Abstract: Background and objective It has been proven that epithelial-mesenchymal transition (EMT) not only correlated with embryonic development but also could promote tumor invasion and metastasis. Transforming growth factor beta-1 (TGF-β1) has been identified as the main inducer of tumor EMT. The aim of this study was to investigate the effects of TGF-β1 on EMT and PI3K/AKT signaling pathway in lung adencarcinoma PC9 cells. Methods Cultured PC9 cells were treated with different concentrations of TGF-β1 for 48 h. The morphological changes were observed under phase-contrast microscopy; EMT relative marker protein changes were assessed by Western blot and immunoflurescence staining. In addition, the expression of AKT and P-AKT were also measured by Western blot. Results The data showed that TGF-β1 could induce PC9 morphological alteration from epithelial to mesenchymal and upregulate the expression of mesenchymal maker protein Fibronectin. Obviously, the expression of P-AKT was downregulated by TGF-β1 treatment for 48 h. Conclusion TGF-β1 might induce EMT of PC9 cells , accompanied by the changes of PI3K/AKT signaling pathway.

Abstract: Gene therapy constitutes a promising strategy for the treatment of osteoarthritis (OA). We assessed the use of electroporation (EP) of non-viral gene vectors, and compared its efficacy with that of adeno-associated virus (AAV) vectors. EP- and AAV-mediated delivery of human interleukin-1 receptor antagonist (hIL-1Ra) was localized performed in the joints of rats following induction of OA. mRNA levels for hIL-1Ra, IL-1β, TNF-α, MMP-13 and ADAMTS-4 in the cartilage and synovial tissues were analyzed. Structural analyses of the subchondral bone at the medial femoral condyle were performed by Micro-CT after treatment. Knee joint specimens were staining with hematoxylin and eosin and Saffron O. Induction of hIL-1Ra by both EP and AAV inhibited inflammatory-induced sub-chondral bone reconstruction, and effectively suppressed IL-1β activity, as evidenced by decreased expression of MMP-13 and ADAMTS-4. Histological analyses revealed significant protection of cartilage, proteoglycan by EP and AAV. hIL-1Ra expression was similar in both the EP and AAV groups. Notably, this gene is not easier degraded transduced by EP compared with AAV. Taken together, these results show that EP offers transfection efficiency comparable to that of AAV, with the potential for longer gene expression, making EP a promising candidate for efficient non-viral delivery of OA gene therapy.

miRNA-29c Suppresses Lung Cancer Cell Adhesion to Extracellular Matrix and Metastasis by Targeting Integrin β1 and Matrix Metalloproteinase2 (MMP2)

Abstract: Our pilot study using miRNA arrays found that miRNA-29c (miR-29c) is differentially expressed in the paired low-metastatic lung cancer cell line 95C compared to the high-metastatic lung cancer cell line 95D. Bioinformatics analysis shows that integrin β1 and matrix metalloproteinase 2 (MMP2) could be important target genes of miR-29c. Therefore, we hypothesized that miR-29c suppresses lung cancer cell adhesion to extracellular matrix (ECM) and metastasis by targeting integrin β1 and MMP2. The gain-of-function studies that raised miR-29c expression in 95D cells by using its mimics showed reductions in cell proliferation, adhesion to ECM, invasion and migration. In contrasts, loss-of-function studies that reduced miR-29c by using its inhibitor in 95C cells promoted proliferation, adhesion to ECM, invasion and migration. Furthermore, the dual-luciferase reporter assay demonstrated that miR-29c inhibited the expression of the luciferase gene containing the 3′-UTRs of integrin β1 and MMP2 mRNA. Western blotting indicated that miR-29c downregulated the expression of integrin β1 and MMP2 at the protein level. Gelatin zymography analysis further confirmed that miR-29c decreased MMP2 enzyme activity. Nude mice with xenograft models of lung cancer cells confirmed that miR-29c inhibited lung cancer metastasis in vivo, including bone and liver metastasis. Taken together, our results demonstrate that miR-29c serves as a tumor metastasis suppressor, which suppresses lung cancer cell adhesion to ECM and metastasis by directly inhibiting integrin β1 and MMP2 expression and by further reducing MMP2 enzyme activity. The results show that miR-29c may be a novel therapeutic candidate target to slow lung cancer metastasis.

