Abstract: Es wird ein überblick über den Entwicklungs- und Etablierungsstand der qualitativen Forschung im deutschen Sprachraum in methodischer Hinsicht gegeben. Nach knappen Bemerkungen zur Geschichte werden die wichtigsten Forschungsperspektiven und Schulen qualitativer Forschung – objektive Hermeneutik, Ethnomethodologie, Narrationsanalysen, Grounded Theory, Lebensweltanalyse, Ethnographie und Cultural und Gender Studies – jeweils vor dem Hintergrund aktueller Entwicklungen skizziert. Der Etablierungsstand qualitativer Forschung wird in Hinblick auf die Verankerung in der Deutschen Gesellschaft für Soziologie (DGS) und in der International Sociological Association (ISA) sowie auf Entwicklungen im Bereich von Lehr- und Handbüchern und die Gründung darauf spezialisierter Fachzeitschriften behandelt. Als methodische Trends werden die Hinwendung zu visuellen und elektronischen Daten, die Triangulation methodischer Zug nge und die Hybridisierung qualitativer Vorgehensweisen diskutiert. Den Abschluss bilden einige Perspektiven, die in Zukunft in der qualitativen Forschung an Gewicht gewinnen werden bzw. Desiderate der weiteren Kl rung darstellen. Neben der Verwendung von Computern und der Kl rung der Verbindung von qualitativer und quantitativer Forschung bzw. der diesbezüglichen Grenzen und Probleme sind hier die weitere Kl rung der Frage der Darstellung von Vorgehensweisen und Ergebnissen und die Entwicklung angemessener und gleichzeitig verbindlicher Gütekriterien qualitativer Forschung zu nennen. Weiterhin werden Schulenbildung und Forschungspragmatik einerseits und Tendenzen der Kl rung und Verkl rung methodischer Vorgehensweisen andererseits als aktuelle Spannungsfelder qualitativer Methodendiskussion ausgemacht. Abschlie end werden die Internationalisierung der qualitativen Forschung und die Beantwortung der Frage der Indikationsstellung qualitativer Methoden als Desiderate behandelt. Regulation of Sirtuin Function by Posttranslational Modifications

Abstract: Sirtuins are homologs of the yeast silencing information regulator 2 protein, an NAD+-dependent (histone) deacetylase. In mammals seven different sirtuins, SIRT1–7, have been identified, which share a common catalytic core domain but possess distinct N- and C-terminal extensions. This core domain elicits NAD+-dependent deacetylase and in some cases also ADP-ribosyltransferase, demalonylase, and desuccinylase activities. Sirtuins have been implicated in key cellular processes, including cell survival, autophagy, apoptosis, gene transcription, DNA repair, stress response, and genome stability. In addition some sirtuins are associated with disease, including cancer and neurodegeneration. These findings suggest strongly that sirtuins are tightly controlled and potentially responsive to different signal transduction pathways. Here, we review the posttranslational regulation mechanisms of mammalian sirtuins and discuss their relevance regarding the physiological processes, with which the different sirtuins are associated. The available data suggest that the N- and C-terminal extensions are the targets of posttranslational modifications (PTM) that can affect the functions of sirtuins. Mechanistically this can be explained by the interaction of these extensions with the catalytic core domain, which appears to be controlled by PTM at least in some cases. In contrast little is known about PTM and regulation of the catalytic domain itself. Together these findings point to key regulatory roles of the N- and C-terminal extensions in controlling sirtuin functions, thus connecting these regulators to different signaling pathways.

