Agrobacterium tumefaciens-mediated transformation of Chaetomium globosum and its T-DNA insertional mutagenesis

Abstract: Agrobacterium tumefaciens-mediated transformation is a routine method in the genetic transformation of a wide range of plant species, and so far, its application has been extended to the transformation of yeasts, fungi and even human cells. By using this transformation system, filamentous fungus Chaetomium globosum was successfully transformed with an efficiency of 60-180 transformants per 10(7) spores. PCR and Southern analysis showed that the T-DNA was integrated into the genome. Among of the examined transformants contained a single copy, and was stable through mitotic cell division. The transformation system mediated by Agrobacterium tumefaciens may prove to be a powerful tool for the filamentous fungi transformation and functional genomic study with its high transformation frequency, simplicity of T-DNA integration, and genetic stability of transformants.
Coherent control of negative refraction based on local-field enhancement and dynamically induced chirality

Abstract: This paper studies the electromagnetic response of a coherently driven dense atomic ensemble to a weak probe. It finds that negative refraction with little absorption may be achieved in the presence of local-field enhanced interaction and dynamically induced chirality. The complex refractive index governing the probe refraction and absorption depends critically on the atomic density, the steady population distribution, the coherence dephasings, and the frequency detunings, and is also sensitive to the phase of the driving field because the photonic transition paths form a close loop. Thus, it can periodically tune the refractive index at a fixed frequency from negative to positive values and vice versa just by modulating the driving phase. Moreover, the optimal negative refraction is found to be near the probe magnetic resonance, which then requires the electric fields of the probe and the drive being on two-photon resonance due to the dipole synchronisation.

Effects of Arr Mutation on Biofilm and Pyocyanin Biosynthesis in Pseudomonas aeruginosa

Abstract: As an opportunistic pathogen, Pseudomonas aeruginosa can produce biofilm and pyocyanin, which play a critical role in its pathogenesis. This study aims to elucidate the function of the Arr gene in formation of biofilm and production of pyocyanin in Pseudomonas aeruginosa. Using the chromosome DNA of Pseudomonas aeruginosa as template, we first cloned the Arr gene by PCR. With the insertion of gentamycin resistance cassette (aacC1), the mutant PA-AG has then been constructed by homologous recombination. The formation of biofilm is determined by staining with crystal violet, and production of pyocyanin is detected with spectrophotometric method. In KMB or LB medium, biofilm formation of the parental strain PAO1 is about 2 folds higher than that of the Arr mutant PA-AG. The biosynthesis of pyocyanin in the Arr mutant PA-AG is about 2.5 folds higher than that in the parental strain PAO1. These results indicate that the Arr gene negatively controls the pyocyanin biosynthesis, but Arr could exert some positive effect on the biofilm formation. It is suggested that regulation mediated by the Arr gene on biofilm and biosynthesis of pyocyanin in Pseudomonas aeruginosa is specific and different.
Ischemic Preconditioning of Rat Livers from Non-Heart-Beating Donors Decreases Parenchymal Cell Killing and Increases Graft Survival after Transplantation

