Protoplast mutagenesis for improving β-glucosidase production of Aspergillus niger

Abstract: 本研究报道了以原生质体诱变技术选育高产β-葡萄糖苷酶的黑曲霉菌株,并研究了其发酵特性。以黑曲霉CGMCC3.316为出发菌株,通过紫外诱变得到突变株3-3M。然后以3-3M为供试菌株,研究了其原生质体制备与再生的条件。最后通过原生质体诱变,选育得到一株β-葡萄糖苷酶活力较高的突变株60B-3D。该菌株具有良好的遗传稳定性,酶活力平均达到23IU/mL,与出发菌株CGMCC3.316相比提高39%。此外,该菌株的木聚糖酶活力也有所增加。同时考察了黑曲霉60B-3D的发酵特性,并与3-3M和出发菌株进行比较,结果表明该菌株有较高的蛋白分泌能力。本研究为发酵生产β-葡萄糖苷酶提供了一株良好的供试菌株。
A Discrete Velocity Traffic Kinetic Model Including Desired Speed

Abstract: We introduce the desired speed variable into the table of games and formulate a new table of games and the corresponding discrete traffic kinetic model. We use the hybrid programming technique of VB and MATLAB to develop the program. Lastly, we compared the proposed model result and the detector data. The results show that the proposed model can describe the traffic flow evolution.
Neurospora COP9 Signalosome Integrity Plays Major Roles for Hyphal Growth, Conidial Development, and Circadian Function

Abstract: The COP9 signalosome (CSN) is a highly conserved multifunctional complex that has two major biochemical roles: cleaving NEDD8 from cullin proteins and maintaining the stability of CRL components. We used mutation analysis to confirm that the JAMM domain of the CSN-5 subunit is responsible for NEDD8 cleavage from cullin proteins in Neurospora crassa. Point mutations of key residues in the metal-binding motif (EXnHXHX10D) of the CSN-5 JAMM domain disrupted CSN deneddylation activity without interfering with assembly of the CSN complex or interactions between CSN and cullin proteins. Surprisingly, CSN-5 with a mutated JAMM domain partially rescued the phenotypic defects observed in a csn-5 mutant. We found that, even without its deneddylation activity, the CSN can partially maintain the stability of the SCFFWD-1 complex and partially restore the degradation of the circadian clock protein FREQUENCY (FRQ) in vivo. Furthermore, we showed that CSN containing mutant CSN-5 efficiently prevents degradation of the substrate receptors of CRLs. Finally, we found that deletion of the CAND1 ortholog in N. crassa had little effect on the conidiation circadian rhythm. Our results suggest that CSN integrity plays major roles in hyphal growth, conidial development, and circadian function in N. crassa.

Abstract: Hemichorea with corresponding putamenal T1 hyper-intensity and T2 hypointensity on MR imaging has occasionally been reported in diabetes mellitus with nonketotic hyperglycemia. However, the signal intensity in pu-tamenal and cerebellum lesion on MR imaging, which is believed to be pathogenetically related to hemichorea, is rarely documented in diabetes mellitus with nonketotic hyperglycemia. We describe a 57-year-old man with nonketotic hyperglycemic hemichorea on his right arm and legs, whose signal intensity in putamenal and cerebellum lesion was demonstrated by MR imaging.

Abstract: The non-functional QoS (quality of service) information helps us to select a proper Web-service from the web applications, by using component services such as UDDI[1](Universal Description, Discovery, and Integration) and MDS(Monitoring and Discovery System). MDS is a suite of web services to monitor and discover resources and service on Grids, but MDS only based on function aspects. This paper studies on an approach to provide the QoS information and a discovery model by using MDS and gives a system deployment and implementation plan. The simulation results show that the method is effective in service discovery.

In this paper, we investigate the eccentric connectivity index and connective eccentricity index of boron-nitrogen fullerenes, respectively. And we give computing formulas of eccentric connectivity index and connective eccentricity index of all boron-nitrogen fullerenes with regular structure.
Evaluation of Medical College Students’ Entrepreneurial Skills and Its Relationship with Social Intelligence  [PDF]

Abstract: Social intelligence and entrepreneurial skills are important factors influencing individual academic performance and career development. Studies have shown that people with high social intelligence have the ability to understand and manage others and have an advantage in innovation. The entrepreneurial personality trait is precisely the courage to innovate. Therefore, there is a close relationship between entrepreneurship and social intelligence. In this study, the author explores the relationship between students’ social intelligence level and entrepreneurial skills based on medical college students. This study used a cross-sectional descriptive method for related research. The results showed that the overall scores of the entrepreneurial skills assessment criteria of the students were lower. This shows that students’ performance levels of entrepreneurial skills are relatively weak. This result shows that we need to further improve the entrepreneurial skills of medical university students and take the necessary measures to improve students’ social intelligence.

Abstract: A comparison analysis based method for computing the water consumption volume needed for electric energy production of optimal scheduling in hydro-thermal power systems is presented in this paper. The electric energy produced by hydroelectric plants and coal-fired plants is divided into 4 components: potential energy, kinetic energy, water-deep pressure energy and reservoir energy. A new and important concept, reservoir energy, is proposed, based on which is divided into a number of water bodies, for example 3 water bodies, and a reservoir is analyzed in a new way. This paper presents an optimal scheduling solution of elec-tric energy production of hydro-thermal power systems based on multi-factors analytic method, in which some important factors, such as load demand, reservoir in-flow, water consumption volume increment rate of hydroelectric plants or converted from coal-fired plants, and so on are given to model the objective function and the constraints. A study example with three simulation cases is carried out to illustrate flexibility, adapta-bility, applicability of the proposed method.

Abstract: This paper examines Chinese residents’ experience in learning English in the 3-D virtual world of Second Life (SL). With an introduction to the current English as a Second Language (ESL) education in China, ESL students’ demand for practice, I analyzed an online BBS created and maintained by Chinese SL residents and conducted interviews in SL to argue that there is an urgent need among Chinese ESL learners to practice their English with native English speakers and SL can function as a platform to allow Chinese ESL learners practice with residents from all over the world with help and support provided by their online learning communities such as BBS or SL friends groups.

Abstract: The aim of the paper is to offer advice about setting on investigating feasibility of a legal system about the reciprocal recognition of civil case jurisdiction among East Asia region. The present conditions for the recognition of foreign jurisdiction in East Asia states are on the whole similar and this is an advantage for the construction of a unified mechanism. This paper gives three selected models of reciprocal recognition of civil case jurisdiction, which are bilateral judicial assistance treaty, regional multilateral treaties and soften the principle of reciprocity.
