Identification of Two Cold Water-Soluble Polysaccharides from the Stems of Ephedra sinica Stapf  [PDF]

Abstract: Two polysaccharides (ESP-A1 and ESP-A2) were isolated from the cold water extract of Ephedra sinica Stapf and purified through ethanol precipitation, deproteinization and by ion exchange and gel-filtration chromatography. Their molecular weight was determined using high performance size exclusion chromatog-raphy and evaporative light scattering detector (HPSEC-ELSD) and their monosaccharide composition was analyzed by high performance capillary electrophoresis (HPCE) based on pre-column derivatization with 1-phenyl-3-methyl-5-pyrazolone (PMP). It was shown that ESP-A1 consisted of xylose, arabinose, glucose, mannose and galactose and ESP-A2 consisted of xylose, arabinose, rhamnose and galactose, in a molar ratio (%) of 3.2: 61.1: 11.1: 12.9: 11.6 and 20.6: 67.7: 5.0: 6.7, respectively. The molecular weights (Mw) of ESP-A1 and ESP-A2 were 5.83 × 104 Da and more than 200 × 104 Da, respectively. To the best of our knowledge, two neutral polysaccharides are now being reported for the first time in this study.
Clinical Teleacupuncture between China and Austria Using Heart Rate Variability in Patients with Depression  [PDF]

Abstract: In previous studies by the same research group it has been shown in post-stroke patients that the autonomic system can be affected by acupuncture. Within this study, teleacupuncture between China and Austria is used for quantifying the effects of heart rate variability (HRV) in Chinese patients suffering from depression. In 22 Chinese depression patients (17 f, 5 m; mean age ± SD 52.3 ± 10.6 years; range 31 - 70 years) electrocardiographic signals before, during and after acupuncture at the acupoint Jianshi (PC 5) were recorded in Harbin and analyzed in Graz using teleacupuncture via internet. HRV data were analyzed in the time and frequency domain and a protocol from Austria was sent to the team in China immediately after the treatment and recording session. Mean heart rate decreased significantly (p < 0.001) during and after acupuncture, whereas total HRV increased significantly during (p < 0.034) and after (p < 0.001) acupuncture, always immediately following manual needle stimulation. Furthermore, there is a marked decrease in balance between sympathetic and parasympathetic activity (low frequency/high frequency HRV ratio) during treatment. This study shows that HRV could be a useful parameter for quantifying clinical effects of acupuncture on the autonomic nervous system. Teleacupuncture between China and Austria over a distance of more than 8,500 km has been successfully used.

Abstract: This second part of a series of Sino-European high-tech acupuncture studies describes the first clinical transcontinental teleacupuncture measurements in patients with insomnia. Heart rate (HR) and heart rate variability (HRV) measurements in 28 patients (mean age ± SD: 41.9 ± 14.6 years) were performed under standardized conditions in Harbin, China, and the data analysis was performed in Graz, Austria. Similar to the first part of the series, the electrocardiograms (ECGs) were recorded by an HRV Medilog AR12 system during acupuncture of the Shenmen point (HT7) on the left hand. HR decreased significantly (<0.001) during and after acupuncture stimulation of the HT7 acupuncture point. Total HRV increased significantly (<0.05) immediately after acupuncture stimulation, but there was no long-lasting effect. The values of the low-frequency (LF) and high-frequency (HF) band increased significantly after the stimulation compared to baseline values; however, the LF/HF ratio showed no significant changes. Together with the results of previous studies, the present results can serve as a solid basis for further development of acupressure or acupuncture stimulation equipment for complementary use in treating insomnia.
Compounds from the Roots and Rhizomes of Valeriana amurensis Protect against Neurotoxicity in PC12 Cells

Abstract: Three new germacrane-type sesquiterpenoids, heishuixiecaoline A–C (compounds 1–3), were isolated along with ten known compounds 4–13 from fraction of Valeriana amurensis roots and rhizomes effective against Alzheimer’s disease (AD). The structures of 1–3 were elucidated on the basis of their spectroscopic data. We also investigated the protective effect of compounds 1–13 on the neurotoxicity of PC12 cells induced by amyloid-beta (Aβ25–25), respectively. As a result, germacrane-type sesquiterpenoids 1–4 and lignans 5–7 were seen to afford protection against Aβ-induced toxicity in PC 12 cells. This study will contribute to revealing the chemical basis for the therapeutic effect of V. amurensis against AD.

Abstract: Three new cycloartenol triterpene saponins, named shengmaxinsides A-C, have been isolated from the ethyl acetate soluble fraction of an ethanol extract of Cimicifuga simplex Wormsk roots. Their structures were established by chemical tests and detailed spectroscopic analysis as 25-O-acetyl-7,8-didehydrocimigenol-3-O-β-D-galactopyranoside (1), 7,8-didehydrocimigenol-3-O-β-D-galactopyranoside (2) and 7,8-didehydro-24S-O-acetylhydroshengmanol-3-O-β-D-galactopyranoside (3), respectively.

