Viability of Frozen-Thawed Epididymal Sperm of Garut Ram Cryopreserved with Modified Tris Extender

Abstract: Sperm collected from cauda epididymis is a source of male gametes. The purposes of this study was to evaluate an quality of frozen-thawed sperm of garut ram which was collected from cauda epididymis and cryopreserved with modified Tris extender, i.e: Tris extender (control, KT), Tris extender + 60 mM lactose (LS), and Tris extender + 60 mM lactose + 0.05% glutathione (GL). Quality of collected sperm including concentration, motility, live sperm, abnormality, cytoplasmic droplet, intact acrosomal cap (IAC), and intact plasma membrane (IPM) were evaluated. Results showed that mean of sperm concentration, percentages of motility, live sperm, abnormality, cytoplasmic droplet, IAC, and IPM of fresh epididymal sperm were 13,993.33 million/ml, 70.83, 82.83, 10.83, 8.5, 85.83, and 81.33%, respectively. Sperm quality after equilibration for LS and GL were significantly (P<0.05) higher than KT. Mean percentages of post thawing sperm motility, live sperm, IAC, and IPM for GL (45, 54.5, 47.83, and 48.83%) were significantly (P<0.05) higher than LS (40, 49.17, 43.83, and 44.5%), and KT (35, 42.5, 39.17, and 41.5%). Mean percentages of post thawing sperm motility, live sperm, IAC, and IPM for LS were significantly (P<0.05) higher than those of KT. Hence, frozen-thawed epididymal sperm of garut ram after slaughter and cryopreserved with Tris extender + 60 mM lactose (LS) and Tris extender + 60 mM lactose + 0.05% glutathione (GL) possibly can be used for artificial insemination (AI) or in vitro embryo production program.

Abstract: It is now ten years since the Council of Europe's grand art exhibition 'Gods and Heroes of the Bronze Age – Europe at the Time of Ulysses' toured Europe as a means to 'increase the awareness of the value and the significance of the archaeological heritage' of Europe (AH 2008). Presenting the 'first golden age' of Europe, the Bronze Age exhibition was one of the first steps towards a more defined European cultural politics. The campaign represented something new in European politics, but the idea of the Bronze Age as a golden age in European history is not new. This article draws attention to the archaeological heritage which enabled such an idea to be put forward: Childean prehistory.

Abstract: The aim of this research was to observe the quality of spotted buffalo epididymal sperm after storage at 4 °C as liquid semen for 12 and 24 hours in andromed containing sucrose 0.2% and 0.4%. Sperm was collected from epididymal tissues by combination of slicing and pressing methods. The parameters observed were the percentage of progressive motility and live sperm. The results showed that the percentage of progressive motility of the liquid semen after storage for 12 hours in andromed (A), andromed + sucrose 0.2% (P1) and andromed + sucrose 0.4% (P2) were 48.33%; 53.33% and 55% (P>0.05), respectively. In addition, the percentage of motility after 24 hours of storage in those extenders were 45%; 46.67% and 46.67% (P>0.05), respectively. The percentage of live sperm in A, P1 and P2 after 12 hours of storage were 70.33%; 73% and 73.33% while after 24 hours of storage, the percentage of live sperm in those extenders were 66.33%; 68.67% and 69.67%, respectively. In conclusion, the addition of 0.2% and 0.4% sucrose in andromed extender could maintain the quality of the spotted buffalo epididymal sperm after storage for 12 and 24 hours at 4 °C.

