Intracellular distribution of Tankyrases as detected by multicolor immunofluorescence techniques

Abstract: Poly(ADP-ribose) polymerases are a family of enzymes that catalyze the conversion of NAD+ into ADP-ribose. Among them, Tankyrases have been found to bind to centrosome, mitotic spindle and microsome proteins, in the cytoplasm, and to telomeres in the nucleus, where they play a relevant role in telomere metabolism. However, their precise intracellular localization during interphase has not been so far fully elucidated. We investigated this aspect in situ by double immunofluorescence experiments using antibodies recognizing Tankyrases 1-2 or other proteins residing in specific organelles (Golgi apparatus, mitochondria, lysosomes, endoplasmic reticulum). We used HeLa cells as a model system in vitro, before and after treatment with either actinomycin D or etoposide, to also investigate the possible relocation of Tankyrases during apoptosis. We observed that Tankyrases are distributed both in the nucleus and in the cytoplasm; in this latter compartment, they were found to colocate with the Golgi apparatus but never with the mitochondria; a pool of Tankyrases also colocates with the endoplasmic reticulum and lysosomes. Interestingly, in cells with clear signs of apoptosis, Tankyrases were detectable in the cytoplasmic blebs: this suggests that they are not massively cleaved during apoptosis and persist in the largely heterogeneous apoptotic remnants which are known to contain components of cytoplasmic and nuclear origin.

Abstract: A demanda por produtos light e funcionais, e o fato de o queijo minas apresentar lugar de destaque na produ § £o nacional de queijos (terceiro lugar), motivaram a realiza § £o do presente estudo. O objetivo foi o desenvolvimento de formula § £o de queijo Minas light padronizado enriquecido com fonte de ferro (citrato f rrico amoniacal). Na fabrica § £o do queijo Minas frescal empregou-se cultura l ctica mista de Streptococcus lactis e Streptococcus cremoris. Nos queijos foram avaliados a composi § £o (prote -na, gordura, lactose, minerais, umidade, acidez, cloretos, c lcio, s 3dio, ferro), valor nutricional e caracter -sticas sensoriais (aroma, sabor, textura, acidez e aceitabilidade geral). Para a avalia § £o dos resultados utilizou-se an lise de vari ¢ncia (ANOVA) e teste de Tukey para diferen §a de m dias. Os resultados demonstraram ser poss -vel uma redu § £o de 59 % no teor de lip -dios e a fortifica § £o com 55 mg de Fe/kg de queijo com citrato f rrico amoniacal, sem ocasionar mudan §as significativas (p<0,05) nas caracter -sticas sensoriais e na aceitabilidade. A produ § £o de queijos com teor reduzido de lip -dios e enriquecido com micronutrientes, oferece novas oportunidades de mercado de produtos diferenciados e com valor agregado.

A new stable basis for RBF approximation

Abstract: It's well know that Radial Basis Function approximants suffers of bad conditioning if the simple basis of translates is used. A recent work of M.Pazouki and R.Schaback gives a quite general way to build stable, orthonormal bases for the native space based on a factorization of the kernel matrix A. Starting from that setting we describe a particular orthonormal basis that arises from a weighted singular value decomposition of A. This basis is related to a discretization of the compact operator which leads to the so-called eigenbasis, and provides a connection with it. We give convergence estimates and stability bound for the interpolation and the discrete least-squares approximation based on this basis, which involves the eigenvalues of such an operator.

Abstract: the present study characterizes the early stages of development of brycon hilarii (valenciennes, 1850) based on morphometric and meristic characters. patterns of development in morphometric characters were analyzed through the fit of linear, quadratic and linear piecewise models. samples were collected from march 2000 to march 2004 in basins of the cuiabá and manso rivers, state of mato grosso, brazil. larvae were identified and classified in different stages (larval yolk, pre-flexion, flexion, and post-flexion) according to notochord development. the morphometric and meristic characters for each larva were, respectively, measured and counted. the standard length ranged from 3.25 to 26.00 mm. the larval yolk stage showed a greater concentration of dendritic pigments along the intestine, and the flexion and post-flexion stages showed an intensified pigmentation intensified along the back. a single spot on the humerus and another on the caudal peduncle were observed at the end of the post-flexion stage. also, in the larval yolk and pre-flexion stages. the intestine is long, the snout is short, the adhesive organ was present in the larval yolk and pre-flexion, and the mouth is terminal throughout the development. the total number of myomeres ranged from 41 to 50 (23 to 30 pre-anal and 16 to 24 post-anal). number of fin rays: dorsal 11; anal 30; pectoral 15 and ventral eight. the analysis of the development indicated higher metamorphosis in the flexion stage.

