Paclitaxel Blocks Immunologic Escape through Up-regulating TAP-1, TAP-2 and Eliminatiing Treg Cells in 3LL-bearing Mice

Abstract: Background and objective Until now, there are still some questions that chemotherapy could block cancer immunologic escape or not. The aim of this study is to explore the mechanism of paclitaxel in the blocking of immunologic escape in 3LL bearing mice. Methods MHC class I components were detected by FACS antibodies TAP-1 and TAP-2 in 3LL group and 3LL pretreated with 5×10(-8) mmol/L paclitaxel. C57BL/6 mice were divided into 3 groups: tumor –bearing group, paclitaxel-treating group and tumor-free group. 3LL cells (0.3×10(6)) were injected via tail vein of mice. On day 8, paclitaxel (0.012 5 mg) was injected subcutaneously in the treatment group. On day 21, murine lung cells were suspended. With FACS triple staining, we analyzed expression of Treg in each group. Results The expression of TAP-1 (5.68%±0.65%), TAP-2 (89.54%±4.8%) was remarkably higher in paclitaxel-pretreating 3LL group than that in 3LL bearing mice group with TAP-1 (1.93%±0.25%), TAP-2 (67.78%±5.08%), P=0.006, P=0.036 respectively. The expression of regulatory T cells (Treg) in tumor bearing mice was significantly higher (25.46%±2.23%) than that of in normal mice (12.46%±1.21%) (P < 0.001). Expression of Treg in paclitaxel treated mice was remarkably inhibited (17.53%±1.24%) compared with that in 3LL bearing mice group (P=0.004). Conclusion Paclitaxel may block immunologic escape in the treatment of 3LL bearing mice partly through up-regulation TAP-1, TAP-2 and eliminatiing Treg cells.
Sex Differences in Scratching Behaviors Induced by Intradermal Injections of Pruritogenic Chemicals in C57BL/6 Mice  [PDF]

Abstract: Pruritus is an individual unpleasant sensation of human sensory nervous system. In the physiological condition it excerts a self-protective mechanism to protect the skin against external harmful agents. Pruritoceptive itch is also a major symptom of skin disease and a common reason for consulting a dermatologist in clinic. It has been well known that both histamine-dependent and histamine-independent pathways mediate acute and chronic itch sensations. Previous studies have showed common neural pathways partially shared by itch and pain sensation, and significant sex differences in pain sensation. However, sex difference in itch sensation has not been given too much attention as the majority of itch studies were done in male mice or rats till now. In the present study, we compared the scratching behaviors induced by pruritogenic agents in male and female C57BL/6 mice. The results showed that both males and females exhibited scratching behaviors in response to the intradermal injection of histamine-dependent and histamine-independent pruritogenic chemicals. Moreover, the number of scratching behaviors in response to compound 4880 and chloroquine were significantly higher in females. These results suggested that sex differences occured in histamine-dependent compound 4880-induced and histamine-independent chloroquine-induced itch sensations, but not in histamine-independent SLIGRL-NH2-induced itch sensation.
Escin may exert a synergistic anti-inflammatory effect with glucocorticoids  [PDF]

Abstract: Escin is a natural mixture of triterpenoid as- ponin isolated from the seed of the horse chestnut and demonstrates anti-oedematous and anti-inflammatory effects. As yet, the pre-cise mechanisms by which escin exerts its anti- inflammatory effects remain unclear. The data from current studies indicate that the anti-in-flammatory properties of escin were attributed to its ability to reduce the adhesiveness of neu-trophils and the associated release of inflam-matory mediators; its ability to decrease hista-minic and serotoninergic activities; its ability to inhibit phospholipase A2; its ability to decrease nuclear factor-κ B activation and down-regulate the expression of tumor necrosis factor-α. All these effects are similar to glucocorticoids. Mo- reover, escin depends on adrenal glands to ex-ert its anti-inflammatory effects. Also, our recent research showed that the serum corticosterone level in mice did not increase after a 7-day in-travenous injection of escin. The results sup-port the hypothesis that escin may exert a syn-ergistic anti-inflammatory effect with glucocor-ticoids. Confirming this hypothesis will play a role in elucidating the anti-inflammatory mech- anisms of escin.
Expression of TIP-1 Confers Radioresistance of Malignant Glioma Cells

