DNA Fingerprinting of Chinese Melon Provides Evidentiary Support of Seed Quality Appraisal

Abstract: Melon, Cucumis melo L. is an important vegetable crop worldwide. At present, there are phenomena of homonyms and synonyms present in the melon seed markets of China, which could cause variety authenticity issues influencing the process of melon breeding, production, marketing and other aspects. Molecular markers, especially microsatellites or simple sequence repeats (SSRs) are playing increasingly important roles for cultivar identification. The aim of this study was to construct a DNA fingerprinting database of major melon cultivars, which could provide a possibility for the establishment of a technical standard system for purity and authenticity identification of melon seeds. In this study, to develop the core set SSR markers, 470 polymorphic SSRs were selected as the candidate markers from 1219 SSRs using 20 representative melon varieties (lines). Eighteen SSR markers, evenly distributed across the genome and with the highest contents of polymorphism information (PIC) were identified as the core marker set for melon DNA fingerprinting analysis. Fingerprint codes for 471 melon varieties (lines) were established. There were 51 materials which were classified into17 groups based on sharing the same fingerprint code, while field traits survey results showed that these plants in the same group were synonyms because of the same or similar field characters. Furthermore, DNA fingerprinting quick response (QR) codes of 471 melon varieties (lines) were constructed. Due to its fast readability and large storage capacity, QR coding melon DNA fingerprinting is in favor of read convenience and commercial applications.
Silencing of Tumor Necrosis Factor Receptor 1 by siRNA in EC109 Cells Affects Cell Proliferation and Apoptosis

Abstract: Tumor necrosis factor receptor 1 (TNFR1) is a membrane receptor able to bind TNF- or TNF-. TNFR1 can suppress apoptosis by activating the NF-B or JNK/SAPK signal transduction pathway, or it can induce apoptosis through a series of caspase cascade reactions; the particular effect may depend on the cell line. In the present study, we first showed that TNFR1 is expressed at both the gene and protein levels in the esophageal carcinoma cell line EC109. Then, by applying a specific siRNA, we silenced the expression of TNFR1; this resulted in a significant time-dependent promotion of cell proliferation and downregulation of the apoptotic rate. These results suggest that TNFR1 is strongly expressed in the EC109 cell line and that it may play an apoptosis-mediating role, which may be suppressed by highly activated NF-B.
Expression of Acetaldehyde Dehydrogenase in Gefitinib-resistant Human Lung Adenocarcinoma HCC-827/GR Cells

Abstract: Background and objective Tumor recurrence and drug resistance are the main causes of death in tumor patients. The family of acetaldehyde dehydrogenase (ALDH) is closely related to the proliferation, migration, invasion and resistance of tumor cells, and different ALDH subtypes are expressed in different tumor cells. The aim of this study is to elucidate the ALDH subtype in human lung adenocarcinoma HCC-827/GR cells, which resistant to the gefitinib. Methods The human lung adenocarcinoma HCC-827 cells were used to generate the gefitinib-resistant HCC-827/GR cells; the expression of ALDH subtype in either HCC-827 or HCC-827/GR was detected by flow cytometry; The proliferative capacity and sensitivity to gefitinib of hcc-827/GR cells were analyzed by MTT assay before and after treatment with 100 μmol/L diethyllaminaldehyde (DEAB); Real-time quantitative PCR was used to detect the expression of ALDH subtypes at mRNA levels in hcc-827 cells and hcc-827/GR cells. Results Compared with HCC-827 cells, the positive rate of ALDH in HCC-827/GR cells increased. The proliferation ability of HCC-827/GR cells decreased after treatment with 100 μmol/L DEAB. Compared with HCC-827 cells, the expression of ALDH1A1 and ALDH1L1 mRNA was increased in hcc-827/GR cells, but the ALDH3B2 expression was decreased. Conclusion ALDH might be used as a molecular biomarker to test the gefitinib-resistant to lung adenocarcinoma cancer cells, and the ALDH1A1 may play a role in gefitinib resistance in lung cancer. ?

Abstract: A novel fabrication method for surface-enhanced Raman scattering (SERS) sensors that used a fast femtosecond (fs) laser scanning process to etch uniform patterns and structures on the endface of a fused silica optical fiber, which is then coated with a thin layer of silver through thermal evaporation is presented. A high quality SERS signal was detected on the patterned surface using a Rhodamine 6G (Rh6G) solution. The uniform SERS sensor built on the tip of the optical fiber tip was small, light weight, and could be especially useful in remote sensing applications.
Preparation and cytocompatibility of polylactic acid/hydroxyapatite/graphene oxide nanocomposite fibrous membrane

Abstract: A series of polylactic acid (PLA) based nanocomposite fibrous membranes, including neat PLA, PLA/hydroxyapatite (HA) and PLA/HA/graphene oxide (GO), were fabricated via electrospinning method. The morphology and composition were investigated by scanning electron microscopy (SEM), X-ray diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) respectively. The thermal stability was determined by thermogravimetric analysis (TGA). To estimate the cytocompatibility of asprepared PLA/HA/GO fibrous membrane, MC3T3-E1 cells were cultured, and the corresponding cell adhesion and differentiation capability were investigated by fluorescence microscopy, SEM and MTT test. The electrospun ternary PLA/HA/GO membrane exhibited three-dimensional fibrous structure with relatively rough surface morphology, which made itself ideal for cell attachment and proliferation in bone tissue regeneration. The fluorescence microscopy, SEM and MTT test confirmed that the PLA/HA/GO nanocomposite fibrous membrane created a proper environment for the seeding and proliferation of MC3T3-E1 cells.
Mid-Field Magnetic Resonance Imaging and Cross-Sectional Anatomy of the Yearling Chinese Equine Distal Forelimb

Abstract: The objective of this study is to perform mid-field (0.4T) Magnetic Resonant Imaging (MRI) on a distal forelimb of a healthy Mongolian yearling horse and obtain a detailed cross-sectional comparison between the MRI and anatomy images. T1-weighted, T2-weighted, PD-weighted and STIR sequence were performed to acquire images of the horse distal limb on the transverse plane. After imaging, the limb was frozen and cut into slices to get anatomy photos. Comparisons between the MR images and corresponding frozen cross-sections of the horse limb were made to identify the anatomic details of the horse limb. The results of this study indicate that mid-field MRI can be used to diagnose lesions in the equine distal forelimb at an early stage of lameness. Besides, this study also provides an atlas of gross anatomy of the carpus and fetlock in mid-field MRI which may benefit veterinary clinical evaluation and medical research.

Abstract: There have always been controversies upon negotiable instrument counterplea and negotiable instrument counterplea right among Chinese negotiable instruments law scholars. If legal scholars cannot clearly understand the contents of negotiable instrument counterplea and negotiable instrument counterplea right, there will be chaos in the whole negotiable instrument counterplea theory which goes against the protection of negotiable instrument obligors’ benefit and the balance of negotiable instrument right system.
