Sodium Coupled Bicarbonate Influx Regulates Intracellular and Apical pH in Cultured Rat Caput Epididymal Epithelium

Abstract: Background The epithelium lining the epididymis provides an optimal acidic fluid microenvironment in the epididymal tract that enable spermatozoa to complete the maturation process. The present study aims to investigate the functional role of Na+/HCO3? cotransporter in the pH regulation in rat epididymis. Method/Principal Findings Immunofluorescence staining of pan cytokeratin in the primary culture of rat caput epididymal epithelium showed that the system was a suitable model for investigating the function of epididymal epithelium. Intracellular and apical pH were measured using the fluorescent pH sensitive probe carboxy-seminaphthorhodafluor-4F acetoxymethyl ester (SNARF-4F) and sparklet pH electrode respectively to explore the functional role of rat epididymal epithelium. In the HEPES buffered Krebs-Henseleit(KH) solution, the intracellular pH (pHi) recovery from NH4Cl induced acidification in the cultured caput epididymal epithelium was completely inhibited by amiloride, the inhibitor of Na+/H+ exchanger (NHE). Immediately changing of the KH solution from HEPES buffered to HCO3? buffered would cause another pHi recovery. The pHi recovery in HCO3? buffered KH solution was inhibited by 4, 4diisothiocyanatostilbene-2, 2-disulfonic acid (DIDS), the inhibitor of HCO3? transporter or by removal of extracellular Na+. The extracellular pH measurement showed that the apical pH would increase when adding DIDS to the apical side of epididymal epithelial monolayer, however adding DIDS to the basolateral side had no effect on apical pH. Conclusions The present study shows that sodium coupled bicarbonate influx regulates intracellular and apical pH in cultured caput epididymal epithelium.

Abstract: The model equations of the growth phase of rHSA fermentation were derived on the base of both elemental balance and metabolic balance. The unknown parameters of the model were estimated by multivariable optimization. The model can preferably describe the relations between different macroscopic reaction rates of the process and provide the key for the high-density cultivation of Pichia pastoris.

Abstract: Objective] Citrus (Citri) is an economically important fruit crop in the world. The anthracnose caused by Colletotrichum gloeosporioides (Penz.) Sacc. is one of the main diseases in citrus production. For exploring of an effective biocontrol measure against citrus postharvest anthracnose, a bacterial biocontrol strain T132 isolated from the rhizosphere soil of citrus was identified and characterized. The efficacy of the biocontrol strain against citrus postharvest anthracnose disease was evaluated. Methods] Identification of strain T132 was carried out by using 16S rDNA sequence homology comparison as well as morphological, physiological and biochemical characteristics. The antagonistic stability of the strain T132 was determined by continuously transferring it on artificial media for 8 generations. The efficacy of strain T132 in controlling citrus postharvest anthracnose was performed by stab inoculation and naturally infected methods. The use of specific primers detect the strain T132 of the pathogenic factor of Burkholderia cepacia as a potential human pathogens. Results] Strain T132 was identified as Burkholderia vietnamiensis based on its morphological traits, phyiological and biochemical characters as well as 16S rDNA sequence analysis. Less change in the suppressive ability of antagonizing against growth of C. gloeosporioides was observed during 8 generations of transfers on artificial media. The strain T132 had significant efficacy in controlling citrus anthracnose disease causing by C. gloeosporioides. The average efficacies of the strain T132 in controlling the disease were 88.2% in artificial inoculation test and 54.9% in natural infection test 20 d and 60 d after of the antangonist applications, respectively. It was no virulence gene of Burkholderia cepacia epidemic strain marker (BCESM) existed. Conclusion] This is the first report of B. vietnamiensis as a biocontrol agent against citrus postharvest anthracnose disease and the relative safety of biocontrol strains on human.
The 16S rRNA Gene Sequences and Growth Characteristics of Two Nitrogen Fixing Endobacteria Strains from Sugarcane Stalk

Abstract: Two N2-fixing isolates, B11S and B8S, were obtained from the surface-sterilized stalks of sugarcane variety GT11and RB86-7515 which were bred in Guangxi, China and introduced from Brazil, respectively. Both isolates showed acetylene reduction in nitrogen free media. Classification of both B11S and B8S was made on the basis of 16S rRNA sequence analysis, and the biological characteristics of the two isolates were studied. The 16S rRNA sequence of the isolate B11S had a 98.5% sequence similarity to those of many Stenotrophomonas maltophili and the 16S rRNA sequence of the isolate B8S had a 100% similarity to that of Agrobacterium tumefaciens (Rhizobium radiobacter). For both growth and nitrogenase activity of the isolates, the optimum temperature was about 31°C. The optimum pH was about 6.0 for their growth but 6.5~7.0 their nitrogenase activity. The two strains propagated more quickly and showed higher nitrogenase activity with the increase of carbon concentration in the culture media, and the effect of sucrose was better than that of the glucose. The growth and nitrogenase activity could be detected when the two strains were cultured in the media containing different concentration NH4+ and NO3-, and they grew better and showed higher nitrogenase activity under suitable nitrogen contents, but the growth of the two isolates was hindered more and more strongly with the increasing of the nitrogen content.