Abstract: Four new glycosides, namely 3β-norpinan-2-one 3- O-β-d-apiofuranosyl-(1→6)-β-d-glucopyranoside ( 1), (6 Z)-3-hydroxymethyl-7-methylocta-1,6-dien-3-ol 8- O-β-d-glucopyranoside ( 2), (6 E)-3-hydroxymethyl-7-methylocta-1,6-dien-3-ol 8- O-β-d-gluco-pyranoside ( 3), and 7-[(β-d-apiofuranosyl-(1→6)-β-d-glucopyranosyl)oxymethy]-8,8-dimethyl-4,8-dihydrobenzo[1,4]thiazine-3,5-dione ( 4), were isolated from the fruits of Xanthium sibiricum Patr together with three known compounds, xanthiside ( 5), adenosine ( 6), and 2,3-dihydroxy-1-(4-hydroxy-3-methoxyphenyl)-propan-1-one ( 7). The structures of the new compounds were determined on the basis of detailed spectroscopic analyses.
Biomedical Teleacupuncture between China and Austria Using Heart Rate Variability, Part 1: Poststroke Patients

Abstract: Background. Acupuncture has been reported to affect the human autonomic system. Within this pilot study, teleacupuncture between China and Austria is used for the first time for quantifying the effects of heart rate variability (HRV) in poststroke rehabilitation. Methods. In 29 Chinese post-stroke patients (15 f, 14 m; mean age ± SD 64.7±11.3 years; range 40–80 years) electrocardiographic signals before, during, and after acupuncture at the acupoint Tongli (HT 5) were recorded in Harbin and analyzed in Graz using teleacupuncture via internet. HRV data were analyzed in the time and frequency domain, and a protocol from Austria was sent to the team in China immediately after the treatment and recording session. Results. Acupuncture does not change heart rate in the post-stroke patients; however, total HRV increased significantly (<.05) during and 5–10 minutes after acupuncture. In addition, balance between sympathetic and parasympathetic activity (low frequency/high frequency HRV ratio) changes markedly during treatment. Conclusions. Based on innovative HRV analysis, it could be demonstrated that teleacupuncture between China/Harbin and Austria/Graz over a distance of about 8,500 km is no longer a future vision; it has become reality.

Abstract: It has been shown in previous studies that the autonomic nervous system can be affected by acupuncture. Within this study, teleacupuncture between China and Austria is used for quantifying the effects of heart rate (HR) and heart rate variability (HRV) in 33 Chinese patients (27 females, 6 males; mean age ± SD 49.5±13.1 years; range 22–72 years) suffering from depression. Electrocardiographic signals before, during, and after acupuncture at the acupoint Baihui (GV20) were recorded in Harbin and analyzed in Graz using teleacupuncture. HRV data were analyzed in the time and frequency domain. Mean HR decreased significantly (<0.05) during and after acupuncture, whereas total HRV increased significantly after the third acupuncture stimulation period (<0.05) and also 5–10 minutes after (<0.05) acupuncture. The study shows that HRV could be a useful parameter for quantifying clinical effects of acupuncture on the autonomic nervous system.

Abstract: Objective: To study the migration of Rutin, Quercetin and Kaemperol in HPCE. Methods: The vdtage of HPCE.pH and concentration of buffer were changed respectively to study the effect for separation. Results: The shape peaks and separation were proved to be good under the condition which pH was 8.65,and voltage was 20kv and concentration was 12mmol/L. Conclusion: The separation degree of the three compounds was mainly determined by pH of buffer, and migration time of them were mainly determined voltage and concentration of buffer.

Abstract: 目的:制备洋金花总黄酮磷脂复合物并考察其理化性质。方法:以洋金花总黄酮与磷脂的复合率为指标,考察复合溶剂、复合时间、复合温度、初始浓度和投料比例对复合率的影响,优化制备工艺;对所得磷脂复合物的理化性质,包括油/水分配系数、粒径、在水溶液中的状态以及形成的单分子膜的特性等进行了研究。结果:洋金花总黄酮磷脂复合物的最佳制备工艺为:洋金花总黄酮与磷脂按1颐1.8 的质量比投料,于30益甲醇中复合1h,总黄酮的浓度为10g·L-1,复合率95%以上。差热扫描显示复合物的相变温度改变;透射电镜显示复合物在水中为圆整均匀的囊泡,平均粒径48.51依9.13nm;在pH 为1.0、6.5、9.0 的条件下,复合物的油/水分配系数分别为总黄酮的4.22、3.06、3.75 倍;和磷脂复合物的结构相对应,Langmuir-blodgett(LB)膜仔-A 等温曲线显示复合物具有比磷脂强的刚性。结论:通过在甲醇中复合,成功制备了洋金花总黄酮的磷脂复合物;复合物较黄酮的脂溶性增加,较磷脂亲水性增加,在水相中组装成纳米胶束。膜天平是研究磷脂复合物的又一有力工具。