Abstract: The increasing number of textbooks and handbooks on qualitative research reflects developments in qualitative research as a field. In this book review symposium—based on a "Meet the Author" Session at the European Sociological Association Conference in Glasgow in 2007—several recent examples of handbooks written or edited by Uwe FLICK are discussed by two commentators. The author of the books then adds his own comments and responses. The discussion covers four main issues. First, tensions between intensifying the reflexivity of qualitative research and consolidating it as a competitor on the market of research, research training and funding are discussed. A second issue is how handbooks contribute to bridging the gaps between different local or language-specific traditions of qualitative research. A third issue is how to integrate more strongly the idea of research design into the methodological discussion (and practice) of qualitative research more strongly. A fourth issue is how to promote and assess the quality of qualitative research and overcome a "legitimation crisis". These issues are discussed by the authors of this book review symposium from different perspectives with a focus on teaching qualitative methodology and on the progress of qualitative research. En este simposio de rese as, basado en la sesión "Conozca al autor" de la Conferencia de Glasgow en 2007 de la Asociación Europea de Sociología, dos comentaristas discutieron varios ejemplos recientes de manuales escritos o editados por Uwe FLICK. El autor de los libros brindó respuestas e hizo comentarios. La discusión abarca cuatros temas principales. Primero, se discuten las tensiones entre intensificar la reflexividad de la investigación cualitativa y consolidarla como un competidor en el mercado de la investigación, la capacitación para la investigación y el financiamiento. Un segundo tema es como los manuales ayudan a superar los huecos entre diferentes tradiciones locales o con lenguajes específicos de la investigación cualitativa. El tercer tema es como integrar más solidamente la idea del dise o de la investigación a la discusión metodológica y práctica de la investigación. El cuarto tema es como promover y evaluar la calidad de la investigación cualitativa. Abstract: Der Aufsatz bietet eine Einführung in die FQS-Schwerpunktausgabe über qualitative Methoden in Europa. Er schildert die besondere Situation der qualitativen Forschung in diesem Raum, die durch Vielfalt und Einheit gepr gt ist. Vielfalt, denn die verschiedenen geistigen Traditionen und institutionellen Strukturen der Sozialwissenschaft, die den Hintergrund der qualitativen Forschung bilden, unterscheiden sich zwischen den einzelnen L ndern betr chtlich. Vielfalt verweist auf eine Reihe von selbst ndigen Weisen der Forschungspraxis, die neben den bekannten anglos chsischen Entwicklungen zu finden sind und diese erg nzen. Eine Einheit findet sich trotz aller Unterschiede der Forschungspraktiken darin, dass sie alle an einem interpretativen Paradigma ausgerichtet sind, eine Ausrichtung, die auf Sinn, Verstehen und Kontextualit t aufbaut. This variation indicates a number of traditional ways to do qual-itative research that complement and complete the well-known Anglo-Saxon development. Temperature prediction in a refrigerated display cabinet: deterministic and stochastic approaches

Abstract: The product temperature in the refrigerated display cabinets is very variable and it influences directly the safety and quality of the food products. This variability is due to the type of display cabinet (vertical, horizontal), the position of the product inside the equipment, the fluctuation of the ambient air temperature in the supermarkets (day/night, opening hours, seasons, air conditioning etc.) and the surrounding condition of the display cabinet (in front of another refrigerated display cabinet or in front of grocery products) etc. This study proposes a thermal model to predict the load temperature in an open vertical display cabinet in function of two random parameters: ambient temperature in the supermarket and radiation temperature of the surrounding walls. The knowledge of the probability density of these parameters, from field measurements or expert knowledge, allows predicting the variability of the product temperatures inside the display cabinet.
Non-adiabatic and time-resolved photoelectron spectroscopy for molecular systems

Abstract: We quantify the non-adiabatic contributions to the vibronic sidebands of equilibrium and explicitly time-resolved non-equilibrium photoelectron spectra for a vibronic model system of Trans-Polyacetylene. Using exact diagonalization, we directly evaluate the sum-over-states expressions for the linear-response photocurrent. We show that spurious peaks appear in the Born-Oppenheimer approximation for the vibronic spectral function, which are not present in the exact spectral function of the system. The effect can be traced back to the factorized nature of the Born-Oppenheimer initial and final photoemission states and also persists when either only initial, or final states are replaced by correlated vibronic states. Only when correlated initial and final vibronic states are taken into account, the spurious spectral weights of the Born-Oppenheimer approximation are suppressed. In the non-equilibrium case, we illustrate for an initial Franck-Condon excitation and an explicit pump-pulse excitation how the vibronic wavepacket motion of the system can be traced in the time-resolved photoelectron spectra as function of the pump-probe delay.
Optimized expression of Plasmodium falciparum erythrocyte membrane protein 1 domains in Escherichia coli

Abstract: Several domains of PfEMP1 genes obtained from different P. falciparum strains were expressed in E. coli as GST-fusion proteins. Expression was carried out under various culture conditions with a main focus on the time point of induction in relation to the bacterial growth stage.When expressed in E. coli recombinant proteins derived from P. falciparum sequences are often truncated and tend to aggregate what in turn leads to the formation of insoluble inclusion bodies. The analysis of various factors influencing the expression revealed that the time point of induction plays a key role in successful expression of A/T rich sequences into their native conformation. Contrary to recommended procedures, initiation of expression at post-log instead of mid-log growth phase generated significantly increased amounts of soluble protein of a high quality. Furthermore, these proteins were shown to be functionally active. Other factors such as temperature, pH, bacterial proteases or the codon optimization for E. coli had little or no effect on the quality of the recombinant protein, nevertheless, optimizing these factors might be beneficial for each individual construct. In conclusion, changing the timepoint of induction and conducting expression at the post-log stage where the bacteria have entered a decelerated growth phase, greatly facilitates and improves the expression of sequences containing rare codons.Qualitative and quantitative production of proteins in heterologous systems is essential for the characterization of any molecule, from determination of antigenicity, functional and structural analysis to vaccine development. Malaria antigens are among the most difficult proteins to express with in vitro methods because of their extreme genetic codon usage.
