Abstract: Given the importance of the thymus to the immune system and the limited knowledge about its development and involution processes, this study evaluated the expression of the vascular endo-thelial growth factor, a key angiogenic factor for the formation of vascular tissue bed and modulation of functions related to cell proliferation and migration, hormone synthesis, and immunomodulation. In this study, thymus samples of dog fetuses (30, 40, 50 and 60 days old), and young (180 days old) and adult (365 days old) dogs were submitted to real-time PCR and Western blot. Time-dependent changes were observed for proteins and genes associated with the development of the vascular system, such as VEGF-A, VEGF-C, Flt-1, KDR, FLT-4, and EG-VEGF. This result demonstrates that these system components play different roles during development and involution of the thymus. Possible functions, such as the modulatory effect on vasculogenesis and microenvironment, influence the proliferation and differentiation of thymocytes, T cells maturation, cell-cell interaction and hormone secretion.
Embryonic thymic development in fetuses of domestic cats (Felis domesticus)

Abstract: During fetal life, and during the neonatal period, the thymus is a very important immune organ, and is the largest lymphatic organ, which exhibits high lymphopoietic activity as a precursor of lymphopoiesis. Morphological studies on the development of the thymus are rare and only include general information. Given the above, this study aimed to characterize the morphological development of the thymus of embryos and fetuses of domestic cats (Felis domesticus), from natural pregnancy, using macroscopic dissection techniques and light microscopy. The thymus of the cats was pale pink and was resting in the region of the cranial mediastinum, medially to the lungs and dorsally to the base of the heart. Histologically, two distinct regions were observed (cortical and medullar). The medullary region had reticular epithelial cells with large nuclei and dendritic extensions. The fetuses had exponential growth and were more pronounced starting on the 35th day of gestation.

Abstract: There were utilised 10 fetuses of jumentos, males and females, to macroscopic study of the thymus vascularization. After the arteries injection, throught the aorta, with Neoprene Latx “450” solution, it was proceded the fixation, in a 10% formalin solution. It was observed that the thymus is reached by direct or indirect arterial branches coming from the right and left internal thoracic, left subclavian arterie, right superficial cervical, left extern thoracic and direct and indirect branches of left and right carotidas arteries. Para a dissec o dos fetos, o conteúdo arterial recebeu solu o aquosa a 50% de Neoprene Látex “450”, corada com pigmento específico, em seguida os fetos foram fixados em solu o aquosa a 10% de formol. Abstract: the increase of heart collagen fibers in diabetics is a well known fact, but the consequences are not defined. the aim was to quantify the cardiac collagen fibers in normal and diabetic rats treated with vitamin c. we selected 32 wistar rats, 16 diabetic animals induced endovenously with streptozootocin, and 16 healthy animals, half of them, diabetics and normals, were treated with vitamin c for 90 days. after the experimental proceeding, the hearts were removed and processed accordingly to conventional protocol for optical microscopy and specific staining for collagen. the results showed that the diabetic rats presented increase in the number of cardiac collagen fibers, but the ones treated with vitamin c showed little accumulation of fibers. it could be concluded that treatment with vitamin c is important for the prevention of heart failure in diabetic animals.

Abstract: the objective of this study was to analyze morphometric histological shatters of the spleen of normal and diabetic animals, comparing the joined results and relating them in the sex and the suplementation of vitamin c. had been used 32 wistar rats, which had been analyzed: number of vases; the number of germinative folicules (white pulp); and the diameter of the vases of each animal. histological and morphometric analyses were applied on 5μm thick samples and showed that: in the amount of germinative folicules, we observe resulted comparing, independent of the sex, animals controls supplemented with vitamin c and controls not supplemented (p£0.05; f=0.1452); in the amount of vases, we observe resulted comparing, of diabetic females supplemented by vitamin c when compared with not supplemented (pd"0.05; f=6.8893); and in the diameter of the vases, we observe resulted comparing females, as much in the group has controlled how much to the diabetic group, supplemented with vitamin c when compared with the females not supplemented with vitamin c (p<0.05; u=121.50; z(u)=2.1234) and (p<0,05; f=4.8134). in a general way, the induction of diabetes modifies the vascular diameter in the females and that the vitamin c administration only intervenes with relative metric data to the vascular diameter in the females.

