Snake venom metalloproteinases and disintegrins: interactions with cells

Abstract: metalloproteinases and disintegrins are important components of most viperid and crotalid venoms. large metalloproteinases referred to as mdc enzymes are composed of an n-terminal metalloproteinase domain, a disintegrin-like domain and a cys-rich c-terminus. in contrast, disintegrins are small non-enzymatic rgd-containing cysteine-rich polypeptides. however, the disintegrin region of mdc enzymes bears a high degree of structural homology to that of the disintegrins, although it lacks the rgd motif. despite these differences, both components share the property of being able to recognize integrin cell surface receptors and thereby to inhibit integrin-dependent cell reactions. recently, several membrane-bound mdc enzymes, closely related to soluble venom mdc enzymes, have been described in mammalian cells. this group of membrane-anchored mammalian enzymes is also called the adam family of proteins due to the structure revealing a disintegrin and metalloproteinase domains. adams are involved in the shedding of molecules from the cell surface, a property which is also shared by some venom mdc enzymes.

Abstract: Metalloproteinases and disintegrins are important components of most viperid and crotalid venoms. Large metalloproteinases referred to as MDC enzymes are composed of an N-terminal Metalloproteinase domain, a Disintegrin-like domain and a Cys-rich C-terminus. In contrast, disintegrins are small non-enzymatic RGD-containing cysteine-rich polypeptides. However, the disintegrin region of MDC enzymes bears a high degree of structural homology to that of the disintegrins, although it lacks the RGD motif. Despite these differences, both components share the property of being able to recognize integrin cell surface receptors and thereby to inhibit integrin-dependent cell reactions. Abstract: The GERDA experiment aims to search for the neutrinoless double beta-decay of 76Ge and possibly for other rare processes. The sensitivity of the first phase is envisioned to be more than one order of magnitude better than in previous neutrinoless double beta-decay experiments. This implies that materials with ultra-low radioactive contamination need to be used for the construction of the detector and its shielding. Therefore the requirements on material screening include high-sensitivity low-background detection techniques and long measurement times. In this article, an overview of material-screening laboratories available to the GERDA collaboration is given, with emphasis on the gamma-spectrometry. Additionally, results of an intercomparison of the evaluation accuracy in these laboratories are presented.
LArGe - Active background suppression using argon scintillation for the GERDA $0νββ$-experiment

Abstract: LArGe is a GERDA low-background test facility to study novel background suppression methods in a low-background environment, for future application in the GERDA experiment. Similar to GERDA, LArGe operates bare germanium detectors submersed into liquid argon (1 m$^3$, 1.4 tons), which in addition is instrumented with photomultipliers to detect argon scintillation light. The scintillation signals are used in anti-coincidence with the germanium detectors to effectively suppress background events that deposit energy in the liquid argon. The background suppression efficiency was studied in combination with a pulse shape discrimination (PSD) technique using a BEGe detector for various sources, which represent characteristic backgrounds to GERDA. Suppression factors of a few times $10^3$ have been achieved. First background data of LArGe with a coaxial HPGe detector (without PSD) yield a background index of (0.12$-$4.6)$\cdot 10^{-2}$ cts/(keV$\cdot$kg$\cdot$y) (90% C.L.), which is at the level of GERDA Phase I. Furthermore, for the first time we monitor the natural $^{42}$Ar abundance (parallel to GERDA), and have indication for the $2\nu\beta\beta$-decay in natural germanium. These results show the effectivity of an active liquid argon veto in an ultra-low background environment. As a consequence, the implementation of a liquid argon veto in GERDA Phase II is pursued.
Calidad de la ejecución del Programa de Atención a Centros con Becarios Extranjeros

Abstract: a research on health systems and services was conducted to evaluate the quality of the implementation of the program of care for centers with foreign scholars in santiago de cuba municipality during 2010. the universe was constituted by 6 centers with foreign scholars, 25 service providers and 26 users. the issues studied were scientific-technical quality (professional competence) and satisfaction of providers and users. the process of evaluation revealed it was inappropriate due to the low professional competence of providers (66.6 %), the high frequency of health controls without results, and the low demand for health (33.4 %); and evaluation of the results was also inappropriate because of failure to comply with the indicators of the program. there was dissatisfaction of providers and users. recommendations related to these problems were made.