Tre fokusgrupper best ende av veiledere som sykepleierstudentene forholder seg til i h gskole og i praksisstudier er blitt intervjuet om helsefremming. Empirien viser at helsefremming i hovedsak knyttes til helseopplysning og forebyggelse av sykdom. Dessuten sp rres det om det er hensiktsmessig skille mellom helsefremming og forebygging. Det argumenteres for at det trengs en bevisstgj ring av hva helsefremming i tr d med Ottawacharteret kan bety i sykepleien.
PRESERVATION OF GARUT RAMS SPERMATOZOON AS A SOURCE OF MALE GERM PLASM

Abstract: This study was conducted to examine the quality of ejaculated sperm by Garut rams to be used for artificial insemination (AI) and viability of sperm that were collected from preserved cauda epididymis (4°C up to 12 days) for assisted reproductive technology. The semen was collected by artificial vagina, with the sperm motility, live sperm, acrosomal intact, and intact plasma membrane observed. Sperm motility was 75%, while for the live sperm, intact plasma membrane and sperm abnormality were 91.5%, 90.0%, and 1.8%, respectively. In the other study, sperm was collected from cauda epididymis by aspiration method and diluted in different media: 1) Brackett Oliphant (BO) media and 2) modified Phosphate Buffer Saline (mPBS). Evaluation of sperm motility and intact plasma membrane were conducted after washing, counting and dilution of the sperm. The results of this study showed that the sperm motility and intact plasma membrane could be maintained better in BO rather than PBS medium although they were not statistically different (P>0.05). At day 12 of preservation, the motility and intact plasma membrane of sperm collected from cauda epididymis were 0.7% and 1.33% for motility and plasma membrane intact, respectively. These findings showed that the Garut rams semen was qualified for AI and frozen processing; in vitro embryo production by introducing the assisted reproductive technology such as intracytoplasmic sperm injection (ICSI) could be applied by using the sperm collected from preserved cauda epididymis until 12 days of preservation at 4°C.
Viabilility of Rams Epididymal Sperm after Preservation in Low Temperature (5oc)

Abstract: The purpose of this research was to evaluate the viability of ram epididymal sperm collected from fresh caudal epididymis (H-0) or after storage in low temperature (5oC, in refrigerator) for one (H-1), two (H-2), and three (H-3) days. Collected sperm were diluted in modified Tris extender and they were preserved in refrigerator up to four days. The viability of diluted sperm was evaluated daily base on motility and sperm live. Results indicated that mean sperm concentration after sperm diluted with 0.05 ml Tris extender of caudal epididymis was 2745 million/ml. Sperm motility and percentage of live for H-0 (71.25% and 82.83%) and H-1 (70.00% and 79.17%) were significantly higher (P<0.05) than H-2 (61.25% and 69.83%) and H-3 (51.67% and 66.17%). Percentages of sperm motility and live of diluted sperm and preserved in refrigerator for H-0 were significantly higher (P<0.05) than H-1, H-2, and H-3. These results showed that epididymal sperm collected from caudal epididymis up to three days of preservation (without further storage of the diluted sperm) could be used for artificial insemination or in vitro fertilization programs. Abstract: h e objective of this study was to obtain the viability and plasma membrane integrity of the spotted buff alo epididymal sperm after addition of dextrose into Andromed extender. Spermatozoa that have been collected from cauda epididymis were diluted with Andromed extender as control (K) and Andromed + 0.2% dextrose (P1) and Andromed + 0.4% dextrose (P2) as treatments. h e results showed that the quality of epididymal spermatozoa decreased during cryopreservation process. h e percentage of motility after thawing in P1 (46%) and P2 (46.67%) were signifi cantly higher (P<0.05) compared to K (41%) as well as the percentage of live sperm in P1 (58.8%) and P2 (60%) compared to K (52.2%). h e percentage of membrane integrity in P1, P2 and K were 67.4; 66.8 and 68 %, respectively. In conclusion, the addition of 0.2 and 0.4% of dextrose into Andromed acted as an extra cellular cryoprotectant and could maintain the viability and membrane integrity of the spotted buff alo epididymal spermatozoa after thawing.
Optimalization of Equilibration and Thawing Methode on Freezing Process of Garut Ram Semen (Ovis aries)

