Effects of Baicalin and Ligustrazine on airway inflammation and remodeling and underlying mechanism in asthmatic rats  [PDF]

Abstract: Aim: To explore the effects of Baicalin and Ligustrazine on airway inflammation and construction and underlying mechanisms through the expressions of GATA-3, IL-5, MMP-9 and TIMP-1 in asthmatic rats. Methods: 30 Wistar rats were randomly divided equally into five groups. Lung tissues were sliced. WBC and Eos in lung tissue were estimated by HE stain and the expressions of IL-5, GATA-3, MMP-9, TIMP-1 and collagen type IV in lung tissue were observed by immunohistochemistry. The airway wall and airway smooth muscle thicknesses were measured by computed image analysis system. Results: Compared with asthma group, EOS counts and the expression of IL-5 and GATA-3 in the lung tissue were significantly lower in normal controlled groups (P < 0.05 or P < 0.01). Additionally, the thickness of airway wall and airway smooth muscle did significantly increase, and the expressions of collagen type Ⅳ, MMP-9 and TIMP-1 were significantly higher in asthma group (P < 0.05). With the intervention of Baicalin or Ligustrazine, EOS decreased, and the thicknesses of airway wall and airway smooth muscle became thinner compared with asthma group. Meanwhile, the expression of collagen type IV, IL-5, GATA-3, MMP-9 and TIMP-1 significantly decreased (P < 0.05). Airway wall thickness and collagen type Ⅳ were associated with Eos, IL-5, TAGA-3, MMP-9, TIMP-1 and MMP-9/ TIMP-1. Conclusion: Two herbs could diminish infiltration of EOS with inhibiting the expressions of IL-5, and GATA-3, meanwhile, decrease the deposition of collagen type IV and the thickness of the airway smooth muscle through regulating MMP-9, TIMP-1 level and the balance between MMP-9 and TIMP-1, additionally, had synergetic effects.
Physicochemical evaluation and essential oil composition analysis of Hyssopus cuspidatus Boriss from Xinjiang, China

Abstract: Background: It is reported that the plant Hyssopus cuspidatus Boriss from Xinjiang has great value. This article deals with the detailed pharmacognostic evaluation of the crude drug H. cuspidatus Boriss. Materials and Methods: The essential oil of H. cuspidatus Boriss from Xinjiang, China, was extracted by the method of hydrodistillation and the chemical composition of the essential oil was analyzed by gas chromatography-mass spectrometry (GC-MS). Results: The yield of essential oil based on the dry weight of the plant was 0.6%(w/w). Fifty compounds accounting for 99.42% of the total oil were identified. The major components were oxygenated terpenes (66.33%), monoterpenes (26.14%), oxygenated sesquiterpenes (1.25%), and octane (1.85%). Conclusion: Oxygenated terpenes were the main group of the compounds. The physicochemical parameters presented in this article may be proposed as parameters to establish the authenticity of H. cuspidatus Boriss and can possibly aid pharmacognostic and taxonomic species identification.
Effect of mixed culture of Saccharomyces cerevisiae and Pichia anomala on fermentation efficiency and flavor compounds in Chinese Liquor

Abstract: 【目的】通过酿酒酵母(Saccharomyces cerevisiae)和异常毕赤酵母(Pichia anomala)在麸皮汁培养基中的混菌发酵,以增加发酵液的风味酯含量并保证发酵效率。【方法】采用两种酵母混合接种、顺序接种混菌发酵方式,以酵母单独接种发酵作对照,测定酵母的发酵性能和发酵液中乙酸乙酯含量,并对发酵结束时风味物质进行半定量;利用无细胞系统,分析两种酵母之间的相互作用。【结果】采用顺序接种混菌发酵方式,避免S.cerevisiae对P.anomala的生长竞争性抑制,使两种酵母均能获得较高的生物量;发酵结束时,乙醇浓度为20.17 g/L,比酿酒酵母单菌种发酵时降低了9.14%;但乙酸乙酯含量达到0.74 g/L,比异常毕赤酵母单菌种发酵时提高了80%;发酵液风味物质的测定结果表明,酿酒酵母与异常毕赤酵母的混合发酵能够形成更多的酯类物质,总酸和高级醇含量却相对较低,有效改善了发酵液的风味特性;在混菌发酵时,碳源是影响酿酒酵母繁殖的重要因素,但酵母的代谢物对异常毕赤酵母产生明显的抑制作用。【结论】混菌发酵,为丰富发酵产物的风味复杂性和增强风格的独特性提供了一条有效的途径。

