Eficacia y toxicidad de los antimoniales pentavalentes (Glucantime? y Pentostam?) en un modelo animal de leishmaniasis cutánea americana: aplicación de la luminometría

Abstract: the pentavalent antimonial compounds glucantime? and pentostam? are the first line drugs used in anti- leishmania treatment. however, no in vivo studies have compared the efficacy and toxicity of these drugs where host variability has been controlled. biochemical studies of leishmania have detected differences between the two drugs with regard to dna topoisomerase i inhibition, a phenomenon that possibly impacts treatment efficacy. to evaluate the clinical efficacy, hamsters were infected intradermally in the right hind foot with 106 promastigotes of a wild type or luciferase-transfected leishmania panamensis. at three weeks post-inoculation, the animals were treated intramuscularly with either glucantime? or pentostam? (30, 60 or 120 mg sbv/kg per day for 20 days). to evaluate parasitological efficacy a luminometry assay was standardized for quantitation of amastigotes in hamster tissues. to evaluate toxicity, hamsters were treated intramuscularly with glucantime? or pentostam? (120, 160 or 240 mg sbv/kg per day for 20 days). animals inoculated with either of the parasite strains and treated with either drug, showed a similar rate of lesion reduction, as compared to untreated controls ( p<0.01). parasite burden was also comparable, and no significant differences were found in the cure rate. no renal or hepatic alterations occurred as evidenced by normal serum levels of creatinine, aspartate aminotransferase, alanine aminotransferase and amylase. hamsters treated with 120 mg sbv/kg per day for 20 days or higher doses of pentostam? showed macro- and microscopic signs of inflammation at the site of injection. these effects were absent in the animals treated with glucantime?. the results confirmed clinical observations regarding the similar efficacy of the two drugs, as well as the higher local toxicity of pentostam?.

Abstract: effects of the substrates agrolite, vermiculite, tezontle as substitutes for agar were evaluated on seed germination, shoot proliferation, rooting, and survival of the seedlings of the citrus ?tristeza? virus (ctv) tolerant rootstocks citrus volkameriana, citrumelo swingle and citrange 35. the medium used was murashige and skoog (ms), which for shoot proliferation was supplemented with 3% saccharose, vitamins, and 1mg·l-1 bap. rooting was achieved using half strength of ms medium and saccharose, supplemented with 2.5mg·l-1 of iba and vitamins. seed germination, shoot proliferation, and rooting were 96-100, 95-100 and 93-100%, respectively, in the three substrates and did not differ from the value of 95-100% in agar (p>0.05). survival was higher (p<0.05) from seedlings obtained in the substrates (91-100%) than in the agar (48-51%). the substrates evaluated can be used instead of agar as a support in tissue culture, being less expensive and increasing seedling survival.

Abstract: the use of the sugars xylose, fructose and 10x20f, as substitutes for sucrose, was evaluated in terms of effects on the number and height of shoots, number of leaves, roots per shoot, and rooting shoots in rootstocks of the citrus ?tristeza? virus (ctv) resistant lemon varieties citrus volkameriana, citrumelo swingle and c-35. the explants were cultured in vermiculite and in agar in order to concurrently compare the effects of these different culture media. an average of two shoots per explant was observed in all three species in all three carbohydrate treatments. shoot height, number of leaves per shoot and number of roots in both culture media, were not found to be significantly different (p<0.05). a higher percentage of rooting shoots was observed in the sucrose and 10x20f treatments in both culture media (99-100%). treatments utilising xylose and fructose showed a response of 75-89% rooting. the results showed that neither carbohydrate source nor culture medium had a significant effect on in vitro multiplication, but that shoot height was higher in the rootstock of c. volkameriana
A novel HPV 16 L1-based chimeric virus-like particle containing E6 and E7 seroreactive epitopes permits highly specific detection of antibodies in patients with CIN 1 and HPV-16 infection

Abstract: The sera of 30 patients with CIN 1 who also tested positive for HPV-16 DNA and of 30 age-matched normal donors negative for HPV infection were tested for the presence of IgG antibodies specific for either VLP-L1 (HPV-16 L1), gVLP (derived from Gardasil), or cVLP by ELISA. The cVLP-reactive sera yielded two distinct groups of results: (H) reactivity levels that presented very strong cVLP-specific titers, and (L) reactivity levels with significantly lower titers similar to those obtained with VLP-L1 and gVLP antigens. Additionally, the sera that presented the higher cVLP titers closely matched those that had significantly stronger reactivity to E6 and E7 epitopes. Interestingly, the samples with the highest titers corresponded to patients with the higher numbers of sexual partners and pregnancies. On the other hand only 4 out of the 12 sera that harbored antibodies with VLP neutralizing ability corresponded to the group with high cVLP antibody titers.We report for the first time that chimeric particles containing HPV-16 L1 protein fused with E6 and E7 seroreactive epitopes enable much better detection of IgG antibodies in the sera of CIN 1 patients positive for HPV-16 infection than those obtained with VLPs containing only the HPV-16 L1 protein. We also found that the sera with higher cVLP antibody titers corresponded to patients with more sexual partners and pregnancies, and not always with to those with a high neutralizing activity. This novel assay could help in the development of a tool to evaluate cervical cancer risk.Infection of the genital epithelium with human papillomavirus (HPV) is a common sexually transmitted disease, as well as a significant public health burden in developing countries. Abstract: hardness of edam cheese through penetrometry technique and texture profile analysis (tpa) was studied. the statistical results indicated that product hardness increases with ripening time but its values depends upon the measurement technique (p<0,05). lineal regression analysis showed a relation of 85 % between two methods where the values of hardness obtained using penetrometry by sphere are underpredicted with respect to values reached with texture profile analysis technique.

Abstract: Se estudió la dureza del queso Edam a través de la técnica de penetrometría y análisis de perfil de textura (TPA). Los resultados estadísticos indicaron que la dureza del producto incrementó con el tiempo de maduración, pero estos valores dependen de la técnica de medición (P < 0,05). El análisis de regresión lineal mostró una relación del 85 % entre los dos métodos, donde los valores de dureza obtenidos utilizando el penetrómetro por esfera son subestimados con respecto a los valores obtenidos con la técnica del análisis de perfil de textura. Effective quantum equations for the semiclassical description of the Hydrogen atom

Abstract: We study the Hydrogen atom as a quantum mechanical system with a Coulomb like potential, with a semiclassical approach based on an effective description of quantum mechanics. This treatment allows us to describe the quantum state of the system as a system of infinite many classical equations for expectation values of configuration variables, their moments and quantum dispersions. It also provides a semiclassical description of the orbits and the evolution of observables and spreadings and their back-reaction on the evolution.
