Linear Thanatin Is an Effective Antimicrobial Peptide against Colistin-Resistant Escherichia coli in Vitro  [PDF]

Abstract: Colistin has been regarded as the last line antibiotic for treatment of infections caused by multidrug resistant gram-negative bacteria. Therefore, the increasing emergence of colistin resistance among gram-negative bacteria represents a serious problem. The objective of this study was to characterize the effectiveness of the chemically synthesized thanatin in linear form against colistin-resistant E. coli isolated from a pig farm in China. Agar diffusion assay and broth microdilution test were employed to analyze the susceptibility of colistin-sensitive E. coli (ATCC25922) and colistin-resistant E. coli (SHP45) to linear thanatin (L-thanatin). Combinatory effect of linear thanatin and colistin against E. coli was also determined by fractional inhibition concentration index (FICI) analysis. The results showed that L-thanatin at a concentration of 1 mg/ml produced larger inhibition zone on agar against ATCC25922 than SHP45. In the quantitative microdilution test, L-thanatin had the same MIC of 3.2 μg/ml for ATCC25922 and SHP45. Based on the FICI analysis, additive effect was obtained with 1.56 μg/ml of L-thanatin and 0.125 μg/ml of colistin for ATCC25922; but with 1.56 μg/ml of L-thanatin and 0.25 μg/ml of colistin or with 2 μg/ml of colistin and 0.39 μg/ml of L-thanatin for SHP45. These data proved that L-thanatin is an effective antimicrobial peptide against colistin-resistant E. coli.

The proportion of female leading cadres in China is also growing year by year; however, as far as the development of female leading cadres is concerned, women are still far behind men in terms of quantity and behavioral influence. The long-term status inequality has inhibited women’s instinct and desire to tap their own potential and the sense of social participation. The career development and life balance of female leaders under multiple role conflicts has become the confusion of most female leaders. Therefore, mastering the balance of female leadership role conflicts is of great significance for improving women’s leadership and creating a happy life.
Efficacy and Safety of rh-Endostatin Combined with Chemotherapy versus Chemotherapy Alone for Advanced NSCLC: A Meta-analysis Review

Abstract: Background and objective In recent years, there has been a large number of studies and reports about the efficacy and safety of recombinant human endostatin (rh-endostatin), an anti-angiogenic drug, in treatment of advanced lung cancer. Authentic assessment of rh-endostatin treatment in lung cancer is important. The aim of this study is to assess the clinical efficacy and safety of rh-endostatin combined with chemotherapy in the treatment of patients with non-small cell lung cancer (NSCLC). Methods Cochrane systematic review methods were used in the data selection, and data were selected from the Cochrane Library, EMBASE, Medline, SCI, CBM, CNKI, and etc electronic database to get all clinical controlled trials. The retrieval time was March 2010. The objects of these randomized controlled trials were advanced NSCLC patients and in the experimental group was rh-endostatin combination chemotherapy, in the control group was chemotherapy alone to compare the efficacy of two groups. The quality of included trials were evaluated by two reviewers independently. The software RevMan 5.0 was used for meta-analyses. Results Fifteen trials with 1,326 patients were included according to the including criterion. All trials were randomized controlled trials, and two trials were adequate in reporting randomization. Thirteen trials didn’t mention the blinding methods. Meta analysis indicated that the NPE arm (Vinorelbine+cisplatin+rh-endostatin) had a different response rate compared with NP (Vinorelbine+cisplatin) arm (OR=2.16, 95%CI: 1.57-2.99). The incidences of severe leukopenia (OR=0.94, 95%CI: 0.66-1.32) and severe thrombocytopenia (OR=1.00, 95%CI: 0.64-1.57) and nausea and vomiting (OR=0.85, 95%CI: 0.61-1.20) were similar in the NPE arm compared with those in the NP arm. The NPE plus radiotherapy (RT) arm had a similar response rate compared with NP plus RT arm (OR=2.39, 95%CI: 0.99-5.79). The incidences of leukopenia (OR=0.83, 95%CI: 0.35-1.94) and thrombocytopenia (OR=0.78, 95%CI: 0.19-3.16) and radiation esophagitis (OR=1.00, 95%CI: 0.40-2.49) were similar in the NPE plus RT arm compared with those in the NP plus RT arm. Conclusion In the treatment of advanced NSCLC, rh-endostatin in combination with platinum-based chemotherapy improve the response rate without obviously raised side effects, however, when radiotherapy are added to NPE arm or NP arm, the response rates have a similar outcome. Owing to the small sample size and poor quality of included trials, more well-designed double-blinded randomized controlled trials should be performed.
Co-Sensitization to Silkworm Moth (Bombyx mori) and 9 Inhalant Allergens among Allergic Patients in Guangzhou, Southern China

