Optimization of the Sample Preparation Method for DNA Sequencing

Abstract: This study reports on the modification and optimization of the sample preparation method for analyzing nucleotide sequence in plant species by capillary electrophoresis. The experimental study was aimed at developing possible ways of increasing the throughput of the method. The optimization conditions modified were purification of the PCR product, concentration adjustment of cycle sequencing kit BDT v3.1 from half to quarter reactions for economic analysis. Concentrations of the tamplet, primer and program of thermocycler were also optimized. Finally the volume of Hi-Di form amide was adjusted for the required sequencing. The accuracy of analysis was greater than 99.99%. The modified method enables quality sequencing results with higher average signal intensities compared to the standard recommended method. With the optimized electrophoresis conditions reported here, greater than 700 base pairs can be analyzed within 75 min using POP7. It was found that deviation from the recommended method consistently delivers higher signal-to-noise ratios and signal intensities and longer Phred20 read lengths. It is more reproducible than that of other methods and enables rapid, more cost effective robustness for nucleotide sequencing in the samples of plant origin received from research laboratory sources.

The two editorials by Jerome Kassirer and Joel Lexchin are outstanding and deeply thought provoking. The article by Martin Van Der Weyden is equally rewarding and has very useful information. The main monograph has been superbly, very competently, and comprehensively written by both of you.In the series "What Medicine Means To Me," I especially liked the piece by Helen Herrman. Her message of public health psychiatry is particularly relevant for developing countries.The book review of David Healy's book by Leemon McHenry is very refreshing and challenging. I congratulate you on your decision to publish it when other journals were still hesitating.And lastly, the obituary reference to my dear friend Dr. Ravi Kapur by Ajit Bhide and you were so touching; I cried all over again.So keep up your good work, dear friends. Heart geometry in rats under diabetes mellitus – the role of individual reactivity factors

Abstract: In 76 rats with alloxan diabetes the relation between heart remodeling and sensitivity to angiotensin II was established. After 14 days, 1, 2 and 3 months the sensitivity of Angiotensin1 receptors was estimated by in vitro test with induced aggregation of platelets. The body and heart mass, volume and thickness of heart ventriculi were measured. The individual peculiarities of Angiotensin1 receptors expression and their role in determination of heart state during diabetes were shown. The hypersensitivity of Angiotensin1 receptors was associated with early myocardial hypertrophy with next dilatation of ventriculi with decrease of their wall thickness.
Cardiovascular mortality: quality diagnostics analysis and causal statistics of lethal outcomes

Abstract: The high cardiovascular mortality (CVM) proves the necessity of verification analysis of cardiovascular death causes. The research goal is to determine the quality, facilities and peculiarities of cardiovascular diseases (CVD) of postmortem diagnostics and statistical recording as a cause of death. 1972 successive cases of CVM in Voronezh, Ryazan and Khanty-Mansiysk have been analyzed. The majority of deaths caused by CVD were classified as fatalities occurring outside hospital and in the absence of medical care (88.0%). Incidence of cardiovascular deaths was confirmed by autopsy in 28.3% cases. With increasing age, the incidence of detecting CVD as a cause of death increased, while the frequency of referral to autopsy consistently decreased. Chronic Ischemic Heart Disease (IHD) forms comprising 48.3% of all CVM were confirmed by autopsy results only in 29.5% of cases. Acute IHD forms (including myocardial infarction) accounted for only 11.6% of CVM; however this diagnosis was based on autopsy results in 72.4% of cases. The high death rate outside hospital and in the absence of medical care with the rare referral to autopsy, especially for elderly having chronic CVD, does not provide reliable statistical information on CVM
The new theoretical and pathomorphological approaches to the problem of tumor metastasis.

Abstract: The new mechanism of tumor metastasis by penetration of tumor components into vessel adventitia due to vessel rotation was described. Then the tumor cells can travel through the all layers of the vessel and get into the vessel lumen. We suppose that arterial hypertension is important risk factor for the tumor metastasis in oncologic patients. We propose the principle: the carcinogenesis replicates embryogenesis. The new method of metastasis prevention by active stimulation of gastrulas in blastomas on the basis of this principle was suggested. The theories of cytomeres and their activity of tumors are evolved. The cytomeres play important role in the immunological reactions, inflammation, tumor transformation. The high tumor cytomeres activity precedes their metastasis. The algorithm of the pathogenesis of many diseases on the basis of different ways of blocking of extravascular transport paths owing to local information deficiency is offered. Due to information deficiency the "pathogenic field" is formed, inside of which the adaptive morphogenesis can not be implemented because of impossibility of structuring of depressed information streams.

Abstract: the electrochemical data obtained by cyclic voltammetry on several camphor-type (a, b and c) ligands and derived complexes are revised. a direct positive correlation is established between the redox potentials (anodic or cathodic) of the camphor species and the σp-hammett parameter of the r group on the camphor skeleton. a study of the effect of the ligand on the redox properties of some pd, pt or cu camphor complexes shows that the ligands influence either the anodic or cathodic processes but the characteristics of the metal drive the potential values
Financing small rumminant operations along gender lines in Imo State, Nigeria

Abstract: This study analyzed the distribution of ruminant livestock in Imo state, the gender-perceived production constraints; the relative contributions of these ruminants to the farm household net income and the implications of these contributions to loan repayments if production is assigned on gender basis. Results showed that within the ruminant class of livestock, small ruminants, particularly goats, dominate the others, followed by sheep and then large ruminant, cattle. Their relative contribution to total farm household net income follows the same order. Female gender class was found to rear higher number of the small-ruminant livestock. However, a c2 analysis showed that no significant difference exists in their net farm income from these two classes of livestock. It was concluded that financing of these enterprises on gender basis would not significantly improve the contribution of ruminants to farm household net income and loan repayment capacity. It was recommended, among others, that the constraints imposed by factors such as poor marketing and research systems be addressed in order to translate the relative technical advantage of production of small ruminants by female farmers into financial advantage. Response of the Commercial Poultry Sub-Sector to National Policy Regimes in Imo State, Nigeria

Abstract: This study assessed the relative performance of rural commercial poultry farmers in Imo State, between 2000-2003 and 2004-2007 and the influence of the policy instruments implemented within these periods on this performance. Twelve out of the Twenty Seven Local Government Areas in the three Agricultural Zones in the area were chosen for the study through simple random sampling technique. Data were subjected to a multiple regression analysis, structural stability test, costs-returns analysis and the χ2-test. Results showed that the output and profit margins of farmers were significantly higher in 2004-2007 period, leading to the conclusion that the policy instruments employed within this time frame had stronger influence on these performance indices. The χ2-test showed that the returns on investment were dependent on the policy instrument implemented in each period. It was recommended, among others, that these instruments should be relied upon to guide the commercial poultry sub-sector operations in the State.
