Abstract: The wheat stripe rust fungus, Puccinia striiformis f. sp. tritici (Pst), does not have a known alternate host for sexual reproduction, which makes it impossible to study gene linkages through classic genetic and molecular mapping approaches. In this study, we compared 4,219 Pst expression sequence tags (ESTs) to the genomic sequence of P. graminis f. sp. tritici (Pgt), the wheat stem rust fungus, using BLAST searches. The percentages of homologous genes varied greatly among different Pst libraries with 54.51%, 51.21%, and 13.61% for the urediniospore, germinated urediniospore, and haustorial libraries, respectively, with an average of 33.92%. The 1,432 Pst genes with significant homology with Pgt sequences were grouped into physical groups corresponding to 237 Pgt supercontigs. The physical relationship was demonstrated by 12 pairs (57%), out of 21 selected Pst gene pairs, through PCR screening of a Pst BAC library. The results indicate that the Pgt genome sequence is useful in constructing Pst physical maps.

Abstract: In this study, we have a analysis of the ARM core unit design of a remote video monitoring system. An important aspect of a remote video monitoring system is that the image information stored in the monitoring system is transmitted to a remote PC through the GSM/GPRS network. There into the application of the ARM core unit is one part of the design of the entire remote video monitoring system. An ARM core unit is designed based on ARM architecture and ARM chips. It includes hardware and software design. Practice shows high stability and reliability of the design.
Identification of expressed genes during compatible interaction between stripe rust (Puccinia striiformis) and wheat using a cDNA library

Abstract: Total RNA, extracted from susceptible infected wheat leaves harvested at 3, 5 and 8 days post inoculation (dpi), was used to create a cDNA library, from which 5,793 ESTs with high quality were obtained and clustered into 583 contigs and 2,160 singletons to give a set of 2,743 unisequences (GenBank accessions: GR302385 to GR305127). The BLASTx program was used to search for homologous genes of the unisequences in the GenBank non-redundant protein database. Of the 2,743 unisequences, 52.8% (the largest category) were highly homologous to plant genes; 16.3% to fungal genes and 30% of no-hit. The functional classification of all ESTs was established based on the database entry giving the best E-value using the Bevan's classification categories. About 50% of the ESTs were significantly homologous to genes encoding proteins with known functions; 20% were similar to genes encoding proteins with unknown functions and 30% did not have significant homology to any sequence in the database. The quantitative real-time PCR (qRT-PCR) analysis determined the transcription profiles and their involvement in the wheat-Pst interaction for seven of the gene.The cDNA library is useful for identifying the functional genes involved in the wheat-Pst compatible interaction, and established a new database for studying Pst pathogenesis genes and wheat defense genes. Global Transcriptome Profiling of Salicornia europaea L. Shoots under NaCl Treatment

Abstract: Background Soil salinity is a major abiotic stress that limits agriculture productivity worldwide. Salicornia europaea is well adapted to extreme saline environments with more than 1,000 mM NaCl in the soil, so it could serve as an important model species for studying halophilic mechanisms in euhalophytes. To obtain insights into the molecular basis of salt tolerance, we present here the first extensive transcriptome analysis of this species using the Illumina HiSeq? 2000. Principal Findings A total of 41 and 39 million clean reads from the salt-treated (Se200S) and salt-free (SeCKS) tissues of S. europaea shoots were obtained, and de novo assembly produced 97,865 and 101,751 unigenes, respectively. Upon further assembly with EST data from both Se200S and SeCKS, 109,712 high-quality non-redundant unigenes were generated with a mean unigene size of 639 bp. Additionally, a total of 3,979 differentially expressed genes (DEGs) were detected between the Se200S and SeCKS libraries, with 348 unigenes solely expressed in Se200S and 460 unigenes solely expressed in SeCKS. Furthermore, we identified a large number of genes that are involved in ion homeostasis and osmotic adjustment, including cation transporters and proteins for the synthesis of low-molecular compounds. All unigenes were functionally annotated within the COG, GO and KEGG pathways, and 10 genes were validated by qRT-PCR. Conclusion Our data contains the extensive sequencing and gene-annotation analysis of S. europaea. This genetic knowledge will be very useful for future studies on the molecular adaptation to abiotic stress in euhalophytes and will facilitate the genetic manipulation of other economically important crops.
The Standard, Intervention Measures and Health Risk for High Water Iodine Areas

Abstract: Our study aims to clarify the population nutrient status in locations with different levels of iodine in the water in China; to choose effective measurements of water improvement(finding other drinking water source of iodine not excess) or non-iodised salt supply or combinations thereof; to classify the areas of elevated water iodine levels and the areas with endemic goiter; and to evaluate the risk factors of water iodine excess on pregnant women, lactating women and the overall population of women. From Henan, Hebei, Shandong and Shanxi province of China, for each of 50~99 μg/L, 100~149 μg/L, 150~299 μg/L, and ≥300 μg/L water iodine level, three villages were selected respectively. Students of 6–12 years old and pregnant were sampled from villages of each water-iodine level of each province, excluded iodized salt consumer. Then the children's goiter volume, the children and pregnant's urinary iodine and water iodine were tested. In addition, blood samples were collected from pregnant women, lactating women and other women of reproductive age for each water iodine level in the Shanxi Province for thyroid function tests. These indicators should be matched for each person. When the water iodine exceeds 100 μg/L; the iodine nutrient of children are iodine excessive, and are adequate or more than adequate for the pregnant women. It is reasonable to define elevated water iodine areas as locations where the water iodine levels exceed 100 μg/L. The supply of non-iodised salt alone cannot ensure adequate iodine nutrition of the residents, and water improvement must be adopted, as well. Iodine excess increases the risk of certain thyroid diseases in women from one- to eightfold.
Differential gene expression in incompatible interaction between wheat and stripe rust fungus revealed by cDNA-AFLP and comparison to compatible interaction