Abstract: BaWO4 nanorods have been successfully synthesized in w/o microemulsion system containing barium ions via a simple reaction between Ba2+ and \"\" . The BaWO4 Nanorods were characterized by XRD, TEM, and SEM, respectively. Results showed that the solvents composition—volume ratio of 4-dioxane and distilled water—played the key role in the formation of BaWO4 Nanorods. Furthermore, the strong vibration at 925 cm1 on its Raman spectrum indicated that the BaWO4 nanorods is good at stimulating Raman scattering in transient and steady-state, making it as a promising candidate material for laser with self-raman conversion of radiation inside the active medium.

The author expresses his opinion and consideration about the standardization of psychological translation in this paper,and used this paper as a way of commemorating Prof. Gao Juefu.

Abstract: “徐锡麟手毙陈伯平”之说首见于当年《民报》上南史氏 (陈去病 )的《徐锡麟传》。翌年 ,陈去病删去“或曰毙于君手”一语 ,并慎重地署上自己的真实姓名 ,上说随之消隐。此说再度出现是近五十年后凌孔彰的《先师徐锡麟事迹纪略》 ,但在纪念辛亥革命 60周年之际 ,凌对陈伯平之死的说法作了根本性的修正 ,称“陈伯平又在奋勇击敌中首先中弹牺牲。”今有的人未加考辨 ,把凌孔彰自己已抛弃的说法又摭拾回来 ,使这个问题平添了新的混乱
Studies on the Photoinduced Interaction between Zn(II) Porphyrin and Colloidal TiO2

Abstract: The interaction of Zn(II) porphyrin (ZnPP) with colloidal TiO2 was studied by absorption and fluorescence spectroscopy. The fluorescence emission of ZnPP was quenched by colloidal TiO2 upon excitation of its absorption band. The quenching rate constant ( ) is ?M?1?s?1. These data indicate that there is an interaction between ZnPP and colloidal TiO2 nanoparticle surface. The quenching mechanism is discussed on the basis of the quenching rate constant as well as the reduction potential of the colloidal TiO2. And the mechanism of electron transfer has been confirmed by the calculation of free energy change by applying Rehm-Weller equation as well as energy level diagram. 1. Introduction Wide-band gap semiconductor particles such as TiO2 have been widely used for different applications in photocatalysis and the environment [1, 2].Over the past decades, considerable interest has been shown in the modification of TiO2 semiconductors by organic dyes to extend the photoresponse to visible light owing to their potential application in solar energy conversion [3–5]. Dye sensitization is considered to be an efficient method to modify the photo response properties of TiO2 particles. The dyes used are erythrosine B [6], rose Bengal [7], metal porphyrin [8–10], and so forth. Porphyrins, (including metal-free porphyrins, metalloporphyrins and supramolecular porphyrins) [11] are recognized to be the most promising sensitizers [12]. The chemistry of porphyrin derivatives has played an important role especially during the past decade in particular branches of new materials science, and many researchers have undertaken projects on the synthesis of variously substituted compounds to obtain new functional materials [13–15]. Metalloporphyrin may be an appropriate candidate because of its high absorption coefficient within the solar spectrum and its good chemical stability in comparison to that of other dyes. They are highly effective photocatalysts due to their very strong absorption in the 400?nm–450?nm region (Soret band) and in the 500?nm–700?nm region (Q-bands) and, in fact, the presence of p-electrons affords the condition for electron transfer during the photoreaction. In the present work we have investigated the electron transfer from excited ZnPP (see Scheme 1) to the conduction band of TiO2 colloid by using absorption and fluorescence spectroscopy. Abstract: Mesoporous silica molecular sieves are a kind of unique catalyst support due to their large pore size and high surface area. Several methods have been developed to immobilize heteropolyacids (HPAs) inside the channels of these mesoporous silicas. The mesoporous silica-supported HPA materials have been widely used as recyclable catalysts in heterogeneous systems. They have shown high catalytic activities and shape selectivities in some reactions, compared to the parent HPAs in homogeneous systems. This review summarizes recent progress in the field of mesoporous silica-supported HPAs applied in the heterogeneous catalysis area and preparation of nanostructured metal oxides using HPAs as precursors and mesoporous silicas as hard templates.
