40C heating drying, and the rate of recovery71% and 51% were respectively. The highest xylanase activity has about l5400u / g by freeerdrying, 15395u / g by heating drying. The related properties of enzyme were analysed. the optimaltemperature is 55t and pH is 4.6. Th tempetatUre for 1oss half achvity(t l / 2) in a hour is 54t.Among the four kinds of hemicellulose, the strongest and the weakest hydrolytic affinity ofxylanase are wheat bran and rice straw respectively.

Abstract: Objective To observe the influence of neuregulin-1 on the cardiac function of post-myocardial infarction rats. Methods Left ventricular MI was created in Sprague-Dawley rats by ligalion of the left anterior descending coronary. Six months after the operation, rats were evaluated with echocardiology methods. 36 rats that had an infarct area and a EF around 60% were randomized into 3 groups: MI group(n=12) were injected a blank vehicle fluid intravenously for 5 days, after which they continued to be raised on standard food and water for 30 days. MI+NRG group(n=12). received NRG-1 10(J.g ? kg ' intravenously for 5 days, after which they continued to be raised on standard food and water for 30 days. Ml+Capl group (n=12) received captopril orally (dissolved in their drinking water 2g/L) for 30 days , after which tap water substituted the solution for 5 days. Final echocardiographic and hcmodynamic measurements were made at the end of 1 month of therapy. Total RNA was extracted from fro/en left ventricular tissues, and was reverse transcribed into first-strand PCR was performed with primers for BNPx ANP. Results Rats treated with neuregulin had a smaller LVDs (F=0.()14), a better LVEF (P=0.004 ),and a tendency towards less lung perfusion than untreated rats. Neuregulin decreased the expression of ANP mRNA in the ventricle (P=0.025 ).Conclusion Neuregulin markedly improved the cardiac function of rats that survived myocardial infarction, and decreased the expression of ANP mRNA in the ventricle.

Analysis of Hepatitis B Virus Intrahepatic Covalently Closed Circular DNA and Serum Viral Markers in Treatment-Naive Patients with Acute and Chronic HBV Infection

Abstract: Background This study aimed to investigate the relationships of intrahepatic cccDNA with serum HBsAg and with HBV DNA in treatment-naive patients throughout acute and chronic HBV infection. Methods A total of 120 patients who had a liver biopsy were enrolled, including 19 with acute hepatitis B (AHB), and 101 patients with chronic HBV infection (CHB) of whom were 10 in immune-tolerant (IT) phase, 59 in immune-clearance (IC) phase, 8 in low-replicative (LR) phase, and 24 in HBeAg-negative hepatitis (ENH) phase. Intrahepatic cccDNA, serum HBsAg and serum HBV DNA levels were comparatively analyzed. Results The median intrahepatic cccDNA levels were 0.18 4.80, 3.81, 0.22 and 0.97 copies/cell for patients with AHB, CHB-IT, CHB-IC, CHB-LR, and CHB-ENH, respectively. In AHB patients, intrahepatic cccDNA was positively correlated with serum HBsAg (r = 0.665, P = 0.003), as well as serum HBV DNA (r = 0.536, P = 0.022). In CHB patients, intrahepatic cccDNA was positively correlated with serum HBsAg in the IC phase (r = 0.392, P = 0.005), and with serum HBV DNA in the IC phase (r = 0.301, P = 0.036) and ENH phase (r = 0.588, P = 0.013). HBV replicative efficiency, defined as the ratio of serum HBV DNA to intrahepatic cccDNA, was obviously lower in AHB and CHB-LR patients than in CHB-IT, CHB-IC and CHB-ENH patients (0.70 and 0.53 vs. 1.12, 1.09 and 0.99, P<0.001, values were logarithmic transformed for analysis). In CHB-IC patients, HBV replicative efficiency was positively correlated with histological activity index of liver inflammation (r = 0.308, P = 0.009). Conclusion Serum HBsAg and HBV DNA levels may reflect the amount of active intrahepatic cccDNA in treatment-naive AHB and CHB-IC patients. Reduced intrahepatic cccDNA and HBV replicative efficiency may imply effective immune control of HBV infection.

Abstract: Taking the Um7 meandering river complex sandstone reservoir of Bohai Bay C oilfield as an example, the interlayer type is defined according to the interlayer lithology and sedimentary genesis, and the identification mode of different types of interlayer is established, and the interlayer comparison of the whole area is carried out by using this mode. The results show that the interlayer can be further defined as the stable distributed interlayer and the randomly distributed interlayer according to the morphology of the interlayer. The distribution characteristics of the two types of interlayers are characterized by the well-to-well comparison method and the data analysis method, and the three-dimensional characterization is carried out. The results more closely reflect the spatial distribution of the interlayer, but also are closer to the underground real situation.
Submicroscopic subtelomeric aberrations in Chinese patients with unexplained developmental delay/mental retardation

Abstract: This study included 451 Chinese children with moderate to severe clinically unexplained DD/MR. The subtelomere-MLPA (multiplex ligation dependent probe amplification) and Affymetrix human SNP array 6.0 were used to determine the subtelomeric copy number variations. The exact size and the breakpoint of each identified aberration were well defined.The submicroscopic subtelomeric aberrations were identified in 23 patients, with a detection rate of 5.1%. 16 patients had simple deletions, 2 had simple duplications and 5 with both deletions and duplications. The deletions involved 14 different subtelomeric regions (1p, 2p, 4p, 6p, 7p, 7q, 8p, 9p, 10p, 11q, 14q, 15q, 16p and 22q), and duplications involved 7 subtelomeric regions (3q, 4p, 6q, 7p, 8p, 12p and 22q). Of all the subtelomeric aberrations found in Chinese subjects, the most common was 4p16.3 deletion. The sizes of the deletions varied from 0.6 Mb to 12 Mb, with 5-143 genes inside. Duplicated regions were 0.26 Mb to 11 Mb, with 6-202 genes inside. In this study, four deleted subtelomeric regions and one duplicated region were smaller than any other previously reported, specifically the deletions in 11q25, 8p23.3, 7q36.3, 14q32.33, and the duplication in 22q13. Candidate genes inside each region were proposed.Submicroscopic subtelomeric aberrations were detected in 5.1% of Chinese children with clinically unexplained DD/MR. Four deleted subtelomeric regions and one duplicated region found in this study were smaller than any previously reported, which will be helpful for further defining the candidate dosage sensitive gene associated with DD/MR.Developmental delay/mental retardation (DD/MR) occurs in 1%-3% of the general population [1,2]. MR is defined as a significant impairment of both cognitive (IQ < 70) and social adaptive functions, with onset before 18 years of age.
