Quenching of fluorescence in membrane protein by hypocrellin B

Abstract: The hypocrellin B (HB) was used as a fluorescence quencher to study the basic physical charactcristics of HB in membrane systems, including the diffusion speed of quencher from aqueous phase into membrane phase, the partition coefficient (P) of quenchtr between membrane and water, and the fluorescence quenching constant of protein (Ksv; Kq,). The experimental results show that the quenching of fluorescence in membrane protein by HB can be determined by the principle of dynamic quenching. The experimental process of fluorescence quenching was observed in detail by using the ESR technique. The signal of HB- was found to arise from an electron transfer from excited trytophan to HB.

Abstract: 125 μm-breath sensor with high sensitivity and rapid response was prepared by using n-type Si: Au material. Its sensitivity coefficient and time constant were 4 V.sec / L and 38 msec, respec-tively. Its working principle was based on ano- malous resistance effect, which not only increa- sed the sensitivity, but also reduced its time con-stant greatly. Its signal processing system can select the breath signals and work stably. Therefore, the small changes of breath system can be measured and, especially, patient’s breath rate can be monitored at a distance.

Abstract: The purified thermophilic bacterium PS3 F1β10 ×His-tag is inserted into the F0F1-ATP synthases of chromatophores isolated from photosynthetic bacteria Rhodospirillum rubrum. The studies of biochemical properties of the hybrid chromatophores show that they have both protonsdriving capability and photophosphorylation. The fluorescent actin filaments, as a marker of its orientation by videomicroscopic experiment, are connected via Maleimido-C3-NTA to the reconstituted β10 ×Histag of F0F1ATP synthases. The clockwise rotation of F0F1ATP synthases driven by light is observed directly when viewed from the F0 side to F1. This system should be valuable for further studying the coupling property of F0F1ATP synthase.

Abstract: The fluorescence of gramicidin A(GA) in lipid bilayer may be quenched by Hypocrellin B. When the phospholipids undergo a phase transition from gel to liquid crystalline phase, the reaction rate decreases unusually. This phenomenon has been analyzed by Marcus’ photoinduced electron theory, and an electron transfer inverted region hypothesis is proposed, which may adequately explain this abnormal phenomenon.

Abstract: 以离体培养的HeLa细胞为材料,用激光共聚焦显微镜,多动能图像分析仪,扫描电镜,透射电镜,荧光分光光度计等研究了一种新型的竹红菌乙素修饰物(5—Br—HB)被HeLa细胞摄取的时间进程、药物在细胞内的显微定位以及对细胞形态结构的光动力损伤。结果表明:HeLa细胞对5—Br—HB摄取在1小时之内细胞内药物浓度随着药液培养时间的增加呈线性增长,3小时后摄取基本达到饱和。HeLa细胞在含10μmol/L5—Br一HB培养基中37℃温育半小时后,敏化剂主要分布在细胞膜和胞浆中。受光动力损伤的细胞膜丧失连续结构,绒毛丧失,细胞表面出现异形突起。加药并光照5分钟组细胞浆内产生大量空泡,线粒体、内质网等细胞器丧失结构完整性,在较高浓度下甚至出现明显的细胞膜破裂和核膜损伤。光动力敏化对核形态产生明显的损伤作用,表现为N/C和NA减小,NFF增大。

Abstract: The fluorescence photobleaching recovery technique was used to examine the effect of HB on the lateral diffusion and fluorescence recovery rate in the Ascitic Hepatoma(AH) cell membrane and solutions. The results showed that the lateral diffusion D HB in membrane is 3.2× 10-9cm2s-1 and fluorescence recovery rate is near 97.8%; The experiments illustrated that in AH cell membrane HB is in free state.

Abstract: Chlorinated organic residual liquid is produced from the distillation process of new refrigerants production. It is difficult to be treated by traditional water treatment process and incineration process. In this study, a carbonization process at atmospheric pressure was used to convert this residual liquid to carbonaceous product and organic gas in 2 h at 230℃ or 260℃. The carbonaceous product was characterized by scanning electron microscope, Fourier-transform infrared spectrometer and thermo gravimetric analysis. The element composition and the high heat value of these products were similar to anthracite and lignite, respectively, showing that they could be used as alternative fuels. The components of organic gas were analyzed using gas chromatography-mass spectrometry and the gas had potential for incineration.

Abstract: In this paper, we primarily studied the HB-sensitized cytotoxicity to murine ascitic hepatoma cell (AH). The results showed that the cell mortality was roughly proportional to the square root of HB concentration. The ability to kill AH cell among hypocrellin A (HA) , HB, protoporphyrin (PP) , and he-motoporphyrin (HP) are in the following order: PP>HB>HA>HP. The ultr-astructure observation using TEM showed that the plasma membranes, mit-ochodria and endoplasmic recticulus were probably the main targets of photose-nsitization. After 10 minutes irradiation in the presence of HB, obvious ves-iculation and dramatic rupture of plasmic membranes were observed, but the nucleic membranes and nuclei did not change significantly. SEM revealed that the shape of the photodamaged cells changed obviously and the microvilli on the cells disappeared. By using active oxygen quenchers, spin trapping and spin silence techniques, we proved that singlet oxygens, superoxide anion radicals and hydroxyl radicals were involved in the process of HB-photosensitized cytotoxi-city.

Abstract: Second order elliptic equation is a class of mathematical model for scientific computing, such as convex-diffusion, oil-reservoir simulation, etc. Based on intrinsic symmetrizable property, a new concept on positively symmetrizable matrix is proposed in this paper. We point that for such kind of equation systems, it is possible to adopt special preconditioning CG algorithm, e.g. 1]-3], instead of the usual iteration procedure for general non-symmetry systems, such as GMRES 3]-4] ) BiCGSTAB 5]. Numerical tests show the new algorithm is effective for solving this kind of second order elliptic discrete systems.
