Proteomic Analysis of Growth Phase-Dependent Expression of Legionella pneumophila Proteins Which Involves Regulation of Bacterial Virulence Traits

Abstract: Legionella pneumophila, which is a causative pathogen of Legionnaires' disease, expresses its virulent traits in response to growth conditions. In particular, it is known to become virulent at a post-exponential phase in vitro culture. In this study, we performed a proteomic analysis of differences in expression between the exponential phase and post-exponential phase to identify candidates associated with L. pneumophila virulence using 2-Dimentional Fluorescence Difference Gel Electrophoresis (2D-DIGE) combined with Matrix-Assisted Laser Desorption/Ionization–Mass Spectrometry (MALDI-TOF-MS). Of 68 identified proteins that significantly differed in expression between the two growth phases, 64 were up-regulated at a post-exponential phase. The up-regulated proteins included enzymes related to glycolysis, ketone body biogenesis and poly-3-hydroxybutyrate (PHB) biogenesis, suggesting that L. pneumophila may utilize sugars and lipids as energy sources, when amino acids become scarce. Proteins related to motility (flagella components and twitching motility-associated proteins) were also up-regulated, predicting that they enhance infectivity of the bacteria in host cells under certain conditions. Furthermore, 9 up-regulated proteins of unknown function were found. Two of them were identified as novel bacterial factors associated with hemolysis of sheep red blood cells (SRBCs). Another 2 were found to be translocated into macrophages via the Icm/Dot type IV secretion apparatus as effector candidates in a reporter assay with Bordetella pertussis adenylate cyclase. The study will be helpful for virulent analysis of L. pneumophila from the viewpoint of physiological or metabolic modulation dependent on growth phase.
La segunda mitad de la Deducción trascendental en la Crítica de la razón pura (B) de Kant
Nakano,Hirotaka;
Ideas y Valores , 2009,
Abstract: the transcendental deduction in the second edition of kant’s critique of pure reason is divided in two parts. nevertheless, the role of the second half is not immediately clear. this article intends to examine the argument presented in the second half after clarifying its purpose. based on this approach, we sustain an interpretation according to which kant tries to establish the validity of categories for all intuition given through sensibility. this interpretation seeks to confirm a conceptual articulation among sensible intuitions that is not regarded as a result of objective judgment.
La primera mitad de la Deducción trascendental en la Crítica de la razón pura (B) de Kant
Nakano,Hirotaka;
Ideas y Valores , 2008,
Abstract: after the publication of strawson's "the bounds of sense", the transcendental deduction in kant's critique of pure reason has been widely regarded as a proof of the objectivity of knowledge as a condition of unity of self-consciousness. nevertheless, many interpreters accept that there are passages which cannot be easily integrated into such a strategy. in this article, through an analysis of the first half of the transcendental deduction (b), i try to point out the need to adopt a different interpretation according to which kant does not consider objectivity as a condition of self-consciousness, but rather self-consciousness as a necessary condition of objectivity.
La distinción kantiana entre la forma de la intuición y la intuición formal
Nakano, Hirotaka;
Signos filosóficos , 2008,
Abstract: in this article i analyze the notions of form of intuition and formal intuition in § 26 of the critique of pure reason. according to kant, formal intuition contains unity of diverse representations that form of intuition doesn't imply. i want to clarify that formal intuition is a hybrid of categories, space and time, which connects the understanding and the sensibility. on the other hand, kant refers to the form of intuition as the concepts of space and time that were exposed in the "transcendental aesthetic" and also receptivity as a faculty.

Redescription of Takahashia citricola Kuwana, 1909, and its transfer to the genus Pulvinaria Targioni Tozzetti (Coccoidea: Coccidae)

ZooKeys , 2012, DOI: 10.3897/zookeys.217.3405
Abstract: The Japanese soft scale Takahashia citricola Kuwana, 1909 is redescribed and transferred to the genus Pulvinaria Targioni Tozzetti as Pulvinaria citricola (Kuwana, 1909), comb. n. (Coccoidea: Coccidae). Pulvinaria gamazumii Kanda, 1960 is synonymized with P. citricola comb. n. and Pulvinaria nipponica Lindinger, 1933, is resurrected as the replacement name for Pulvinaria citricola Kuwana, 1914 (nec Kuwana, 1909). The adult female of P. citricola (Kuwana, 1909) is redescribed and illustrated.
Abstract: In this article I analyze the notions of form of intuition and formal intuition in § 26 of the Critique of Pure Reason. Theory of Membrane in Heegaard Diagram Expansion
Hirotaka Sugawara
Physics , 2003,
Abstract: The vacuum amplitude of the closed membrane theory is investigated using the fact that any three-dimensional manifold has the corresponding Heegaard diagram (splitting) although it is not unique. We concentrate on the topological aspect with the geometry considered only perturbatively. In the simplest case where the action describes the free fields we find that the genus one amplitudes (lens space) are obtained from the S3 amplitude by merely renormalizing the membrane tension. The amplitudes corresponding to the Heegaard diagram of genus two or higher can be calculated as the Coulomb amplitudes with arbitrary charge distributed on a knot or a link which corresponds to the set of branch points of a given regular or an irregular covering space. We also discuss the case of membrane instanton.
Highly distorted apparent horizons and the hoop conjecture
Hirotaka Yoshino
Physics , 2007, DOI: 10.1103/PhysRevD.77.041501
Abstract: By analyzing the apparent horizon (AH) formation in the collision of two pp-waves with rectangular sources in four dimensions, we study to what extent the AH can be distorted without violating the energy conditions. It is shown that the highly distorted AH can form in this system although it cannot be arbitrarily long. The hoop conjecture is examined for the formation of such highly distorted AHs, and our result gives a strong support to the hoop conjecture. We also point out the possible relation between the AH topology theorem and the hoop conjecture.