Abstract: a research on health systems and services was conducted to evaluate the quality of implementation of the surveillance and control program of other culicids in "xx aniversario" and "armando garcía aspurú" health areas of santiago de cuba municipality in the period 2009-2010. the sample consisted of 28 natural breeding sites, 10 specific capture points, 11 providers and 60 users. the aspects studied were scientific and technical quality (professional competence) and satisfaction of providers and users. evaluation of the structure revealed that it was inadequate due to specialized personnel deficit (50.0 %), absence of the program and limited availability of batteries and carter buckets; evaluation of the process revealed low professional competence and failure to comply with biological and physical treatments; and that of the results showed dissatisfaction with the training activities (81.1 %) and lack of material resources (63.6 %). the percentage of positive breeding sites was found to be high, larval densities and bite rates were out of permissible parameters, and poor sanitation prevailed. recommendations related to identified problems were given.

Abstract: Se realizó una investigación sobre sistemas y servicios de salud para evaluar la calidad de la ejecución del Programa de Vigilancia y Control de otros Culícidos en las áreas de salud "XX Aniversario" y "Armando García Aspurú" del municipio de Santiago de Cuba, en el período 2009-2010. El universo estuvo constituido por 28 criaderos naturales, 10 puntos de captura específica, 11 prestadores y 60 usuarios. Las dimensiones estudiadas fueron la calidad científico-técnica (competencia profesional) y la satisfacción de prestadores y usuarios. La evaluación de la estructura reveló que fue inadecuada por déficit de personal especializado (50,0 %), por la ausencia del programa y por la escasa disponibilidad de pilas y cucharones carter; la del proceso, por baja competencia profesional e incumplimientos de tratamientos biológicos y físicos; la de los resultados, por insatisfacción con las actividades de capacitación (81,1 %) y por déficit de recursos materiales (63,6 %). El porcentaje de positividad de los criaderos resultó ser elevado, las densidades larvarias e índices de picada estuvieron fuera de parámetros permisibles y predominaron las malas condiciones de saneamiento. Se emitieron recomendaciones relacionadas con los problemas identificados. Las dimensiones estudiadas fueron la calidad científico-técnica (competencia profesional) y la satisfacción de prestadores y usuarios. La evaluación del proceso reveló que fue inadecuado por la baja competencia profesional de los prestadores (66,6 %), por la alta frecuencia de inspecciones sanitarias sin resultados y por la baja exigencia sanitaria (33,4 %); la de los resultados, por incumplimiento de indicadores del programa. Hubo insatisfacción de usuarios y prestadores. Se emitieron recomendaciones relacionadas con los problemas identificados. Abstract: This Letter of Intent has been submitted to the Scientific Committee of the INFN Laboratori Nazionali del Gran Sasso (LNGS) in March 2004. It describes a novel facility at the LNGS to study the double beta decay of 76Ge using an (optionally active) cryogenic fluid shield. The setup will allow to scrutinize with high significance on a short time scale the current evidence for neutrinoless double beta decay of 76Ge using the existing 76Ge diodes from the previous Heidelberg-Moscow and IGEX experiments. An increase in the lifetime limit can be achieved by adding more enriched detectors, remaining thereby background-free up to a few 100 kg-years of exposure.

Abstract: an observational, descriptive and cross-sectional study was conducted in 52 patients with acute respiratory failure admitted to the intensive care unit of "orlando pantoja tamayo" general teaching hospital of contramaestre municipality in santiago de cuba, from january 2010 to december 2011, who required noninvasive mechanical ventilation (with the delay or avoidance of invasive mechanical ventilation) in order to demonstrate the efficacy of this therapeutic modality. in the series a prevalence of people affected over 70 years and the female sex was observed, as well as a high frequency of chronic obstructive lung disease, heart failure and respiratory infections. the application of this technique decreased hospital stay in this service and increased the quality of patients' life.