Abstract: Objectives This study aimed to investigate the profile of sensitization to silkworm moth (Bombyx mori) and other 9 common inhalant allergens among patients with allergic diseases in southern China. Methods A total of 175 patients were tested for serum sIgE against silkworm moth in addition to combinations of other allergens: Dermatophagoides pteronyssinus, Dermatophagoides farinae, Blomia tropicalis, Blattella germanica, Periplaneta americana, cat dander, dog dander, Aspergillus fumigatus and Artemisia vulgaris by using the ImmunoCAP system. Correlation between sensitization to silkworm moth and to the other allergens was analyzed. Results Of the 175 serum samples tested, 86 (49.14%) were positive for silkworm moth sIgE. With high concordance rates, these silkworm moth sensitized patients were concomitantly sensitized to Dermatophagoides pteronyssinus (94.34%), Dermatophagoides farinae (86.57%), Blomia tropicalis (93.33%), Blattella germanica (96.08%), and Periplaneta americana (79.41%). Moreover, there was a correlation in serum sIgE level between silkworm moth and Dermatophagoides pteronyssinus (r = 0.518), Dermatophagoides farinae (r = 0.702), Blomia tropicalis (r = 0.701), Blattella germanica (r = 0.878), and Periplaneta americana (r = 0.531) among patients co-sensitized to silkworm moth and each of these five allergens. Conclusion In southern Chinese patients with allergic diseases, we showed a high prevalence of sensitization to silkworm moth, and a co-sensitization between silkworm moth and other five common inhalant allergens. Further serum inhibition studies are warranted to verify whether cross-reactivity exists among these allergens.

Abstract:

This paper primarily calculates the similar coefficients of the industrial structure in Changchun and Jilin City, the results show that the similarity of the three industries in the two cities is high, but the similarity of the subdivided manufacture industries is low. We also find evidence that the conditions for the industrial integration of the two cities are improved. The paper also explores the reasons for the current situation of the industrial integration in Changchun and Jilin city, then propose the measures to develop the industrial integration.

Abstract: This paper tests the popular continuous-time interest rate models for Chinese repo market to address what and how the interest rates change with the marketlization in China. Using Bandi [1]’s method, we get the functional nonparametric estimation of drift and diffusion terms and the local time of the process. We find that the interest rates of China during the period from 1993 to 2003 are bimodal distributed and propose a two-regime model which can fit the data better. We also study the probabilities that the process will stay the two regimes respectively and its transition probability that the process transfers from one regime to another regime.

Industrial tourism, as one of the forms which curious tourists are fond of, has been rapidly developed throughout the country. Cities are the main carriers of industrialization, also are the important places for Industrial tourism. This paper selects Shanghai and Huangshi as sample cities for research, comparing the two cities and showing the difference between the development stages, development types and development models of industrial tourism in the two cities.

Abstract: In the present study, using specific antibody against D1 protein, we detected four aggregates of D1 protein in thylakoid membranes from spinach leaves illuminated at high light (800–2500 μmol photons·m 2·s 1) for 3 h. Their accumulations were dependent on the light intensity to which the leaves had been subjected. Further immunoblot analysis indicated that 70 kD aggregate was a product of D1 protein cross-linked with CP43, 65 and 60 kD aggregate were two cross-linked products between D1 and D2 proteins, and 41 kD aggregate was one cross-linked D1 with α-subunit of cytochrome b 559 (Cyt b 559). This result provided the evidence for the existence of the aggregation of the D1 protein in vivo. The maximal level of D1/Cyt b 559 aggregate occurred at 1000 μmol photons·m 2·s 1 but drastically decreased with further increasing light intensity. Immunoblot analysis with phosphothreonine (Thr (P)) antibody indicated that D1/CP43 and D1/Cyt b 559 aggregates contained the phosphorylated protein(s). In vitro dephosphorylation experiment also showed that D1/Cyt b 559 and D1/CP43 aggregates lost the immunoreactivity with Thr (P) antibody after the phosphatase treatment of the membranes from high-light-illuminated leaves. Our results demonstrated that strong illumination of spinach leaves induced cross-linked products of D1 protein with its nearby polypeptides of PS, some of which co.n-tained the phosphorylated D1 protein.

Abstract: The specific interaction between angiogenin and aptamer has been investigated by using AFM. The specificity of the interaction is revealed by comparing the binding probability of aptamer to other elements in a series of control experiments. The results have shown that there is specific interaction force between angiogenin and aptamer. Moreover, the single molecular pull-off force between angiogenin and aptamer has also been determined using the Poisson statistical method to be 133.7±11.7 pN. These findings obtained are helpful to the better revelation of recognition mechanism between angiogenin and aptamer, which provided basis for further understanding the inhibition of the aptamer to angiogenic activity.