Abstract: Generally, the problem of semen freezing process is cold shock effect and intracelullar change due to water release which is related to ice crystal formation. One factor to solve the problem is finding out optimal equilibration time and thawing method so there is only a little damage spermatozoa during freezing process. The research has been done to explore the influence of equilibration time and thawing methode on frozen semen quality of garut rams. The results shown that the mean of percentages of progressive motile sperm, percentages of viable sperm, percentages of plasma membrane and acrosomal intact on four hours equilibration (52.50% ; 62.33%; 57.17% and 56.42%) were not significantly different (P>0.05) from five hours equilibration (52.27%; 63.67%; 56.92% and 57.58 %) and six hours equilibration (54.17%; 61.00%; 59.42% and 58.58%) respectively. The percentages of progressive motile sperm, percentages of viable sperm, percentages of plasma membrane and acrosomal intact on the thawing method on the temperature of 370C for 30 seconds (53.33% ; 62.39%; 57.94% dan 58.61) were not significantly different (P>0.05) from the thawing methode on the temperature of 250C for 45 seconds (52.22% ; 62.28%; 57.72% dan 56.44) respectively. The conclusion shown that the treatment of four hours equilibration, five hours equilibration, six hours equilibration and also the treatment of thawing method on the temperature of 370C for 30 seconds and the thawing methode on the temperature of 250C for 45 seconds do not have effect on garut ram freezing semen quality. Estimating domestic wood burning emissions of particulate matter in two Nordic cities by combining ambient air observations with receptor and dispersion models

Abstract: The major emission source of primary PM2.5 in many Nordic countries is wood burning for domestic heating. Though direct measurements of wood burning emissions are possible under controlled conditions, emission inventories for urban scale domestic heating are difficult to calculate and remain uncertain. As an alternative method for estimating these emissions, this paper makes use of ambient air measurements, chemical analysis of filter samples, receptor models, dispersion models, and simple inverse modelling methods to infer the emission strengths. A comparison of dispersion models with receptor models indicates that the dispersion models tend to overestimate the contribution from wood burning. The inverse modelling results are found to agree with those from the receptor modelling. Though both the receptor and inverse modelling point to an overestimation of the wood burning emissions of PM2.5, it is not possible to assign this solely to errors in the emissions inventory as a dispersion model error can be significant. It is recommended to improve plume rise and urban canopy meteorological descriptions in the dispersion models before these models are of sufficient quality to allow quantitative assessments of emission inventories.
Doubtful outcome of the validation of the Rome II questionnaire: validation of a symptom based diagnostic tool

Abstract: Volunteers from a population based colonoscopy study were included (n = 1011), together with patients seeking general practice (n = 45) and patients visiting a gastrointestinal specialists' clinic (n = 67). The questionnaire consists of 38 questions concerning gastrointestinal symptoms and complaints. Diagnoses are made after a special code. Our validation included analyses of the translation, feasibility, predictability, reproducibility and reliability. Kappa values and overall agreement were measured. The factor structures were confirmed using a principal component analysis and Cronbach's alpha was used to test the internal consistency.Translation and back translation showed good agreement. The questionnaire was easy to understand and use. The reproducibility test showed kappa values of 0.60 for GERS, 0.52 for FD, and 0.47 for IBS. Kappa values and overall agreement for the predictability when the diagnoses by the questionnaire were compared to the diagnoses by the clinician were 0.26 and 90% for GERS, 0.18 and 85% for FD, and 0.49 and 86% for IBS. Corresponding figures for the agreement between the printed and the digital version were 0.50 and 92% for GERS, 0.64 and 95% for FD, and 0.76 and 95% for IBS. Cronbach's alpha coefficient for GERS was 0.75 with a span per item of 0.71 to 0.76. For FD the figures were 0.68 and 0.54 to 0.70 and for IBS 0.61 and 0.56 to 0.66. The Rome II questionnaire has never been thoroughly validated before even if diagnoses made by the Rome criteria have been compared to diagnoses made in clinical practice.The accuracy of the Swedish version of the Rome II is of doubtful value for clinical practice and research. The results for reproducibility and reliability were acceptable but the outcome of the predictability test was poor with IBS as an exception.
