Fructus Gardenia Extract Ameliorates Oxonate-Induced Hyperuricemia with Renal Dysfunction in Mice by Regulating Organic Ion Transporters and mOIT3

Abstract: The potent anti-hyperuricemia activities of Fructus Gardenia Extract (FGE) have been well reported. The aim of this study was to evaluate the uricosuric and nephro-protective effects of FGE and explore its possible mechanisms of action in oxonate-induced hyperuricemic mice. FGE was orally administered to hyperuricemic and normal mice for 1 week. Serum and urinary levels of uric acid, creatinine and blood urea nitrogen (BUN), and fractional excretion of uric acid (FEUA) were measured. The mRNA and protein levels of mouse urate transporter 1 (mURAT1), glucose transporter 9 (mGLUT9), ATP-binding cassette, subfamily G, 2 (mABCG2), organic anion transporter 1 (mOAT1), mOAT3, oncoprotein induced transcript 3 (mOIT3), organic cation/carnitine transporters in the kidney were analyzed. Simultaneously, Tamm-Horsfall glycoprotein (THP) levels in urine and kidney were detected. FGE significantly reduced serum urate levels and increased urinary urate levels and FEUA in hyperuricemic mice. It could also effectively reverse oxonate-induced alterations in renal mURAT1, mGLUT9, mOAT1 and mOIT3 expressions, as well as THP levels, resulting in the enhancement of renal uric acid excretion. Moreover, FGE decreased serum creatinine and BUN levels, and up-regulated expression of organic cation/carnitine transporters, improving renal dysfunction in this model. Furthermore, FGE decreased renal mABCG2 expressions in hyperuricemic mice, contributing to its beneficial actions. However, further investigation is needed in clinical trials of FGE and its bioactive components.
Improvement of Amperometric Sensor Used for Determination of Nitrate with Polypyrrole Nanowires Modified Electrode

Abstract: Polypyrrole(PPy) nanowire modified electrodes were developed by template-freeelectrochemical method based on graphite electrode. The modified electrode wascharacterized by their amperometric response towards nitrate ions. Before reduction ofnitrate ions, electrochemical solid-phase extraction (EC-SPE) of nitrate in/on modifiedelectrodes was conducted. It is found that the unusual nanowired structure of polypyrrolelayer (instead of well known cauliflower structure) allows us to increase the effectivesurface area of the electrode and subsequently the sensitivity. And the effects ofelectrochemical preparation parameters of PPy nanowire modified electrodes on theircorresponding characters were evaluated. The experimental results show that theelectrochemical preparation parameters of the modified electrodes such as scan rate,polymerization potential, temperature of polymerization solution and polymerization timehave significantly effects on the morphology of PPy nanowires and subsequently effectivesurface area of the electrode and electroreduction current density of nitrate. Thedetermination sensitivity may be varied according to the modification parameters. Under acertain polymerization conditions, the corresponding sensitivity reaches 336.28 mA/M cm2 and the detection limit is 1.52×10-6 M. The proposed method was successfully applied in thedetection of nitrate in the real samples.

Microstructure of Ni–Al powder and Ni–Al composite coatings prepared by twin-wire arc spraying

Abstract: Ni–Al powder and Ni–Al composite coatings were fabricated by twin-wire arc spraying (TWAS). The microstructures of Ni-5wt%Al powder and Ni-20wt%Al powder were characterized by scanning electronic microscopy (SEM) and energy dispersive spectroscopy (EDS). The results showed that the obtained particle size ranged from 5 to 50 μm. The morphology of the Ni–Al powder showed that molten particles were composed of Ni solid solution, NiAl, Ni3Al, Al2O3, and NiO. The Ni–Al phase and a small amount of Al2O3 particles changed the composition of the coating. The microstructures of the twin-wire-arc-sprayed Ni–Al composite coatings were characterized by SEM, EDS, X-ray diffraction (XRD), and transmission electron microscopy (TEM). The results showed that the main phase of the Ni-5wt%Al coating consisted of Ni solid solution and NiAl in addition to a small amount of Al2O3. The main phase of the Ni-20wt%Al coating mainly consisted of Ni solid solution, NiAl, and Ni3Al in addition to a small amount of Al and Al2O3, and NiAl and Ni3Al intermetallic compounds effectively further improved the final wear property of the coatings. TEM analysis indicated that fine spherical NiAl3 precipitates and a Ni–Al–O amorphous phase formed in the matrix of the Ni solid solution in the original state.

Abstract: In this paper, a new illation of the principle of PWP (Pressure Wave Propagation) method is presented, and the results, after comparing the PWP method with the PEA (Pulsed Electro-Acoustic) method, indicate that the two methods very resemble with each other in physics. In order to testify the conclusion, the measurement of space charge distribution in XLPE by using the combined system of piezo-PWP/PEA has been corried out, and the experimental results confirm the conclusion.