Abstract: Background Malignant gliomas represent one group of tumors that poorly respond to ionizing radiation (IR) alone or combined with chemotherapeutic agents because of the intrinsic or acquired resistance. In this study, TIP-1 was identified as one novel protein that confers resistance of glioma cells to IR. Methodology/Principal Findings Meta-analysis indicated that high TIP-1 expression levels correlate with the poor prognosis of human malignant gliomas after radiotherapy. Studies with established human glioma cell lines demonstrated that TIP-1 depletion with specific shRNAs sensitized the cells to IR, whereas an ectopic expression of TIP-1 protected the glioma cells from the IR-induced DNA damage and cell death. Biochemical studies indicated that TIP-1 protein promoted p53 ubiquitination and resulted in a reduced p53 protein level. Furthermore, p53 and its ubiquitination are required for the TIP-1 regulated cellular response to IR. A yeast two-hybrid screening identified that TIP-1, through its single PDZ domain, binds to the carboxyl terminus of LZAP that has been studied as one tumor suppressor functioning through ARF binding and p53 activation. It was revealed that the presence of TIP-1 enhances the protein association between LZAP and ARF and modulates the functionality of ARF/HDM2 toward multi-ubiquitination of p53, while depleting TIP-1 rescued p53 from polyubiquitination and degradation in the irradiated glioma cells. Studies with a mouse xenograft model indicated that depleting TIP-1 within D54 cells improved the tumor growth control with IR. Conclusions/Significance This study provided the first evidence showing that TIP-1 modulates p53 protein stability and is involved in the radioresistance of malignant gliomas, suggesting that antagonizing TIP-1 might be one novel approach to sensitize malignant gliomas to radiotherapy.
Evaluation and Adjustment of Coordinated Development between Land Use and Eco-Environment in Wuxi City  [PDF]

Abstract: The purpose of this paper is to assess the coordinated development degree of land use and eco-environment in Wuxi City and choose the appropriate adjustment mode. Methods employed include documentation analysis, the coordinated development degree model analysis and BP neural network model analysis. The result shows that the coordinated development degree of land use and eco-environment in Wuxi City presents overall ever-increasing trends and that the overall consideration mode can reduce the contradiction between land use and eco-environment, which is a suitable mode for Wuxi City. It is concluded that the coordinated development degree model and BP neural network model have high reliability and practicability in evaluating and adjusting the coordinated development degree of land use and eco-environment in Wuxi City.
Molecular Markers for the Prediction of Anti-EGFR Monoclonal Antibody Treatment Efficacy in Metastatic Colorectal Cancer  [PDF]

Abstract: The implementation of individualized targeted therapy for metastatic colorectal cancer (mCRC), in addition to standard chemotherapeutic regimens, currently is a topic under debate. Approximately 35% - 45% of mCRC patients exhibit mutated KRAS, which is considered to be an independent predictor of poor response to treatment with epidermal growth factor receptor (EGFR) monoclonal antibody. However, only about 50% of patients with wild-type KRAS respond to anti-EGFR therapy. Two major EGFR-dependent signaling pathways, RAS-RAF-MAPK and PI3K-PTEN-AKT, may be involved in the poor response to anti-EGFR. Increased EGFR gene copy number as detected by fluorescence in situ hybridization, but not increased EGFR protein expression, correlates with efficacy of anti-EGFR treatment. The identification of mutations in BRAF and PIK3CA (exon 20) and deletions in PTEN also may help clinicians screen for anti-EGFR resistance in mCRC patients with wild-type KRAS. To guide health professionals through the realm of individualized targeted therapies for mCRC, we review recent progress on identifying negative predictors and prognostic markers of anti-EGFR treatment efficacy.
Efficacy of Yiqi Fumai (Freeze-Dried Powder) on Ischemic Diastolic Heart Failure  [PDF]

