The Range Extension of the Critically Endangered, Poecilotheria smithi in Sri Lanka, with Notes on its Sociality

Abstract: Poecilotheria smithi is a Critically Endangered Theraposid known only from the type locality Haragama in the Kandy District, Sri Lanka. It was thought to be distribution specific to Haragama. During a survey on the genus Poecilotheria, which was initiated in 2011 by the authors, P. smithi was recorded, the first confirmed observation of P. smithi outside of its type locality from the Matale district about 31.42 Km in aerial distance northwest of the type locality. Distribution of this species extended in Sri Lanka by this novel record. P. smithi displays the social behaviour of sharing same microhabitat with few individuals. As demonstrated for P. smithi, we suggest the large group size and social behaviour observed was in response to unavailability of suitable micro habitat for the mature individuals.
Ammonia poisoning causes muscular but not liver damage in cattle

Abstract: twelve steers were intraruminally administered a high dose (0.5g/kg bw) of urea to study the damage effect of ammonia poisoning on liver and/or muscles. blood samples were collected to determine ammonia and activities of ggt, ast and ck. eleven steers were successfully poisoned and treated properly, but one succumbed. poisoned cattle showed high concentration of ammonia, and higher activities of ast and ck. the higher the ammonia, the greater were the activities of ast (r=0.59) and ck (r=0.61). the correlation between ast and ck was high and significant (r=0.80), but not between ast and ggt (r=0.19). the activities of ast and ck were higher after the beginning of the convulsive episodes due to ammonia poisoning. those results showed that occurred muscle damage instead of liver damage since ck is a typical enzyme from skeletal muscle; ast is found either in skeletal muscle and hepatocytes, while ggt is present in hepatic cells.
Detection and analysis of apoptosis in peripheral blood cells from breast cancer patients

Abstract: Apoptosis is a well-known specific process of cell death that normally occurs in physiological situations such as tissue or organ development and involution. During tumor growth there is a balance between proliferation and cell death which involves apoptotic mechanisms. In the present study genomic DNAs from 120 breast tumor biopsies were analyzed by agarose gel electrophoresis and none of them presented the fragmentation pattern characteristic of the apoptosis process. However, 33% of the 105 breast cancer patients clearly showed the apoptotic pattern when DNA from blood cells was analyzed. None of the DNAs from healthy volunteer blood cells showed any trace of apoptosis. Since the breast cancer patients were not receiving chemo- or hormone therapy, the possible relationship between blood cortisol levels and the apoptotic pattern found in patient blood cells was investigated. Using a chemoluminescence immunodetection assay, similar cortisol levels were observed in breast cancer patient sera presenting or not apoptotic blood cells and in healthy volunteer sera. Analysis of the clinical data obtained from 60 of these patients showed that patients bearing tumors of smaller size (under 20 mm) were more susceptible to the apoptotic effect in blood cells. According to the Elston grade, it was observed that 7 of 12 patients with grade III tumors (58%) presented apoptotic peripheral blood cells, in contrast to 10 of 48 patients with grade I and grade II tumors. These observations may reflect the immunosuppression characteristic of some breast cancer patients, which may contribute to tumor growth. Therefore, further studies are necessary to elucidate the factor(s) involved in such massive blood cell death.
Association between EcoRI fragment-length polymorphism of the immunoglobulin lambda variable 8 (IGLV8) gene family with rheumatoid arthritis and systemic lupus erythematosus

Abstract: the human immunoglobulin lambda variable 8 (iglv8) subgroup is a gene family containing three members, one of them included in a monomorphic 3.7-kb ecori genomic fragment located at the major lambda variable locus on chromosome 22q11.1 (gene iglv8a, embl accession no. z73650) at 100% frequency in the normal urban population. the second is a polymorphic rflp allele included in a 6.0-kb ecori fragment at 10% frequency, and the third is located in a monomorphic 8.0-kb ecori fragment at 100% frequency, the last being translocated to chromosome 8q11.2 and considered to be an orphan gene. our southern blot-ecori-rflp studies in normal individuals and in patients with rheumatoid arthritis (ra) or with systemic lupus erythematosus (sle), using a specific probe for the iglv8 gene family (probe pvl8, embl accession no. x75424), have revealed the two monomorphic genomic fragments containing the iglv8 genes, i.e., the 3.7-kb fragment from chromosome 22q11.1 and the 8.0-kb fragment from 8q11.2, both occurring at 100% frequency (103 normal individuals, 48 ra and 28 sle patients analyzed), but absence of the 6.0-kb iglv8 polymorphic rflp allele in all ra or sle patients. as expected, the frequency of the 6.0-kb allele among the normal individuals was 10%. these findings suggest an association between the absence of the 6.0-kb ecori fragment and rheumatoid arthritis and systemic lupus erythematosus.
Asynchronous expression of myeloid antigens in leukemic cells in a PML/RARalpha transgenic mouse model