China is located in the eastern Eurasian continent and the brink of the world's largest ocean - the Pacific Ocean. It has more than 20,000 kilometers border, 18,000 kilometers coastline and 5,000 offshore islands. The strong climatic differences between land and sea, as well as vast land area and the complex terrain form abundant wind energy resources in China. Evaluation of Dermal Substitute in a Novel Co-Transplantation Model with Autologous Epidermal Sheet

Abstract: The development of more and more new dermal substitutes requires a reliable and effective animal model to evaluate their safety and efficacy. In this study we constructed a novel animal model using co-transplantation of autologous epidermal sheets with dermal substitutes to repair full-thickness skin defects. Autologous epidermal sheets were obtained by digesting the basement membrane (BM) and dermal components from rat split-thickness skins in Dispase II solution (1.2 u/ml) at 4°C for 8, 10 and 12 h. H&E, immunohistochemical and live/dead staining showed that the epidermal sheet preserved an intact epidermis without any BM or dermal components, and a high percentage of viable cells (92.10±4.19%) and P63 positive cells (67.43±4.21%) under an optimized condition. Porcine acellular dermal matrixes were co-transplanted with the autologous epidermal sheets to repair full-thickness skin defects in Sprague-Dawley rats. The epidermal sheets survived and completely re-covered the wounds within 3 weeks. Histological staining showed that the newly formed stratified epidermis attached directly onto the dermal matrix. Inflammatory cell infiltration and vascularization of the dermal matrix were not significantly different from those in the subcutaneous implantation model. Collagen IV and laminin distributed continuously at the epidermis and dermal matrix junction 4 weeks after transplantation. Transmission electron microscopy further confirmed the presence of continuous lamina densa and hemidesmosome structures. This novel animal model can be used not only to observe the biocompatibility of dermal substitutes, but also to evaluate their effects on new epidermis and BM formation. Therefore, it is a simple and reliable model for evaluating the safety and efficacy of dermal substitutes.
The Ryukyu Current east of Amami-Ohshima during 2004 to 2006

Abstract: An inverse calculation using twelve repeat hydrographic section data collected from 2004 to 2006 yields velocity structures and volume transports (VTs) of the Ryukyu Current (RC) in the region east of northern Ryukyu Islands. The results show that the RC is dominated by subsurface velocity core with maximum velocities varying from 15.1 to 80.0 cm·s-1. The positions of subsurface maximum velocity core distribute between 110 and 600 m and 27.2°~28.2° N along the AE line. The mean velocity section exhibits a perfect subsurface velocity core with a mean maximum velocity of 21.3 cm·s-1, and vertical and horizontal dimensions of 800 m and 30 km, respectively. The seasonal mean velocity sections exhibit that the RC is the strongest in autumn and the weakest in summer. The mean VT for winter, spring, summer, autumn, and all twelve observations are 10.9, 10.1, 5.9, 23.9, and 12.7×106 m3·s-1, respectively. The mean net VT of the Kuroshio south of Japan is estimated to be 52.7×106 m3·s-1. The rate of Kuroshio VT supplied by the Kuroshio from Tokara Strait and by the RC from the region southeast of Amami-Ohshima is 0.40.
Fuzzy Petri net and its application in Web services trust evaluation

Abstract: In order to improve the accuracy of trust evaluation and the success rate of interaction of Web services,this paper proposed a new Web services trust evaluation model WS-TEM based on the fuzzy Petri net. This model gave the fuzzy trust rules of evaluation, based on it, proposed a reasoning algorithm based on the fuzzy Petri net. This model used the knowledge of fuzzy Petri net to represent and run basic algorithm which applied to comprehensive evaluation trust relation of Web services entities by parallel processing of more than one recommended trust. The result of simulative experiment and analysis show that this model on the trust evaluation of Web services becomes more accurate and reliable, consequently the interaction risk is reduced and ensured the success rate of interaction.
Resonant tunneling through double-barrier structures on graphene

Abstract: Quantum resonant tunneling behaviors of double-barrier structures on graphene are investigated under the tight-binding approximation. The Klein tunneling and resonant tunneling are demonstrated for the quasiparticles with energy close to the Dirac points. The Klein tunneling vanishes by increasing the height of the potential barriers to more than 300 meV. The Dirac transport properties continuously change to the Schrodinger ones. It is found that the peaks of resonant tunneling approximate to the eigen-levels of graphene nanoribbons under appropriate boundary conditions. A comparison between the zigzag- and armchair-edge barriers is given.