Abstract:

Objective: To evaluate the efficacy of Yiqi Fumai freeze-dry powder (YFP) on ischemic diastolic heart failure. Methods: 100 patients diagnosed with unstable angina accompanying ischemic diastolic heart failure (IDHF) were selected randomly. 52 patients with TCM syndrome of qi-yin deficiency were divided into Chinese and Western combination therapy group. 48 patients have no TCM syndrome of qi-yin deficiency, and were treated with standard western medicine. After treatment, Seattle Angina Questionnaire Evaluation, ECG, conventional and stress echocardiography (SE) index, NT-proBNP were compared between before and after treatment. Results: The differences of PL, TS, DP between before and after treatment by YFP were statistically significant (P < 0.05). SE parameters of LVEF, E/A were significantly increase; E/e' decreased. The differences were statistically significant. However, echocardiography parameters showed no significant differences after treatment. After the combination treatment, NT-proBNP level had negative correlation with LVEF, E/A (r = -0.432, -0.643, both P < 0.01). Conclusion: Yiqi Fumai freeze-dry powder is safe and effective to patients with ischemic diastolic heart failure. Stress echocardiography can improve the diagnostic of ischemic diastolic heart failure, and maybe an effectively predict treatment response.

One-Step Seedless and Catalyst—Free Growth of Hierarchical ZnO Film Promising for Photoelectrochemical Application  [PDF]

Abstract: A facile one-step method was developed for the fabrication of hierarchical ZnO film on substrate. Neither seed nor catalyst layer is necessary for the growth of hierarchical ZnO film. Three kinds of nucleation process were found, and the influences of growth time, growth electrolyte, growth temperature on the morphology of ZnO film were evaluated. Hierarchical ZnO film can absorb more than 97% of incident photons with wavelength shorter than 380 nm. Such hierarchical ZnO film would be a promising scaffold for photoelectrochemical application.
Coordinating optimization-based sliding mode variable structure control for electro-hydraulic servo system

Abstract: A sliding mode variable structure control (SMVSC) based on a coordinating optimization algorithm has been developed. Steady state error and control switching frequency are used to constitute the system performance indexes in the coordinating optimization, while the tuning rate of boundary layer width (BLW) is employed as the optimization parameter. Based on the mathematical relationship between the BLW and steady-state error, an optimized BLW tuning rate is added to the nonlinear control term of SMVSC. Simulation experiment results applied to the positioning control of an electro-hydraulic servo system show the comprehensive superiority in dynamical and static state performance by using the proposed controller is better than that by using SMVSC without optimized BLW tuning rate. This succeeds in coordinately considering both chattering reduction and high-precision control realization in SMVSC.

Abstract: In the title compound, C13H17NO3·0.5H2O, a new caffeic acid amide derivative, the solvent water molecule lies on a twofold axis and the terminal ethyl group appears disordered with occupancy factors of 0.525 (6) and 0.475 (6). The benzene ring makes an angle of 17.3 (2)° with the C=C—C—O linker. The presence of an ethylenic spacer in the caffeic acid amide molecule allows the formation of a conjugated system, strongly stabilized through π-electron delocalization. The C=C double bond in the linker is trans, similar to those previously reported in caffeic esters. The crystal is stabilized by O—H...O, N—H...O and C—H...O hydrogen bonds. The molecules of the caffeic acid amide form a supermolecular planar structure through O—H...O hydrogen bonds between a hydroxy group of one caffeic acid molecule and a carbonyl O atom of another. These planes interact via C—H...O, O—H...O and N—H...O hydrogen bonds to form a three-dimensional network.
