Unusual Detection of Lathosterol in Amniotic Fluids Investigated for the Determination of Cholesterol and 7-Dehydrocholesterol for Suspected Smith-Lemli-Opitz Syndrome  [PDF]

Abstract: The Smith-Lemli-Opitz syndrome (SLOS), is an autosomal recessive disorder caused by a 7-dehydrocholesterol reductase deficiency, which is characterized by abnormally elevated amniotic fluid 7-DHC (7-dehydrocholesterol) concentrations. A GC/FID (gas-chromatography with a flame ionization detector) and GC/MS (gas-chromatography with a mass detector) method was optimized for the detection of cholesterol and 7-DHC in amniotic fluids. The quantitative determination of cholesterol in 39 control amniotic fluids evidenced that between the fourth and fifth month of pregnancy the levels of this analyte are quite constant, the concentration of total and free cholesterol being respectively 10.3 μg·mL﹣1 (SD = ±3.6) and 1.7 μg·mL﹣1 (SD = ±0.91), while the analysis of 60 amniotic fluids potentially related to SLOS, showed a higher variability of cholesterol levels. Moreover, in 13 samples an analyte which did not correspond either to cholesterol or to 7-DHC was detected. A GC/MS investigation allowed us to identify this compound as lathosterol, a precursor of cholesterol in the biosynthetic pathway.
Scaling cigars in the Cuban tourism economy

Abstract: taking scale as an object of analysis (tsing 2000), this article examines how competing scale-making projects emerge in different layers of the cuban tourism economy. i consider how tourists and cubans interact along contested chains of production and distribution of cigars, and therefore become entangled in bundles of ideas, discourses, practices and materialities. these constitute different scaling propositions on which tourists and cubans converge or diverge, and which inform the success of encounters within the tourism economy. highlighted is the importance of understanding how scaling propositions are enacted and negotiated within tourism. i conclude that the realm of tourism is a particularly fruitful platform to investigate how competing scale-making projects and notions of scale are played out in the contemporary world.

Abstract: An electrolysed solution of sodium chloride was examined for its disinfection potential in drinking and bathing water. The electrolysis of NaCl ((=1%) in tap water was performed at room temperature using a 10 A electric current in an electrolysis apparatus. Some laboratory tests were made, initially to determine the stability and efficiency of the disinfectant. Chemical and microbiological measurements of the treated water (according to DIN 19643 for bathing water) were then carried out before and after addition of the disinfectant agent.

With this powerful geometric tool of complex functions in one variable, we are able to prove some theorems concerning the size of images under holomorphic mappings, including the celebrated Picard's theorems. The article concludes with a brief insight into the theory of Kobayashi hyperbolic complex manifolds.
High Doses of Ursodeoxycholic Acid Up-Regulate the Expression of Placental Breast Cancer Resistance Protein in Patients Affected by Intrahepatic Cholestasis of Pregnancy

Abstract: Background Ursodeoxycholic acid (UDCA) administration in intrahepatic cholestasis of pregnancy (ICP) induces bile acids (BA) efflux from the foetal compartment, but the molecular basis of this transplacental transport is only partially defined. Aim To determine if placental breast cancer resistance protein (BCRP), able to transport BA, is regulated by UDCA in ICP. Methods 32 pregnant women with ICP (14 untreated, 34.9±5.17 years; 18 treated with UDCA - 25 mg/Kg/day, 32.7±4.62 years,) and 12 healthy controls (33.4±3.32 years) agreed to participate in the study. Placentas were obtained at delivery and processed for membrane extraction. BCRP protein expression was evaluated by immunoblotting techniques and chemiluminescence quantified with a luminograph measuring emitted photons; mRNA expression with real time PCR. Statistical differences between groups were evaluated by ANOVA with Dunn’s Multiple Comparison test. Results BCRP was expressed only on the apical membrane of the syncytiotrophoblast. A significant difference was observed among the three groups both for mRNA (ANOVA, p = 0.0074) and protein (ANOVA, p<0.0001) expression. BCRP expression was similar in controls and in the untreated ICP group. UDCA induced a significant increase in placental BCRP mRNA and protein expression compared to controls (350.7±106.3 vs 100±18.68% of controls, p<0.05 and 397.8±56.02 vs 100±11.44% of controls, p<0.001, respectively) and untreated ICP (90.29±17.59% of controls, p<0.05 and 155.0±13.87%, p<0.01). Conclusion Our results confirm that BCRP is expressed only on the apical membrane of the syncytiotrophoblast and show that ICP treatment with high dose UDCA significantly upregulates placental BCRP expression favouring BA efflux from the foetal compartment.

Abstract: over the past few decades a process of construction and deconstruction of the concept of old age has been noticed in the western societies, a theme which has proved to be an object of further investigation and reflection in several fields of knowledge. old age has equally become an object of increasing intervention, which is generated among different agencies and agents pertaining to the same trend. thus, the production of legitimate discourses concerning old age has been followed by a number of actions that have been organized on some basic assumptions. in the academic field such a tendency has given rise to geriatricians, specialized doctors, and gerontologists, specialized non-doctors, who have been both legitimated by the institutions they create, such as the prestigious sociedade brasileira de geriatria e gerontologia (sbgg), which holds scientific knowledge about old age. this paper presents some of the issues, which have arisen in a wider project on age categories, concerning the discourse and praxis of geriatricians and gerontologists. such discourse and praxis are related to family topics in an attempt to clarify some of their non-explicit assumptions, which guide their actions.