Abstract: A total of 52,992 transcript derived fragments (TDFs) were generated with 64 primer pairs and 2,437 (4.6%) of them displayed altered expression patterns after inoculation with 1,787 up-regulated and 650 down-regulated. We obtained reliable sequences (>100 bp) for 255 selected TDFs, of which 113 (44.3%) had putative functions identified. A large group (17.6%) of these genes shared high homology with genes involved in metabolism and photosynthesis; 13.8% to genes with functions related to disease defense and signal transduction; and those in the remaining groups (12.9%) to genes involved in transcription, transport processes, protein metabolism, and cell structure, respectively. Through comparing TDFs identified in the present study for incompatible interaction and those identified in the previous study for compatible interactions, 161 TDFs were shared by both interactions, 94 were expressed specifically in the incompatible interaction, of which the specificity of 43 selected transcripts were determined using quantitative real-time polymerase chain reaction (qRT-PCR). Based on the analyses of homology to genes known to play a role in defense, signal transduction and protein metabolism, 20 TDFs were chosen and their expression patterns revealed by the cDNA-AFLP technique were confirmed using the qRT-PCR analysis.We uncovered a number of new candidate genes possibly involved in the interactions of wheat and Pst, of which 11 TDFs expressed specifically in the incompatible interaction. Resistance to stripe rust in wheat cv. Suwon11 is executed after penetration has occurred. Abstract: We developed L3SN, a scalable, longevous, adaptive, and internet accessible wireless sensor network system for agriculture information monitoring, which is meticulously designed to meet the requirement of thousands hectares coverage, years of time monitoring and the adverse environment. The system architecture, the agriculture sensor device, the mesh protocol, and the web-based information processing platform are introduced. We also presented some implementation experience. The mesh protocol (LayerMesh) is highlighted, in which “stair scheduling” and “distributed dynamic load-balancing” are proposed to response the scalability, longevity and adaptivity requirements. We believe the design of L3SN is useful to many other large-scale, longevous applications such as hydrologic monitoring, geological monitoring etc.
CFD Application to Hydrogen Risk Analysis and PAR Qualification

Abstract: A three dimensional computation fluid dynamics (CFD) code, GASFLOW, is applied to analyze the hydrogen risk for Qinshan-II nuclear power plant (NPP). In this paper, the effect of spray modes on hydrogen risk in the containment during a large break loss of coolant accident (LBLOCA) is analyzed by selecting three different spray strategies, that is, without spray, with direct spray and with both direct and recirculation spray. A strong effect of spray modes on hydrogen distribution is observed. However, the efficiency of the passive auto-catalytic recombiners (PAR) is not substantially affected by spray modes. The hydrogen risk is significantly increased by the direct spray, while the recirculation spray has minor effect on it. In order to simulate more precisely the processes involved in the PAR operation, a new PAR model is developed using CFD approach. The validation shows that the results obtained by the model agree well with the experimental results.
Interferon-Gamma Enhance Immune Response and Efficacy of Recombinant Subunit Vaccine Against Eimeria tenella in Chickens

Abstract: To effectively control coccidiosis with a subunit vaccine, the refractile body antigen SO7 adjuvanted with Chicken Interferon-gamma (ChIFN-gamma) was administered to chickens and its ability to protect against virulent challenge by E. tenella was evaluated. About 5 days old chickens were subcutaneously immunized with SO7 antigen (100 μg per chick) mixed with various doses of ChIFN-gamma or without adjuvant. A boost of the same dose was given at 12 and 19 days of age. The specific antibody response and lymphocyte proliferation response to SO7 antigen were measured at 5, 12, 19 and 26 days of age. Chickens in all groups were orally challenged with 1x105 homologous sporulated oocysts of E. tenella at 26 days of age and killed at 8 days postchallenge. Results showed that when immunized by antigen adjuvanted with ChIFN-gamma, specific lymphocyte proliferation in chichens was obviously observed at 19 and 26 days of age and specific antibody response was obviously observed at 12, 19 and 26 days of age. All groups administered with antigen mixed in adjuvant showed higher protective immunity compared to PBS control, non-adjuvanted antigen control in which the group given adjuvanted antigen with 5,000 units ChIFN-gamma showed significantly stronger adjuvant effects in terms of lower mortality, oocyst output, lesion score and higher weight gain (p<0.05). Protective immunity was affected by dose of the adjuvant, no linear dose relationship was observed. These results suggest that ChIFN-gamma has a strong adjuvant effect at a appropriate dose and may potentially serve as a vaccine adjuvant against avian coccidiosis.