Abstract: the sinoatrial node, for being topographically installed as the initial component of the conduction system, is responsible for the production of the nervous impulses, which determines the cardiac contraction. there have been made studies related to the node's morphology in order to know the origin, distribution and quantification of the vases in this tissue, however, in spite of the results and quantitative data of the nodal irrigation - arterial vascular conduct and arterial vascular density, at the nodal level - the literature is scarce. with this objective 27 srd swine hearts, injected with colored resin for macroscopic analysis of the origin and distribution of ansa (sinoatrial node artery), 3 others injected with watery solution of colloidal coal (dyed nanquim) were used, to mark the route of the vases on the node level for stereological analysis. the atrial arteries originated as well from the right coronary artery as from the left one, with predominance of the first (66.66% and 33.33%, respectively). when originated from the right coronary artery, there existed the following branches: aadam (right cranial medial atrial artery) in 14 cases, aadai (right cranial intermedial atrial artery) in 2 cases, and aadal (right cranial lateral atrial artery) in 2 cases. in 9 cases (33.33%) the following branches originated from the left coronary artery: 4 through branch aaspl (left caudal lateral atrial artery), 2 through branch aasai (left cranial intermedial atrial artery) and 3 through branch aasam (left cranial medial atrial artery). interarterial anastomoses with vessels responsible for irrigation of the sinoatrial node area were observed in most of the cases (25 hearts). the volume or reference volume (v(ref)) of the hearts were 35.32x104μm3. for the stereological variables analyzed, the estimate density of the vessel length (lv) was 766; the vessel length (l) - mm - was 27.06x105μm; the area's surface density (sv) was 182 and the area's surface (s) - mm2 - was 64.3

Abstract: the thymus is an organ with great immunological relevance during foetus life and neonatal period, it is the predecessor of lymphopoiesis and shows high lymphopoietic activity, being also the largest lymphoid organ during intrauterine life and at birth. in this study, the morphological aspects of the prenatal development of dog thymus were described in foetus of different ages and sexes. twenty-four foetus of mongrel domestic dogs, males and females, were divided into four age groups. the thymus presented two lobes linked by a rose-colored tissue connection that was localized in the cranial mediastinal space. the cranial portion protrudes slightly beyond the first pair of ribs. the cells that formed the thymus parenchyma were well organized. concentric aggregates named hassal corpuscles were observed, involved by a thin capsule of connective tissue thickened by adipose tissue. the vessels size gradually increased with the foetus age, being larger in animals that were closer to birth. the volume (vref) and size parameters of female thymus were larger than in males.
Assessment of NADPH-diaphorase stained myenteric neurons of the jejunum of diabetic rats supplemented with ascorbic acid

Abstract: the relation between hyperglycemia and diabetic neuropathy has already been demonstrated in some studies. among the theories proposed for its etiology the oxidative stress stands out. the performance of nitric oxide as a link between the metabolic and vascular neuropathogenic factors that triggers the diabetic neuropathy has already been put forward. this study aimed to assess the quantification and measurements of the cell body profile area (cbpa) of nadph-diaphorase reactive (nadph-dp) myenteric neurons of the jejunum of diabetic rats (induced by streptozotocin) supplemented with ascorbic acid (aa). these changes in the myenteric neurons seem to be related to the gastrointestinal disturbances observed in diabetes mellitus (dm). twenty male wistar rats (rattus norvegicus) were distributed in 4 groups (n=5): controls (c), control supplemented (cs), diabetic (d), and diabetic suplemented (ds). dm was induced by estreptozotocin (50mg/kg body wt). one week after the induction and confirmation of the dm (glycemia exam), animals of the groups cs and ds received 50mg of aa three times a week by gavage. after 90 days of experiment, the animals were anesthetized with lethal thiopental dose (40mg/kg) and the collected jejunum processed for the histochemistry nadph-diaphorase technique. whole-mount preparations were obtained for quantitative and morphometric analysis of the myenteric neurons. a quantity of jejunum neurons in the group d (96±7.5) was not different (p>0.05) from group ds (116±8.08), c (92±9.7), and cs (81±5.4), but in group ds the quantity was higher (p<0.05) than in group c and cs. the cbpa of neurons from group d (189.50±2.68μm2) and ds (195.92±3.75μm2) were lower (p<0.05) than from group c (225.13±4.37μm2) and cs (210.23±3.15μm2). the streptozotocin-induced dm did not change the jejunum-ileum area, the jejunum myenteric plexus space organization and the density of nadph-dp neurons. the 50g aa-supplementation, three times a week, during 90 days, did not decre

New PHA products using unrelated carbon sources

Abstract: polyhydroxyalkanoates (pha) are natural polyesters stored by a wide range of bacteria as carbon source reserve. due to its chemical characteristics and biodegradability pha can be used in chemical, medical and pharmaceutical industry for many human purposes. over the past years, few burkholderia species have become known for production of pha. aside from that, these bacteria seem to be interesting for discovering new pha compositions which is important to different industrial applications. in this paper, we introduce two new strains which belong either to burkholderia cepacia complex (bcc) or genomovar-type, burkholderia cepacia sa3j and burkholderia contaminans i29b, both pha producers from unrelated carbon sources. the classification was based on 16s rdna and reca partial sequence genes and cell wall fatty acids composition. these two strains were capable to produce different types of pha monomers or precursors. unrelated carbon sources were used for growth and pha accumulation. the amount of carbon source evaluated, or mixtures of them, was increased with every new experiment until it reaches eighteen carbon sources. as first bioprospection experiments staining methods were used with colony fluorescent dye nile red and the cell fluorescent dye nile blue a. gas chromatography analysis coupled to mass spectrometry was used to evaluate the pha composition on each strain cultivated on different carbon sources. the synthesized polymers were composed by short chain length-pha (scl-pha), especially polyhydroxybutyrate, and medium chain length-pha (mcl-pha) depending on the carbon source used.
