Effect of recipes replenishing qi and activating blood on cell proliferation and apoptosis in the liver of aging rats

Abstract: Objective: To observe the effect of recipes replenishing qi and activating blood on cell proliferation and apoptosis in the liver of natural aging rats. Methods: Natural aging rats were under administration of recipes replenishing qi or activating blood for 4 months. The liver of the rats was prepared into cell suspension for determination of cell proliferation and apoptosis with PI-staining and flow cytometer. Results: (1) Compared with those of the young rats, the cells in G0-G1 phase in the liver tissue of aging rats were increased (P<0.01), and apoptosis cells were increased (P<0.01), while the cells in S and G2-M phases were decreased(P<0.01). (2) Compared with those of the aging rats, the cells in G0-G1 phase in the liver tissue of aging rats administered recipes replenishing qi or activating blood were decreased (P<0.01), and it was more obvious in activating blood group than in replenishing qi group (P<0.01); the cells in S and G2-M phases were increased (P<0.01), and there was no significant difference between the activating blood group and the replenishing qi group (P>0.05). (3) The apoptosis cells in replenishing qi or activating blood group were decreased significantly (P<0.01), and the effect of replenishing qi was better than that of activating blood (P<0.01). Conclusion: (1) Cell proliferation is decreased and apoptosis is increased in the liver tissue of natural aging rats. (2) Recipes replenishing qi or activating blood can accelerate cell proliferation in the liver tissue of natural aging rats,and the effect of activating blood was slightly stronger than that of replenishing qi. (3) Recipes replenishing qi or activating blood can inhibit cell apoptosis in the liver tissue of natural aging rats,and the effect of replenishing qi was better than that of activating blood.

Abstract: A new method is proposed for determining the backscattering factor using information provided by the background off an direct Auger spectrum N(E). Complicated calculations such as the Monte Carlo method were avoided, and the quantitative Auger analysis accuracy could be further improved since the calculations of backscattering factor was based on the spectra. The examples of determining the compositions of Au-Cu and Ag-Cu alloys are given. These results are compared with that obtained by other mothods.

Identification and target prediction of miRNAs specifically expressed in rat neural tissue

Abstract: In order to obtain as complete a picture of specific miRNA expression in rat neural tissues as possible, customized miRNA microarrays with 152 selected miRNAs from miRBase were used to detect miRNA expression in 14 rat tissues. After a general clustering analysis, 14 rat tissues could be clearly classified into neural and non-neural tissues based on the obtained expression profiles with p values < 0.05. The results indicated that the miRNA profiles were different in neural and non-neural tissues. In total, we found 30 miRNAs that were specifically expressed in neural tissues. For example, miR-199a was specifically expressed in neural tissues. Of these, the expression patterns of four miRNAs were comparable with those of Landgraf et al., Bak et al., and Kapsimani et al. Thirty neural tissue-specific miRNAs were chosen to predict target genes. A total of 1,475 target mRNA were predicted based on the intersection of three public databases, and target mRNA's pathway, function, and regulatory network analysis were performed. We focused on target enrichments of the dorsal root ganglion (DRG) and olfactory bulb. There were four Gene Ontology (GO) functions and five KEGG pathways significantly enriched in DRG. Only one GO function was significantly enriched in the olfactory bulb. These targets are all predictions and have not been experimentally validated.Our work provides a global view of rat neural tissue-specific miRNA profiles and a target map of miRNAs, which is expected to contribute to future investigations of miRNA regulatory mechanisms in neural systems.MiRNAs are a large class of tiny non-coding RNAs (~22 nt long). Abstract: A simple method has been developed in our laboratory, namely, lonization Loss Spectros-copy (ILS) for unoccupied DOS measurement which can be performed with existing Auger electron spectroscopy apparatus.The results of spin-orbit splitting and binding energy obtained by ILS are in excellent agreement with that of ESCA. The ILS spectra of 3d transition metals, Ti, Cr, Ni, Cu have been measured. The typical integral spectra exhibit a peak-like shape with different intensity and width which reflects the width of the unoccupied DOS. Since the unoccupied 3d DOS of Cu is zero, the ILS spectrum of Cu measured is shown by a step-like structure resulting from the flat, narrow 4s DOS.

Abstract: The surface phenomena of Pb97.4Sn2.6 alloy under the Ar+ ion sputtering have been investigated using PHI-590 scanning Auger microprobe at room temperature. Theoretical analyses of experimental data showed that the ion-bombardment-enhanced diffusion occured with diffusion coefficient up to 10-15 cm2/s and the thickness of alter layer was about 260 ?. The possible explanation is given.

Abstract: Leprosy is a disabling chronic infection, with insidious onset that often evades early detection. In order to detect new leprosy cases in a timely manner, we conducted surveillance visits in some difficult-to-reach mountain areas in South West China where the disease is still prevalent. Our data confirm that Chinese multibacillary (MB) leprosy patients have strong antibody responses against Mycobacterium leprae antigens ND-O-BSA and LID-1. Contacts of clinically diagnosed patients were then monitored at regular intervals by both physical examinations and the laboratory determination of antibody responses in sera collected during these examinations. Elevations in antibody titers indicated the onset of MB leprosy in one of the contacts, and diagnosis was subsequently confirmed on physical examination. Our data indicate that rising antibody titers can be used as a trigger for physical examination or increased monitoring of particular individuals in order to provide early leprosy diagnosis. 1. Introduction Leprosy is the clinical manifestation of infection with Mycobacterium leprae. ?With a slow growth rate of division every 12-13 days at an optimum temperature of 30°C, M. leprae bacteria infect macrophages and Schwann cells [1]. Infection of Schwann cells predisposes infected individuals to nerve damage, rendering leprosy, a chronic disabling infection with insidious onset typically characterized by the early involvement of skin and peripheral nerves. The immune response of the leprosy patient shapes the clinical presentation [2]. The response can limit bacterial growth and dissemination, resulting in few localized skin lesions and the definition of such patients as paucibacillary (PB). In cases where the response does not control bacterial growth, high bacterial burden arises, infection becomes systemic and many disseminated lesions and significant nerve function impairment can be observed; these patients are defined as multibacillary (MB). If such cases are left untreated, leprosy can progress to disfigurement and disability. Although early detection is recommended by World Health Organisation (WHO), diagnosis is currently achieved only upon the recognition of clinical symptoms by skilled clinicians [3]. As leprosy is usually associated with low socioeconomic development, patients often have limited access to medical care and evade early detection. Abstract: A universal linear-temperature dependence of the uniform magnetic susceptibility has been observed in the nonmagnetic normal state of iron-pnictides. This non-Pauli and non-Curie-Weiss-like paramagnetic behavior cannot be understood within a pure itinerant picture. We argue that it results from the existence of a wide antiferromagnetic fluctuation window in which the local spin-density-wave correlations exist but the global directional order has not been established yet.