Reactive oxygen species have been implicated in age-related skin disorders. Dietary bioactive antioxidant compounds, such as polyphenols, have beneficial effects on skin health. The advantage of a nutritional administration route is that blood delivers nutraceutical bioactive compounds continuously to all skin compartments, ie, the epidermis, dermis, and subcutaneous fat. The purpose of this study was to evaluate the topical and systemic effects of a dietary supplement containing resveratrol and procyanidin on age-related alterations to the skin, the skin antioxidant pool, and systemic oxidative stress levels. Methods: An instrumental study was performed in 50 subjects (25 treated with supplements and 25 with placebo) to identify clinical features induced by chronoaging or photoaging. Product efficacy was evaluated after 60 days of treatment in terms of in vivo and in situ skin hydration, elasticity, and skin roughness levels, systemic oxidative stress levels by plasmatic derivatives of reactive oxygen metabolites and oxyadsorbent tests, and extent of the skin antioxidant pool. Results: After 60 days of treatment, values for systemic oxidative stress, plasmatic antioxidant capacity, and skin antioxidant power had increased significantly. Additionally, skin moisturization and elasticity had improved, while skin roughness and depth of wrinkles had diminished. Intensity of age spots had significantly decreased, as evidenced by improvement in the individual typological angle. Role of Phosphatidyl-Serine in Bone Repair and Its Technological Exploitation

Abstract: In the 1970s, morphological evidence collected by electron microscopy linked mineral deposition (“calcification” or “mineralization”) in newly-forming bone to membrane-encapsulated particles of a diameter of approximately 100 nm (50–200 nm) that were called “matrix vesiscles”. As the characterisation of these vesicles progressed towards their biochemical composition, the role of lipids in the biomineralization process appeared to be crucial. In particular, a group of cell-membrane phospholipids were identified as major players in the crystal formation process. Indeed, in the 1980s it became clear that phosphatidylserine, together with proteins of the annexin family, was among the most important molecules in binding calcium ions and that this phospholipid was involved in the regulation of the early stages of mineralization in vivo. During the same period of time, the number of surgical implantations of orthopaedic, dental and maxilo-facial devices requiring full integration with the treated bone prompted the study of new functionalisation molecules able to establish a stable bonding with the mineral phase of the host tissue. In the late 1990s studies started that aimed at exploiting the potential of calcium-binding phospholipids and, in particular, of the phosphatidylserine as functionalisation molecules to improve the osteointegration of artificial implants. Later, papers have been published that show the potential of the phophatidylserine and phosphatidylserine-mimicking coating technology to promote calcification both in vitro and in vivo. The promising results support the future clinical application of these novel osteointegrative biomaterials.
Severe community acquired pneumonia: what should we predict?

Abstract: What is the concept of severe CAP based on? CAP severity could be based on pathophysiological disorders that would allow physicians to objectively define severity. Alternatively, CAP severity could be based on treatment intensity, which would certainly facilitate its uptake and the development of clinical tools to assist physicians in decision-making. This second approach, which seems more clinically relevant, will be assumed for the rest of this letter.What are the most relevant criteria for severe CAP? Basically, there are two possibilities: admission to ICU or respiratory/circulatory support requirement [1]. The former criterion is often perceived as too vague as it remains a matter of individual judgment [2]. The second is less prone to subjectivity, but may be too restrictive, as intensive care benefit is not limited to patients that require vital support. However, one must keep in mind the final objective of developing prediction tools for severe CAP; in this context, the definition exclusively based on respiratory/hemodynamic support requirement is less restrictive. Therefore, consistent with Chalmers [3], we suggest keeping this second definition of CAP severity.At this point, several features regarding prediction of severe CAP may be considered. Given the increasing evidence that patients with incipient severe sepsis may benefit from intensive care and from early referral, this prediction would apply to CAP patients presenting to the emergency department with rapidly progressive pneumonia at high risk of developing organ failure but with no overt respiratory/hemodynamic distress [4]. Indeed, predicting what is obvious is of no value. In contrast, patients that are too sick to be eligible for ICU admission have to be excluded. Additionally, regardless of the 30-day risk of severe CAP after presentation to the emergency department, if the 7-day risk is extremely low, CAP patients could be hospitalized in the wards. This paper aims to analyze the questions on the European citizenship and the democratic debt of the supranational institutions. In this sense it addresses the institution of the European Provider of Justice. It is about an individual appointed by the European Parliament with the task of, either by its own initiative or based on complaints presented by European citizens and inhabitants, investigating and proceeding to investigation about irregularities on the actuation of the communitarian organs, as well as to emit recommendations with the aim of correcting the cases of bad administration and violation of the human rights detected by it. The object of actuation of the European Provider of Justice is the enforcement of political activity and the providing of public services in a communitarian set. It is an access channel for the citizenship to the organs of political power, controlling the bad actuation of the institutions or communitarian organisms and the violation of rights, with broad powers of supervision with the aim of the enforcement of the fundamental right to a good administration.
