Roles of the DYRK Kinase Pom2 in Cytokinesis, Mitochondrial Morphology, and Sporulation in Fission Yeast

Abstract: Pom2 is predicted to be a dual-specificity tyrosine-phosphorylation regulated kinase (DYRK) related to Pom1 in Schizosaccharomyces pombe. DYRKs share a kinase domain capable of catalyzing autophosphorylation on tyrosine and exogenous phosphorylation on serine/threonine residues. Here we show that Pom2 is functionally different from the well-characterized Pom1, although they share 55% identity in the kinase domain and the Pom2 kinase domain functionally complements that of Pom1. Pom2 localizes to mitochondria throughout the cell cycle and to the contractile ring during late stages of cytokinesis. Overexpression but not deletion of pom2 results in severe defects in cytokinesis, indicating that Pom2 might share an overlapping function with other proteins in regulating cytokinesis. Gain and loss of function analyses reveal that Pom2 is required for maintaining mitochondrial morphology independently of microtubules. Intriguingly, most meiotic pom2Δ cells form aberrant asci with meiotic and/or forespore membrane formation defects. Taken together, Pom2 is a novel DYRK kinase involved in regulating cytokinesis, mitochondrial morphology, meiosis, and sporulation in fission yeast.
Interaction between NBS1 and the mTOR/Rictor/SIN1 Complex through Specific Domains

Abstract: Nijmegen breakage syndrome (NBS) is a chromosomal-instability syndrome. The NBS gene product, NBS1 (p95 or nibrin), is a part of the Mre11-Rad50-NBS1 complex. SIN1 is a component of the mTOR/Rictor/SIN1 complex mediating the activation of Akt. Here we show that NBS1 interacted with mTOR, Rictor, and SIN1. The specific domains of mTOR, Rictor, or SIN1 interacted with the internal domain (a.a. 221-402) of NBS1. Sucrose density gradient showed that NBS1 was located in the same fractions as the mTOR/Rictor/SIN1 complex. Knockdown of NBS1 decreased the levels of phosphorylated Akt and its downstream targets. Ionizing radiation (IR) increased the NBS1 levels and activated Akt activity. These results demonstrate that NBS1 interacts with the mTOR/Rictor/SIN1 complex through the a.a. 221–402 domain and contributes to the activation of Akt activity.
Effect of Carbon and Nitrogen Sources on the Accumulation of Poly-β-Hydroxybutyrate by Purple Non-Sulfur Photosynthetic Bacteria

Abstract: The effect of carbon and nitrogen sources on the accumulation of Poly-beta-Hydroxybutyrate (PHB) by purple non-sulfur photosynthetic bacteria (PNSB)was investigated. The results showed that ammonia nitrogen and organic nitrogen could benefit the PNSB accumulating PHB; under certain ratio of carbon and nitrogen low carbon concentration can accumulate more PHB, but high carbon concentration is better for PNSB accumulating PHB with the growth of PNSB. As to different substrates, butyrate is the best for accumulating of PHB, but acetate is better and propionate is the poorest. Mixed substrate with butyrate as the main content are better than single substrate; carbon dioxide have certain effect on the PNSB accumulating PHB, and high carbon dioxide concentration can improve PHB content when using acetate or butyrate as substrate.

Abstract: 选用来自国内外的白菜黑斑病菌三个种Alternaria brassicicola,A. brassicae和A. japonica为研究材料,以生长速率为评价指标,对其适宜生长温度、pH值、光照、培养基和菌落特征等基本培养条件和性状进行了对比研究。结果表明,三个种的病菌最适生长温度均在20℃~25℃,变化幅度较小,但A. brassicicola 和A. japonica菌丝体生长温度范围和已有文献报道相比提高了5℃;A. brassicae中国菌株菌丝体生长的pH值范围为4~8,其它所有供试菌株生长的pH值为3~11;24h光照、12h光暗交替和24h黑暗对国内外A. brassicicola菌株生长速率影响均不大,而A. brassicae在24h光照和12h光暗交替下的生长速率比24h黑暗处理的显著增大,24h光照和24h黑暗比12h光暗交替更适合A. japonica生长;PDA是白菜黑斑病菌营养生长的最适培养基,SNA除了适合其生长外,还对供试三个种的病菌生长有选择性;菌落直径及颜色可以作为区分三个种的参考指标。供试的白菜黑斑病菌三个种在基本培养条件方面存在种水平上的差异。
Genetic diagnosis strategy of hereditary non-polyposis colorectal cancer

Abstract: AIM: To study the characteristics of mismatch repair gene mutation of Chinese hereditary non-polyposis colorectal cancer (HNPCC) and hMLH1 gene promoter methylation, and to improve the screening strategy and explore the pertinent test methods.METHODS: A systematic analysis of 30 probands from HNPCC families in the north of China was performed by immunohistochemistry, microsatellite instability (MSI), gene mutation and methylation detection.RESULTS: High frequency microsatellite instability occurred in 25 probands (83.3%) of HNPCC family. Loss of hMLH1 and hMSH2 protein expression accounted for 88% of all microsatellite instability. Pathogenic mutation occurred in 14 samples and 3 novel mutational sites were discovered. Deletion of exons 1-6, 1-7 and 8 of hMSH2 was detected in 3 samples and no large fragment deletion was found in hMLH1. Of the 30 probands, hMLH1 gene promoter methylation occurred in 3 probands. The rate of gene micromutation detection combined with large fragment deletion detection was 46.7%-56.7%. The rate of the two methods in combination with methylation detection was 63.3%.CONCLUSION: Scientific and rational detection strategy can improve the detection rate of HNPCC. Based on traditional molecular genetics and combined with epigenetics, multiple detection methods can accurately diagnose HNPCC.

Bit Confidence-Based Soft Decision Metric Generation for QAM

Abstract: In modern communication systems, a channel decoder usually employs the soft input to guarantee a high channel decoding gain. A new soft decision metric for QAM is presented in this paper. Unlike traditional soft decision metric based on minimum Euclidean distance, the proposed metric is based on the bit confidences. Compared to traditional soft decision metric, the one presented in this paper can guarantee the channel decoder performance with less computation complexity. The numerical results applying the proposed method to the DTMB system indicate that the proposed soft decision metric reduces the computation complexity without any performance degradation. These results also show that the proposed metric is a good alternative for soft decision generations.

Fuzzy image registration algorithms of sub-pixel level

Abstract: On account of the fact that today's point feature registration algorithms can't match edge fuzzy images effectively, this paper proposed the extreme gray offset feature. Firstly, defined the feature point as the pixel with the most dramatic offset from the local mean value. And then, located the extreme offset feature in sub-pixel level by derived function, while fitted the function curve by the least square. Finally, realized the registration by comparing the distance among those 32D feature vectors built by NMI. This feature was easy to extract and calculate, the results of experiment show that the algorithm has high matching accuracy applied in the fuzzy image registration.