Abstract: A critical shortage of donors exists for liver transplantation, which non-heart-beating cadaver donors could help ease. This study evaluated ischemic preconditioning to improve graft viability after non-heart-beating liver donation in rats. Ischemic preconditioning was performed by clamping the portal vein and hepatic artery for 10?min followed by unclamping for 5?min. Subsequently, the aorta was cross-clamped for up to 120?min. After 2?h of storage, livers were either transplanted or perfused with warm buffer containing trypan blue. Aortic clamping for 60 and 120?min prior to liver harvest markedly decreased 30-day graft survival from 100% without aortic clamping to 50% and 0%, respectively, which ischemic preconditioning restored to 100 and 50%. After 60?min of aortic clamping, loss of viability of parenchymal and nonparenchymal cells was 22.6 and 5.6%, respectively, which preconditioning decreased to 3.0 and 1.5%. Cold storage after aortic clamping further increased parenchymal and non-parenchymal cell killing to 40.4 and 10.1%, respectively, which ischemic preconditioning decreased to 12.4 and 1.8%. In conclusion, ischemic preconditioning markedly decreased cell killing after subsequent sustained warm ischemia. Most importantly, ischemic preconditioning restored 100% graft survival of livers harvested from non-heart-beating donors after 60?min of aortic clamping. 1. Introduction Liver transplantation surgery is a viable alternative for patients with end-stage liver disease but the number of heart-beating cadavers suitable for liver donation remains a key limitation. In human kidney transplantation, organ donation from non-heart-beating cadavers is now employed successfully at many centers [1]. Organ donors are typically terminally ill patients who do not meet the criteria of brain death and whose life support is withdrawn at the request of the family. After cardiac arrest occurs and death is pronounced several minutes later, the organs are harvested. The use of livers from non-heart-beating donors is also emerging as an important stratagem to expand the liver donor pool [2]. Organs from non-heart-beating cadaver donors typically experience several minutes of warm ischemia prior to cold preservation. Warm ischemic injury that occurs to livers after cardiac arrest can severely compromise graft viability. Early clinical results with livers from non-heart-beating donors were poor, and two-month graft survival was only 50% even for donors that were extubated in an operating room setting [3]. Abstract: The phase state and distribution rule could recognized clearly and its control factors could investigation, as a means of the PVT experimental data of 80 layers and the physical and chemical characters of these hydrocarbons produced from oil and gas pools in Kuqa petroleum system. The main type was gaseity and subordinative type was liquid in hydrocarbon fluid system in this system. The system divided two or three belts-south, middle and north, and two segments-east and west, and the north belt was rich in gas pools, including dry gas pools and wet gas pools as will as poor condensate gas pools, and the south belt contained more abundant condensate gas pools with fewness partner oil pools and volatilty oil pools. However the west segment of whole system contained more superabundant and abundant condensate gas pools and oil pools in this system, for example the Yangtake, Qule and Wush(Yilake) regions concentrated abundant condensate gas pools and oil pools. It would be characteristic of history of forming-pool of the early stage accumulated normal oil and abundant condensate gas on south belt in main, late stage accumulated dry gas, wet gas and pool condensate gas on north belt in main. Naturalness more dryer gas generated during late stage would charged into the early pools, and the hydrocarbon fluid system in these pools changed them phase state owing to them component admixed each other. Of course the early traps profited forming abundant condensate gas pools and oil pools, whereas the late traps were prone to form more dryer gas pools. So the traps surrounding early uplift and on early uplift were prone to form normal oil and abundant condensate gas. Special most lithologic traps formed early stage and could be finded more rich-oil pools.
Development of EST-SSR markers in Pleurotus ostreatus and construction of primary core collection

Abstract: 根据已知的粗皮侧耳、灵芝、香菇EST序列开发出9条粗皮侧耳EST-SSR引物、58条灵芝EST-SSR引物和35条香菇EST-SSR引物,从中筛选出4条多态性好、条带清晰的引物,对选取的185株粗皮侧耳栽培种进行扩增分析。共扩增得到59条带,每个引物扩增条带数为12–19条,扩增片段在100–1,100bp之间。根据扩增结果构建了一个含47株粗皮侧耳栽培种的初级核心种质库P-core 47。分析得出P-core 47占原群体样品数25%,占资源库的13%,有效等位基因数和多态性位点占有率与原群体一致,多样性指数均值比原群体高0.12747,在原群体的低频率等位基因有所增强。P-core 47内菌株来源广泛,基本已覆盖各个栽培地区,统计的质量性状和数量性状均可在其中找到代表性菌株。表明了用EST-SSR分子标记构建粗皮侧耳栽培种初级核心种质库的可行性。
Constructing muti-level TIN with voronoi diagrams

Abstract: To study the LOD techniques for large scale terrain visualization, this paper focused on the design of muhiresolution TIN. First, a new approach of constructing multiresolution TIN that represents terrain well was given based on Voronoi diagrams and Delaunay triangulation networks. Second, multiresolution TIN was incorporated, thus making the management and load of it even more easily. The results show that the response speed and display effect are very satisfying. It can make real time and seamless walkthrough in large scale 3D visualization.
Reinforcement Learning Ramp Metering without Complete Information

Abstract: This paper develops a model of reinforcement learning ramp metering (RLRM) without complete information, which is applied to alleviate traffic congestions on ramps. RLRM consists of prediction tools depending on traffic flow simulation and optimal choice model based on reinforcement learning theories. Moreover, it is also a dynamic process with abilities of automaticity, memory and performance feedback. Numerical cases are given in this study to demonstrate RLRM such as calculating outflow rate, density, average speed, and travel time compared to no control and fixed-time control. Results indicate that the greater is the inflow, the more is the effect. In addition, the stability of RLRM is better than fixed-time control.
