Purificación de antígeno de Candida albicans para pruebas cutáneas: estudio preliminar

Abstract: the assessment of cellular immunity by means of the response produced by t-lymphocytes stimulated by the presence of a determined antigen, is of great interest in the treatment of several infectious diseases during the course of burns, lesions, traumas, etc., and due to this it is very important to obtain purified antigens for their usage in the skin tests used in such assessment. the authors developed a purification method to obtain one of these antigens, from candida albicans. they carried out modifications on the method described by buckley et al., and found that the methodology was adequate and reproducible, since the antigen obtained had similar characteristics to those of the reference antigen.

Abstract: La evaluación de la inmunidad celular mediante la respuesta producida por los linfocitos T, que son estimulados por la presencia de un antígeno determinado, es de sumo interés para el tratamiento de diversas enfermedades infecciosas que se producen durante el transcurso de quemaduras, lesiones, traumatismos, etcétera, por lo que la obtención de antígenos purificados para su uso en pruebas cutáneas que se emplean para dicha evaluación es de gran importancia. En nuestro trabajo nos dimos a la tarea de desarrollar un método de purificación para la obtención de uno de estos antígenos, a partir de la Candida albicans. El método utilizado fue el descrito por Buckley et al., al cual se le realizaron modificaciones. Se encontró que la metodología era adecuada y reproducible, pues el antígeno obtenido tenía características similares al antígeno de referencia. Abstract: avian antibodies (igy) display some advantages in relation to the mammalian igg antibodies due to its easy procedure for generation and purification and besides for its lower production cost. the objective of this work was to analyze the feasibility of binding igy antibodies to polystyrene particles and to evaluate its performance in latex-agglutination tests in slide. for this purpose, the detection of hepatitis b surface antigen (hbsag) was used as a model. leghorn hens were immunized with the active pharmaceutical ingredient (mpa) used in the manufacturing of the cuban recombinant vaccine heberbiovac hb . the obtained antibodies were purified and used in the preparation of the latex particles (0.8 μm) for the detection of the hbsag by agglutination. the performance of the test was evaluated with 50 serum samples against similar commercial reagent. the results obtained were satisfactory and demonstrated the feasibility of binding such avian antibodies to latex particles. these results open a perspective to the use of this type of assay for the rapid diagnostic of many diseases.

Abstract: The National Database of Nursing Quality IndicatorsTM (NDNQI ) is the only national nursing database that provides quarterly and annual reporting of structure, process, and outcome indicators to evaluate nursing care at the unit level. Linkages between nurse staffing levels and patient outcomes have already been demonstrated through the use of this database. Currently over 1100 facilities in the United States contribute to this growing database which can now be used to show the economic implications of various levels of nurse staffing. The purpose of this article is to describe the work and accomplishments related to the NDNQI as researchers utilize its nursing-sensitive outcomes measures to demonstrate the value of nurses in promoting quality patient care. After reviewing the history of evaluating nursing care quality, this article will explain the purpose of the NDNQI and describe how the database has been operationalized. Accomplishments and future plans of the NDNQI will also be discussed.

Reacción de inmunoglobulinas de distintas especies con el factor reumatoideo en inmunoensayos de látex aglutinación

Abstract: the rheumatoid factor is considered as the fundamental responsible for the interferences (false +) produced in the immunoassays, since it reacts unspecifically with different epitopes of the molecules of immunoglobulin g (igg). this paper was aimed at studying the igg from 7 species (human, rabbit, donkey, lamb, horse, hen and mouse; and of the latter the subclasses igg1, igg2a, igg2b). particles of polystyrene of 0.8 m that were covered with 3 concentrations (0.02, 0.01. 0.05 mg/m2) of every 1 g were used. reactivity was evaluated against 6 concentrations of rheumatoid factor (1 000, 500, 250, 64, 16, 8 ui). in all cases, it was demonstrated that the reactivity decreases as the covering concentration diminishes and that the decreasing reactivity order was: human, rabbit, donkey, lamb, and horse; whereas for the murine igg it was igg1, igg2a and igg2b. the hen igg did not react against the rheumatoid factor. it was the ideal species to make latex reagents to avoid the interferences of the rheumatoid factor.

Abstract: El factor reumatoideo es considerado el responsable fundamental de las interferencias (falsos +) producidas en los inmunoensayos, ya que reacciona inespecíficamente con diferentes epitopos de las moléculas de inmunoglobulina G (IgG). El objetivo de este trabajo fue estudiar la IgG de 7 especies (humana, conejo, burro, carnero, caballo, gallina y ratón, de esta última las subclases IgG1,IgG2a,IgG2b). Se utilizaron partículas de poliestireno de 0,8 μ que fueron recubiertas con 3 concentraciones (0,02, 0,01, 0,05 mg/m2 ) de cada Ig. La reactividad se evaluó frente a 6 concentraciones de factor reumatoideo (1 000, 500, 250, 64, 16, 8 UI ). Se evidenció en todos los casos que la reactividad disminuye a medida que baja la concentración de recubrimiento y que el orden de reactividad decreciente fue: humana, conejo, burro, carnero y caballo, y para las IgG murinas fue IgG1, IgG2a e IgG2b. La IgG de gallina no reaccionó frente al factor reumatoideo, la cual constituyó la especie ideal en la elaboración de reactivos de látex para evitar las interferencias del factor reumatoideo. Abstract: sickle cell disease is associated with glomerular and tabular structural abnormalities, hemodynamic changes and renal hormone synthesis alterations (erythropoietin, renin, prostaglandins). these changes appear in childhood with consequences including chronic anemia, increased blood flow and vaso-occlusion, especially within the renal medulla. in ss disease, the glomerular filtration rate (gfr) is markedly higher in young patients but this drops with age. as a consequence of abnormalities in the distal nephron function there is reduced capacity of urine acidification, potassium excretion and hyposthenuria. among the clinical effects of the latter are polyuria, nocturia, enuresis and a susceptability for dehydration. abnormalities of the proximal tubular function include increased reabsorption of phosphates and b2 microglobulin and incresed secretion of uric acid and creatinine. for this reason, creatinine clearance significantly overestimates the glomerular filtration rate making creatinine learance inappropriate to detect early renal function impairment. proteinuria occurs in 29-50% of over 10-years-old ss patients with 2/3 progressing to chronic renal failure. the use of angiotensin-converting enzyme inhibitor proves to be successful to reduce proteinuria which may have an impact of the progression of renal failure. microalbuminuria is a sensitive marker of sickle nephropathy that precedes proteinuria. in pediatric population a prevalence of 19% of microbuminuria was observed; a percentage that increases with age and has a positive association with lower hemoglobin levels, higher leucocyte counts and acute chest syndrome. screening for microalbuminuria is recommended from the age of 10 years old. a multicentric trial is ongoing with the aim of demonstrate potential benefits of early hydroxyurea therapy in the prevention of chronic organic lesions.