Abstract: acute promyelocytic leukemia (apl) is characterized by the expansion of blasts that resemble morphologically promyelocytes and harbor a chromosomal translocation involving the retinoic acid receptor a (rara) and the promyelocytic leukemia (pml) genes on chromosomes 17 and 15, respectively. the expression of the pml/rara fusion gene is essential for apl genesis. in fact, transgenic mice (tm) expressing pml/rara develop a form of leukemia that mimics the hematological findings of human apl. leukemia is diagnosed after a long latency (approximately 12 months) during which no hematological abnormality is detected in peripheral blood (pre-leukemic phase). in humans, immunophenotypic analysis of apl blasts revealed distinct features; however, the precise immunophenotype of leukemic cells in the tm model has not been established. our aim was to characterize the expression of myeloid antigens by leukemic cells from hcg-pml/rara tm. in this study, tm (n = 12) developed leukemia at the mean age of 13.1 months. morphological analysis of bone marrow revealed an increase of the percentage of immature myeloid cells in leukemic tm compared to pre-leukemic tm and wild-type controls (48.63 ± 16.68, 10.83 ± 8.11, 7.4 ± 5.46%, respectively; p < 0.05). flow cytometry analysis of bone marrow and spleen from leukemic tm identified the asynchronous co-expression of cd34, cd117, and cd11b. this abnormal phenotype was rarely detected prior to the diagnosis of leukemia and was present at similar frequencies in hematologically normal tm and wild-type controls of different ages. the present results demonstrate that, similarly to human apl, leukemic cells from hcg-pml/rara tm present a specific immunophenotype.

Abstract: the pediatric glasgow coma scale was modified for use in veterinary medicine. thirty adult dogs with neurological alterations susceptible to evaluation of the conscience were studied. the modified scale for dogs was applied at three moments with intervals of 48 hours, resulting in 90 events. the scale was applied for evaluation of the ocular opening (oo), the best answer associated to vocalization (bav), and the best motor answer (bma). with the factorial analysis for the indicators oo, bav, and bma the initial values of 2.482, 0.302, and 0.215, respectively, were obtained. the variance was of 82.7%, 10.1%, and 7.2%, respectively, and cumulative of 82.7%, 92.8%, and 100%, respectively. the expected extraction of the main factor, oo, was 2.482 with variance of 82.7% and cumulative of 82.7%. the evaluation result of the conscience was normal in 10% of the animals, light alteration in 20%, moderate in 45%, and severe in 25%, with scores of glasgow equal to 15, from 13 to 14, nine to 12, and eight to three, respectively. the pediatric glasgow coma scale modified for dogs was a safe tool for evaluation of the conscience of adult dogs.

Abstract: a severe cutaneous reaction in a female dog after administration of penicillin and cephalexin is described, in which the main signs presented by the patient were fever, lack of appetite, and detachment of epidermis in the dorsal region of body.the established treatment was surgical debridement of the region, topical application of aloe vera with sugar, and systemic antibiotic therapy with ciprofloxacin, resulting in the recovery and wound closure in approximately 45 days.

Abstract: the objective of this paper was to analyze two protocols for the production of platelet rich plasma (prp) in dogs. peripheral blood of 20 adult dogs was collected into three tubes. the first was processed through protocol a - single centrifugation at 1200rpm for 10min - , the second was submitted to protocol b - a first centrifugation at 1200rpm for 10min and a second centrifugation at 1600rpm for 10min - and the third was used to perform platelet count in whole blood, which served as a parameter for values obtained in prp. the protocol in which it was possible to achieve a higher platelet count was tested in other 20 dogs to evaluate its reproducibility. protocol b resulted in a superior platelet count in 100% of the samples, concluding that the referred protocol is effective for prp production in dogs.
