Abstract: Osmotic dehydration of wheat seedlings in 0.5 MPa polyethylene glycol (PEG) solutions for 24, 48 and 72 h resulted in mild, moderate and severe water stress respectively in leaves, but only caused mild water stress in roots as reflected by the changes in relative water content (RWC). In response to the above water stress conditions, leaf total respiratory rate (Vt) decreased progressively, and the alternative pathway (AP) capacity (Valt) and its actual operation activity (ρValt) decreased more severely. Water stress also led to continuous reduction in cytochrome pathway (CP) activity ((ρ′ V cyt) and different changes in the contribution of ρValt and ρ′Vcyt to Vt, in leaves, with ρValt/ Vt, decreasing and ρ′Vcyt/Vt increasing. The change pattern of root Vt was similar to that of its RWC, while root Valt and ρValt were found to decrease during the first 24 h of stress and thereafter recover to a level close to that of the control (0 h). These data indicate that the alternative pathway is sensitive to water stress and can adapt mild water stress. The results of northern hybridization using an alternative oxidase gene (Aox) as probe revealed that there was a good correlation between Valt (also ρValt) and Aox mRNA levels in both leaves and roots, suggesting that water stress affects the development and operation of AP respiration through affecting the expression of the alternative oxidase gene.
Studies on the relationship between cyanide-resistant respiration and expression of alternative oxidase in mung bean using antibodies prepared by synthetic polypeptide

Abstract: Twelve peptides, including eight conservative amino acid residues in the amino acid sequence of hydrophilic S helix of the alternative oxidase (AOX), were synthesized by solid-phase method. The polypeptide was coupled with α-chymotrypsinogen, and the antibodies were obtained through immunizing domestic rabbit by injecting this complex. By using these antibodies, which were raised to immunoreact with total proteins of purified mitochondria from different organs of mung bean seedlings, we find that there are two hybridizable AOX bands in mitochondria. Their molecular weights are about 35 and 38 ku, respectively. Moreover, the respiratory parameters of hypocotyl, true leaf and cotyledon of mung bean seedlings show that true leaf has the highest total respiration (Vt), alternative pathway (AP) capacity (Valt) and the activity of AP (ρValt) among the three organs. Vt andρV alt of cotyledon ranked the second. Hypocotyl has the lowest Vt andρV alt, but its Valt is higher than that of cotyledon. These results are consistent with the analysis of Western blotting of expression of AOX. The highest Vt andρV alt in true leaf are accompanied two hybridizable polypeptides of AOX protein, 35 ku and 38 ku respectively. The next is cotyledon Vt andρV alt with only one 38 ku hybridizable polypeptide of AOX protein. HypocotylV t andρV alt is the lowest and its immunoblotting band is similar to that of cotyledon, but the expressive amount of 38 ku protein is less than that of cotyledon. The results suggest that the 35 ku AOX may contribute mainly to true leafρV alt.

Abstract: M ssbauer spectrum measured for the iron components of photosystem II (PS II) particles of spinach is a superposition of 4 doublets. Quadrupole splitting and chemical shifting of doublets I–IV are characteristics of proteins with oxidized cytochrome b-559, reduced cytochrome b-559, Fe3+-Q complex and Fe2+ -Q complex respectively. After the PS II particles are treated with La3+, two doublets of Fe2+ disappear and Fe2+ is converted into Fe3+, indicating that the reduced cytochrome b-559 has been converted into the oxidized cytochrome b-559, and Fe2+ -Q complex into Fe3+ -Q complex. The M ssbauer spectrum of PS II particles treated with La3+ and Ca2+ shows that Ca2+ can weaken the inhibitory effect of La3+ in part, and a portion of the reduced cytochrome b-559 and Fe-Q complex still exist.

Abstract: A 3-month feeding experiment was conducted in an in-door seawater system to investigate the effect of dietary vitamin E(Ve) on the sperm quality of turbot(Scophthalmus maximus). D-α-tocopherol acetate was supplemented to the basal(control) diet(65.14 mg kg–1 Ve) to obtain low and high levels of dietary Ve(244.60 mg kg–1, LVe; 721.60 mg kg–1, HVe). Compared with the control, sperm concentration was significantly increased in Ve-supplemented groups(LVe and HVe); while relative sperm volume and testis-somatic index were significantly increased in group HVe only. Sperm motility duration was significantly longer in group HVe than in the control, but no significant difference was observed in percent motility among groups. Sperm size, the uniformity of mitochondrial size, and the integrity of mitochondria cristae and plasma membrane were improved by dietary Ve, especially in HVe. The content of Ve in testis and liver as well as polyunsaturated fatty acids in sperm increased with dietary Ve. These results suggested that dietary Ve, especially at the high level(721.60 mg kg–1), significantly improved sperm concentration and motility duration and maintained normal sperm morphology of turbot

Abstract: In the compatible interaction between potato tuber (Solanum tuberosum L) and soft rot bacteria(Erwinia carotovora pv. Carotovora), the characteristic of active oxygen species production is different in different compatible interactions. In the stronger compatible interaction, there was no obvious production of active oxygen species during 24 hours interaction. But in the weaker compatible interaction, the production of active oxygen species after 8 hours became obvious and reached a peak at 20 hour, which enhanced host resistance to soft rot disease. Experiments also showed that the compatible interactions induced cyanide-resistant alternative pathway in host and SHAM treatment depressed the host susceptibility. At the same time, SHAM treatment also enhanced the production of active oxygen species and a peak appeared. These results showed the host susceptibility was enhanced by cyanide-resistant alternative pathway through inhibition of host active oxygen species production in the compatible interaction.

Effect of insect pest on activated oxygen metabolism in lanceolate leaves of Populus euphratica under different water condition

Abstract: This paper studied the effect of insect pest on the activities of protective enzymes SOD and CAT and the contents of H2O2 and O2*- in lanceolate leaves of Populus euphratica under different soil water conditions. The results showed that soil water deficiency did not increase SOD and CAT activities and H2O2 and O2*- contents in lanceolate leaves, but under this condition, insect pest increased them. When soil water was abundance, insect pest couldn't increase the activities of SOD and CAT and the contents of H2O2 and O2*- in lanceolate leaves. It's concluded that insect pest had an important role in death of Populus euphratica under soil water deficiency.