Abstract: To study the hydrologic cycle, water balance, the use and management of water resources, it is often improtant to have a better understanding of the variations and the change trend of discharges for the system. This paper presents the statistical analysis of the discharge and precipitation data for the Jialing River at the Beibei Hydrologic Gaging Station. 60 years of data have been used in this study for the Kendall correlation analysis, the cumulative mass analysis, and the Mann-Kendall statistical analysis for the change points and change trend in precipitation and flows. Thk Kendall correlation analysis and the Mann-Kendall statistical analysis showed that the discharges in the Jialing River had a decreasing thend with the change point in 1992. The cumulative discharge analysis showed that there were two clear high flow periods, one average flow period, and two dry periods. The Mann-Kendall analysis showed that precipitation had a significant reduction since 1993. Discharges correlate well with the precipitation within the river basin ,which indicated that precipitation is the primary cause of flow reduction in the river. Other factors contributed to the reduction of flow include urbanization, dam constructions on the river, and usage of water in the upper regions.

Inhibition of Longzhi extracts on tumor growth of transplanted H22 and S180 in mice

Abstract: Objective: To study the inhibitory effect of Longzhi extracts (extracts from a Chinese herbal compound) on the growth of transplanted hepatoma H22 and sarcoma S180 in mice. Methods: Certain dosage of Longzhi extracts was dissolved into distilled water to obtain the suspension of definite concentration. The suspension of 0.2 ml was planted on the right forefoot of each mouse. The mice were randomly divided into groups 24 hours later and treated for 7 days. One day after the treatment stopped, the mice were sacrificed and the tumor masses were taken out and weighed. Results: The inhibition rates of Longzhi extracts on the tumor of transplanted H22 and S180 were 63% and 41% respectively. Conclusion: Longzhi extracts has antitumor action.

Abstract: The bacterial community structure of 5 high and medium temperature Chinese liquor Daqu were investigated using PCR-DGGE (Polymerase chain reaction-denaturing gradient gel electrophoresis) as a culture-independent method. DNA sequencing was proceeded to obtain the dominant bacterial population information. The result of DGGE profile showed that Weissella cibaria, Lactobacillus helveticus, L. fermentum and L. panis were commonly detected in all the five Daqu. Thermoactinomyces sanguinis was detected only in the Jiangqu sample. Compared with culture-dependent method, DGGE was able to detect Staphylococcus xylosus and Klebsiella oxytoca. The correlation between the craftwork of Daqu and the bacterial community structure was obvious. The Daqu bacterial Shannon-Wiener index decreased as the craftwork temperature of Daqu increasing. PCR-DGGE was proved to be a powerful tool for gaining detailed insight into the bacterial diversity of the Chinese liquor Daqu.
Folding rate prediction using complex network analysis for proteins with two- and three-state folding kinetics  [PDF]

Abstract: It is a challenging task to investigate the different in- fluence of long-range and short-range interactions on two-state and three-state folding kinetics of protein. The networks of the 30 two-state proteins and 15 three-state proteins were constructed by complex networks analysis at three length scales: Protein Contact Networks, Long-range Interaction Networks and Short-range Interaction Networks. To uncover the relationship between structural properties and folding kinetics of the proteins, the correlations of protein network parameters with protein folding rate and topology parameters contact order were analyzed. The results show that Protein Contact Networks and Short-range Interaction Networks (for both two-state and three-state proteins) exhibit the “small-world” property and Long-range Interaction networks indicate “scale-free” behavior. Our results further indicate that all Protein Contact Networks and Short- range Interaction networks are assortative type. While some of Long-range Interaction Networks are of assortative type, the others are of disassortative type. For two-state proteins, the clustering coefficients of Short-range Interaction Networks show prominent correlation with folding rate and contact order. The assortativity coefficients of Short-range Interaction Networks also show remarkable correlation with folding rate and contact order. Similar correlations exist in Protein Contact Networks of three-state proteins. For two-state proteins, the correlation between contact order and folding rate is determined by the numbers of local contacts. Short- range interactions play a key role in determining the connecting trend among amino acids and they impact the folding rate of two-state proteins directly. For three-state proteins, the folding rate is determined by short-range and long-range interactions among residues together.