Abstract: On the base of element and metablism balancing, the mathematical model of the cultivation process with Streptomyces aureofaciens was developed, and the unknown parameters in the model were estimated with the method of nonlinear optimization. Firstly the energetic coefficient of CTC biosynthesis was gained, which was 1.8 - 2.8 mol-ATP x C-mol(-1). The macroscopic reaction rates were predicted in the process and compared with the experimental values. The results show that the model can preferably describe the relationships between several macroscopic reaction rates in the process and can supervise the optimization of CTC fermentation process theoretically.

Abstract: Citrus bacterial canker is an important disease of Citrus species in China. The disease severely occurs especially in the coastal area. Integrated control system has been used for the control of the disease, in which chemotherapy plays an important role at present. The chemotherapy-dominant control system brought many problems to the environment, such as chemical residua in the products and induction of resistance to fungicide(s) by the pathogen. To solve these problems, an intensive study on biocontrol of citrus bacterial canker is needed. Isolations and characterizations of biocontrol agents are the basis for biocontrol of the disease. A bacterial strain Bt8 with strong inhibiting ability against Xanthomonas axonopodis pv. citri (Hasse) Vauterin, was isolated from citrus orchard soil in Nanning, China. The isolated bacterial strain was identified and characterized as Acinetobacter baumannii Bouvet et Grimont on the base of its morphology and 16S rDNA sequence analysis as well as physiological and biochemical characters. The inhibiting activity of the bacterium suspension against the pathogen was significantly influenced by environmental factors, such as temperatures, pHs and media. At temperatures of 18 degrees C to 33 degrees C, both the inhibiting activity of the bacterium suspension and the biomass of the bacterium increased with the increases of temperatures, suggesting that the influence of temperature on inhibiting activity of the bacterium suspension was in dependence on the bacterial biomass. In NA liquid medium of pH 10, the bacterium suspension showed the highest inhibiting activity against Xanthomonas axonopodis pv. citri, which was not in dependence on biomass of the bacterium. The bacterium suspension provided 55.2% inhibition against bacterial canker under greenhouse conditions. The results showed that Acinetobacter baumannii has potential as biocontrol agent against bacterial canker disease. Acinetobacter baumannii was reported as the pathogens infecting human and animals. The present study enriched the understanding on biological diversity in Acinetobacter baumannii to sciences. This is the first report on the isolation of Acinetobacter baumannii with strong inhibiting ability against plant pathogen.

Abstract: 为进一步明确香蕉叶斑病菌喙突脐蠕孢的生物学特性,调查了部分环境因素(碳氮源、植物成分培养基)对供试菌株(CLER09、D087和JL05)的营养生长和产孢的影响。结果显示,分生孢子主要先从基部萌芽,在28℃下约培养10h后出现两端萌芽;供试菌株对测试的20种碳源及26种氮源显示相似的生长反应,均可利用除菊糖外的测试碳源及氮源进行营养生长和产孢;测试的6种植物成分培养基对供试菌株的营养生长效果皆优于PDA培养基;除燕麦培养基外,其余5种培养基对菌株CLER09和D087的产孢作用均优于PDA培养基;测试的6种培养基对菌株JL05的产孢作用均优于PDA培养基。不同植物成分培养基对该菌分生孢子形态影响较大,以米糠、象草培养基对菌株D087 和JL05的分生孢子长度和玉米粉培养基对菌株CLER09的分生孢子宽度的增长效果最为明显。

Parameter Estimation of Ground Moving Targets Based on SKT-DLVT Processing

Abstract: It is well known that the motion of a ground moving target may induce the range cell migration, spectrum spread and velocity ambiguity during the imaging time, which makes the image smeared. To eliminate the influence of these factors on image focusing, a novel method for parameter estimation of ground moving targets, known as SKT-DLVT, is proposed in this paper. In this method, the segmental keystone transform (SKT) is used to correct the range walk of targets simultaneously, and a new transform, namely, Doppler Lv's transform (LVT) is applied on the azimuth signal to estimate the parameters. Theoretical analysis confirms that no interpolation is needed for the proposed method and the targets can be well focused within limited searching range of the ambiguity number. The proposed method is capable of obtaining the accurate parameter estimates efficiently in the low signal-to-noise ratio (SNR) scenario with low computational burden and memory cost, making it suitable to be applied in memory-limited and real-time processing systems. The effectiveness of the proposed method is demonstrated by both simulated and real data.
A New Parameter Estimation Algorithm Based on Sub-band Dual Frequency Conjugate LVT

Abstract: A new parameter estimation algorithm, known as Sub-band Dual Frequency Conjugate LVT (SDFC-LVT), is proposed for the ground moving targets. This algorithm first constructs two sub-band signals with different central frequencies. After that, the two signals are shifted by different values in frequency domain and a new signal is constructed by multiplying one with the conjugate of the other. Finally, Keystone transform and LVT operation are performed on the constructed signal to attain the estimates. The cross-term and the performance of the proposed method are analyzed in detail. Since the equivalent carrier frequency is reduced greatly, the proposed method is capable of obtaining the accurate parameter estimates and resolving the problem of ambiguity which invalidates Keystone transform. It is search-free and can compensate the range walk of multiple targets simultaneously, thereby reducing the computational burden. The effectiveness of the proposed method is demonstrated by both simulated and real data.
